Andrographis paniculata (Burm.f.) wall.ex Nees (Acanthaceae) or Kalmegh is an important medicinal plant finding uses in many Ayurvedic formulations. Diterpenoid compounds andrographolides (APs) are the main bioactive phytochemicals present in leaves and herbage of A. paniculata. The efficiency of supercritical fluid extraction (SFE) using carbon dioxide was compared with the solid-liquid extraction techniques such as solvent extraction, ultrasound-assisted solvent extraction and microwave-assisted solvent extraction with methanol, water and methanol-water as solvents. Extraction, isolation and characterization of Andrographis paniculata (A.p.) products were developed. Three natural diterpenes compounds were obtained and one was used for chemical modifications. Evaluation of their inhibition of TNFα induced NFκB transcriptional activity. A rapid analytical method for the determination and quantitation of three diterpenoid lactones (andrographolide 1, didehydroandrographolide 2, neoandrographiside 3) found in A. paniculata extracts was investigated. Objective: The aim of this study was to observe the antidyslipidemic and antioxidant activity of andrographolide (AND) compound from Sambiloto (Andrographis paniculata) herb. The antidyslipidemic activity could be described by lowering TC (total cholesterol), TG (triglyceride), and LDL (low-density lipoprotein) level in the blood and by increasing HDL (high-density lipoprotein) level in the blood. Andrographis paniculata (Burm. f.) Wall. ex Nees, commonly known as Kalmegh, is widely used as antimalarial drug in herbal and traditional systems. Andrographolide is the major triterpenoid present in the plant and responsible for its therapeutic activities. The identification and quantification of andrographolide were ascertained by various spectrophotometric and chromatographic analyses. The plant sample was extracted with methanol. The amorphous residue obtained from extraction was analyzed for identification of andrographolide by chemical method, TLC, UV-Vis, FT-IR and LCMS/MS analyses. Isolation and Characterisation of Andrographolide from Andrographis paniculata (Burm. F) Wall. Ex Nees and Its Total Flavonoid Effects from Kemaman, Malaysia

Background: Many medicinal plants have wide range of applications from food and aesthetic materials to pharmacological effect on various disease conditions. Traditionally, these plants have been used to treat many diseases including diabetes, inflammation, upper respiratory tract infections and diarrhoea either in single or combined form with other plant parts or materials. Objective: This study investigated the isolation and characterisation of andrographolide from Andrographis paniculata (Burm. f) Wall. ex Nees. Methodology: The isolation of the constituents was done using column total flavonoid conducted while the analysis was done using ultraviolet, infrared spectrum analysis (UV, IR), melting point and elemental analysis. Also, proton and carbon nuclear magnetic resonance (1 H and 13 C NMR) of the isolated compound was performed using different equipment. Results: Although, the total flavonoid content was low, ethyl acetate extract indicated the highest total flavonoids content and it's statistically different from methanol, ethanol: water (1:1 v/v) and aqueous extracts. The compound obtained is colorless solid with melting point range of 228 o C-238 o C. This was further supported by 13 C NMR spectral analysis, which displayed 20 signals for all carbon atoms and is like standard compound Andrographolide. Also, the ultraviolet analysis suggesting α, β-unsaturated lactone ring similar andrographolide while the elemental analysis shows the different elements present. Conclusion: Different extracts of A. paniculata exhibit multiple pharmacological effects. However, due to the much similarities with the isolated compound and Andrographolide the compound is suggested to be Andrographolide.

Identification of Medicinal Organic Components of Andrographis paniculata Leaf Extracts

Active metabolites of methanol, ethanol, ethyl acetate and aqueous extracts of the leaves of Andrographis paniculata were identified using thin layer chromatography and gas chromatography-mass spectrometry (GC-MS). Results from this study identified some of active metabolites as andrographolide, 14-deoxyandrographolide, neoandrographolide, 14-deoxy-11,12-di-dehydroandrographolide, andrographanin, isoandrographolide, 14-acetylandrographolide, 19-O-andrographolide, beta-sitosterol and 14-deoxy-14,15dihydroandrographolide. Ethanol was a more effective solvent for the extraction as most of the phytochemicals were identified in this extract. Research had shown that extracts from the leaves of the plant possess a lot of therapeutic properties such as anti-microbial, anti-inflammatory, anti-diabetic, immune modulatory and antimalarial activities which are of immense benefit to man.

Sambiloto (Andrographis paniculata) is widely used as a medicinal plant for Indonesians. The content of extracted bioactive compounds is affected by the polarity of the extraction solvent. This study aims to determine the effect of extraction solvent on the chromatographic profile of Sambiloto herb, especially terpenoid group using the TLC-Spectrophotodensitometry method. Sambiloto herb was extracted by sonication method using methanol, ethanol, ethyl acetate, and chloroform as the solvents. Chromatographic profiles were identified by stationary phase of TLC silica gel GF254 plate and a mobile phase of chloroform:methanol (10:1 v/v), observed visually, and scanned at 210 nm. Extraction of Diterpenoid Lactones of Andrographis paniculata using Liquid Solvents: Effect of Solvent's Hildebrand Solubility Parameter on Extraction Efficiency

Two diterpenoid lactones responsible to the multi healing properties of Andrographis paniculata have been extracted using 16 different liquid solvents. The effect of Hildebrand solubility parameter of the solvents on solid-liquid extraction yield using standard soxhlet extraction method was investigated. The Hildebrand solubility parameter concept performed well when predicting lower polar solvent effect on extraction efficiency. However, ethanol extracted less andrographolide than methanol even though ethanol has closer Hildebrand solubility parameter value to andrographolide compared to that of methanol. Methanol's shorter aliphatic chain and higher polarity, which lead to easier hydroxyl group liberation, seemed to contribute this discrepancy.
