Boronic acid compounds as potential pharmaceutical agents

Boronic acid compounds have been used, because of their unique structural features, for the development of potent enzyme inhibitors, boron neutron capture agents for cancer therapy, and as antibody mimics that recognize biologically important saccharides. Boronic Acids and Their Derivatives in Medicinal Chemistry: Synthesis and Biological Applications

Boron containing compounds have not been widely studied in Medicinal Chemistry, mainly due to the idea that this group could confer some toxicity. Nowadays, this concept has been demystified and, especially after the discovery of the drug bortezomib, the interest for these compounds, mainly boronic acids, has been growing. In this review, several activities of boronic acids, such as anticancer, antibacterial, antiviral activity, and even their application as sensors and delivery systems are addressed. The synthetic processes used to obtain these active compounds are also referred. Noteworthy, the molecular modification by the introduction of boronic acid group to bioactive molecules has shown to modify selectivity, physicochemical, and pharmacokinetic characteristics, with the improvement of the already existing activities. Advances in the field of boron chemistry have expanded the application of boron from material use to medicine. Boron-based drugs represent a new class of molecules that possess several biomedical applications including use as imaging agents for both optical and nuclear imaging as well as therapeutic agents with anticancer, antiviral, antibacterial, antifungal and other disease-specific activities. For example, bortezomib (Velcade®), the only drug in clinical use with boron as an active element, was approved in 2003 as a proteasome inhibitor for the treatment of multiple myeloma and non-Hodgkin’s lymphoma. Several other boron-based compounds are in various phases of clinical trials, which illustrates the promise of this approach for medicinal chemists working in the area of boron chemistry. A standard goal of medicinal chemists has been to discover efficient and potent drug candidates with specific enzyme-inhibitor abilities. In this regard, boron-based bioactive compounds have provided amphiphilic properties to facilitate interaction with protein targets. Boron Containing Compounds as Protease Inhibitors

In this context, the organoboron compounds are fast becoming compounds used for many things from starting materials in organic synthesis to approved drugs. Researchers are increasingly looking to boron containing molecules as protease inhibitors. Moreover, there are many interesting applications of some boronic acid compounds which target the HIV proteases. The elucidation of their mechanism of action as anti-HIV agents may lead to more biological targets for boronic acid compounds. An increasing research activity in this area is expected because of the enormous potential it presents.

Boric acid: a simple molecule of physiologic, therapeutic and prebiotic significance

Boric acid, H3BO3, is a weak acid and at physiological pH is in the form of an uncharged small molecule. Behaving as a Lewis acid, it forms complexes with amino- and hydroxy acids, carbohydrates, nucleotides and vitamins through electron donor-acceptor interactions. These interactions are believed to be beneficial for human health. Synthetic bis-chelate complexes of boric acid with organic biomolecules are therefore considered for nutritional and/or pharmaceutical applications. The use of boric acid for BNCT has gained attention due to the short biological half-life, solubility, plasma circulation and the non-selective soft tissue accumulation properties of this simple molecule.
