Background: Theileria equi is a tick borne protozoan parasite which causes piroplasmosis among equines worldwide. The present study was aimed to determine seroprevalence of T. equi in donkeys, horses, and mules from two equine populated districts (Peshawar and Charsadda) of Khyber Pakhtunkhwa (KPK), Pakistan. Methods: A total of 393 equine (195 horses, 194 donkeys and 4 mules) serum samples were collected from five and four randomly selected localities in Charsadda (n = 193) and Peshawar (n = 200), respectively. The presence of antibodies to T. equi was determined using a commercially available competitive enzyme-linked immunosorbent assay. A comparative study of serological tests and PCR for the diagnosis of equine piroplasmosis

Working donkeys (n ¼ 395) originating from three different districts of the Oromia region of Ethiopia were examined October 2009 and April 2010 for the prevalence of equine piroplasmosis between. Apart from hematological and serological examination, tick vectors were collected and determined. Intraerythrocytic stages of Theileria equi and Babesia caballi were detected in 12.2% and 1.8% of examined stained blood smears, respectively. The seroprevalence of T. equi and B. caballi was 55.7% and 13.2%, respectively. The majority of active infection coincided with anemia. Only 16 donkeys were infested with four species of hard ticks: Rhipicephalus turanicus, Rhipicephalus evertsi evertsi, Rhipicephalus pulchellus, and Amblyomma variegatum.

Equine piroplasmosis is significant tick-borne disease with wide distribution. The prevalence of equine piroplasmosis in Serbia, Montenegro and Bosnia and Herzegovina is unknown. In aim to obtain a first insight into the prevalence we performed molecular epidemiological study which included 142 horses, on seven locations in these three countries. We first performed PCR for the detection of a 450bp long section of the 18S rRNA of piroplasma-specific region. For all positive samples we have done multiplex PCR for the species detection. Species determination was further confirmed by sequencing PCR products of 10 randomly selected Theileria equi and all Babesia caballi samples. The overall prevalence rates in analysed region for T. equi and B. caballi were 22.5% and 2.1%, respectively. Possible risk factors (such as location, age, sex and activity) associated with PCR positivity were evaluated. Marked differences were found in prevalence between geographic areas. There was no significant association between positivity and age group. T. equi was more prevalent in females and farming horses. This is the first report on the molecular survey of T. equi and B. caballi in central Balkan. Further prevalence studies on definitive host and vectors in this region are necessary.

Equine Piroplasmosis (EP) is an infectious disease caused by the hemoprotozoan parasites Theileria equi, Babesia caballi, and the recently identified species T. haneyi. Hereby, we used a multiplex PCR (mPCR) targeting the 18S rRNA gene of T. equi and B. caballi for the simultaneous detection of EP in Egyptian equids and examined the presence of T. haneyi infections in Egypt. Blood samples from 155 equids (79 horses and 76 donkeys) collected from different governorates of Egypt were examined by mPCR and PCR targeting T. hayeni. The mPCR method revealed a prevalence of T. equi of 20.3% in horses and of 13.1% in donkeys and a prevalence of B. caballi of 1.2% in horses. Equine piroplasmosis, an economically important disease of equids caused by the hemoprotozoan parasites Theileria equi, T. haneyi, and Babesia caballi, has a worldwide distribution. These parasites are transmitted by ixodid ticks. To improve the detection of horses in Nigeria exposed to piroplasm parasites, 72 horses with variable clinical signs of piroplasmosis were sampled from Northwest and Northcentral Nigeria and tested by nPCR and cELISA. Blood and serum samples were collected from each horse via jugular venesection. Individually, nPCR or cELISA failed to identify all horses exposed to piroplasms. A combination of species-specific nPCR and the OIE-approved T. equi and B. caballi cELISAs enhanced the detection of horses exposed to parasites. heileria equi is a tick-borne hemoprotozoan parasite and one of the causative agents of equine piroplasmosis. Data on T. equi infection in donkeys are scarce in Assiut governorate, Egypt. This study was conducted on the prevalence of T. equi infection in donkeys by using micropscopic examination and polymerase chain reaction (PCR) assay. Out of 50 donkeys, blood samples examined for T. equi, 14% (7/50) were positive by microscopic examination and 38% (19/50) by PCR. Prevalence of T. equi had high significant differences between microscopic examination and PCR assay. Prevalence of T. equi in male donkeys was 13.6% (3/22) by microscopic examination and 36.4% (8/22) by PCR. In female donkeys it was 14.3% (4/28) by microscopic examination and 39.3% (11/28) by PCR. No significant difference between the prevalence rate of infection in males and females were recorded. Microscopic examination of donkeys' blood smears stained with Giemsa stain revealed forms of theilerial schizogony in lymphocytes (Kochs blue bodies) and intra-erythrocytic relatively small, less than 2 μm long pear-shaped and ring shaped merozoites of T. equi. The B1 gene specified for T. equi was detected by PCR in 19 blood samples. All positive samples with microscopic examination were also positive with PCR. The sensitivity, specificity and accuracy of PCR with respect to blood film examination were 61.3%, 100% and 82%, respectively. So, PCR was found to be more sensitive, specific and accurate than blood film examination.The present results suggest that T. equi parasite is widely spread in Assiut governorate due to high exposure to ticks.

Background: Equine piroplasmosis (EP) is the cause of persistent tick-borne infection with no symptoms, but the most important problem of EP is due to the persistent carrier state. Carrier animals to Babesia (Theileria) equi (Laveran 1901) and B. caballi (Nuttall, 1910) infestation could be identified by extremely sensitive PCR-based method. The purpose of this study was to identify the causative agents of equine piroplasmosis based on molecular and microscopic assays in equids from Kurdistan Province, Iran.
