Evaluation of the immunomodulatory effects of five herbal plants

A group of medicinal plants including, Silybum marianum, Matricaria chamomilla, Calendula officinalis, Cichorium intybus and Dracocephalum kotschyi which grow in Iran, were extracted with ethanol 70% and the mitogenic activity was examined both on human peripheral blood lymphocytes and thymocytes. Effect of these extracts on proliferative responsiveness of human lymphocytes to phytohemagglutinin (PHA) and on the mixed lymphocyte reaction (MLR) was also investigated. The results obtained indicated that none of the extracts had a direct mitogenic effect on human lymphocytes or thymocytes (stimulation index, SI B0.07). Among the plants studied, C. intybus and C. officinalis showed a complete inhibitory effect on the proliferation of lymphocytes in the presence of PHA (SI range 0.01-0.49). A dose dependent inhibitory effect was obtained in the case of D. kotschyi. Extract of M. chamomilla showed almost no stimulatory effect. A significant decrease in proliferation assay due to 0.1-10 mg/ml of S. marianum was observed (SI B0.46, PB 0.05). In MLR, a markedly stimulatory effect with some lower concentrations of all the extracts except Dracocephalum was detected. The highest stimulatory effect was due to 100 mg/ml of S. marianum (SI 2.82). Treatment of mixed lymphocytes with 0.1-10 mg/ml of C. officinalis (SI range 1.34-1.80) and 10 mg/ml of M. chamomilla and C. intybus (SI 2.18 and 1.70, respectively) strongly increased the cell proliferation. In conclusion, this in vitro study revealed the capacity of all the extracts except Dracocephalum to enhance the proliferation of lymphocytes after stimulation with the allogenic cells.

We assessed the effects of Cleistocalyx nervosum var paniala, Gynostemma pentaphyllum, Gynura procumbens, Houttuynia cordata, Hyptis suaveolens, Portulaca grandiflora, Phytolacca americana and Tradescantia spathacea on lymphocyte proliferation and the effects of C. nervosum, G. pentaphyllum, H. suaveolens and P. grandiflora on natural killer (NK) cells activity. All of the extracts significantly stimulated human lymphocyte proliferative responses at various concentrations depending on each extract. The extracts of C. nervosum and H. suaveolens were significantly enhanced NK cells activity while those of G. pentaphyllum and P. grandiflora did not alter NK cells function. Our results suggested that the extracts of those plants have stimulating activity on human lymphocytes and could be clinically useful for modulating immune functions of the body.
In Vitro Modulatory Effects of Certain Plant Extracts on T-lymphocytes Proliferation
Herbal drugs are believed to enhance the natural immunity of the human body against viral and microbial infection and improve response; their immunomodulatory activities have been reported in numerous plants. In this study, fifty plant alcoholic extracts were screened for their potential immunomodulatory activity using in-vitro human lymphocyte transformation assay (LTA). Among the screened extracts, Aralia victoria, Boswellia carteri, Cyperus rotundus, Ginkgo biloba, Glycyrrhiza glabra, Olea europaea and Rosmarinus officinalis were capable to induce 96.67%, 32.22%, 11.11%, 5.56%, 25.56%, 13.33% and 2.22% respectively of lymphocyte transformation in comparison to Concanavalin-A (20.5%). Subsequently, each extract was further subjected to bio-guided fractionation using LTA. The bio-guided fractionation revealed that Dichloromethane and Petroleum-ether fractions from Glycyrrhiza glabra and Boswellia carteri stimulate the transformation of human lymphocytes by 86% and 28% respectively. This finding stimulated the interest to pursue a comprehensive study on the possible contribution of the commonly known and structurally related triterpens of Glycyrrhiza glabra and Boswellia carteri and their derivatives as potential immunomodulators and their possible use as immune enhancer in many infectious and cancer diseases.
Review on traditional plants with phytochemical having immunomodulatory activities

Immunomodulatory agents initiate the activation of non-specific immune responses and work without antigenic specificity. They directly activate different cell populations of the immune system and modify the production of cytokines and initiate the production of various effect or molecules that take part in the modulation and enhancement of the immune response. Since ancient times, plants have been an exemplary source of medicine. Some of the medicinal plants valued in Ayurvedic Rasayana for their therapeutic potential have been scientifically investigated with promising results. A number of plant-based principles have been isolated with potential immunomodulatory activity that can explain and justify their use in traditional medicine in the past and can form the basis for further research in the future as well. The present work constitutes a review of the medicinal plants whose immunomodulator activity has been proven. We performed PUBMED, EMBASE, Google scholar searches for research papers of medicinal plants having immunomodulator activity. This review shall hopefully encourage researchers to undertake further work on medicinal plants with potential immunomodulatory activity.
Screening of fourteen Moroccan medicinal plants for immunomodulating activities
The aim of this study is to determine the immunomodulating ability of total extract of fourteen plants used in traditional Moroccan medicine, Citrullus colocynthis L., Elletaria cardamomum L., Piper cubeba L., Ammi Visnaga L., Urtica dioïca L., Juncus acutus L; Capparis spinosa L., Aristolochia longa L., Marrubium vulgare L., Delphinium staphysagria L., Sinapis nigra L., Tetraclinis articulata L., Lepidium sativum L. and Datura stramonium L. The immunomodulatory activity was tested using primary culture of rabbit splenocytes. Cell proliferation in presence or absence of plant extracts and with or without a mitogen (Concanavalin-A, Con-A) was assayed by MTT assay. We observed firstly that (i) the addition of plant extract to splenocytes didn't show significant cytotoxicity; (ii) except for Citrullus colocynthis L., Elletaria cardamom L., and Piper cubeba L., other plant extracts stimulate splenocyte proliferation with high effect for Ammi Visnaga L., (iii) Citrullus colocynthis impaired partially the mitogenic action of Con-A. In the second time, we investigate the hemaglutination properties of plant extracts. Results obtained showed that the plant extracts which stimulate splenocyte proliferation bind to blood cells inducing hemagglutination except for Ammi visnaga L., and Urtica dioïca L. This study revealed interesting immunomodulating actions of plants which could explain their traditional use.

To investigate the effects of 20 methanolic extracts from Malaysian selected plants on CD18/11a expression and phagocytosis activity of leukocytes using flow cytometry analysis. Methods: The effects of methanolic extracts on CD18/11a expression and phagocytosis of leukocytes were measured by labelling the cells with CD18-fluorescein isothiocyanate and ingestion labelled with Escherichia coli-fluorescein isothiocyanate and then analyzed using flow cytometer. Results: About 12 out of 20 methanolic extracts of selected Malaysian medicinal plants significantly (P≤0.05) inhibited the CD18/11a expression of leukocytes at both concentrations of 6.25 μg/mL and 100 μg/mL in dose dependent manner. The most active inhibitory was shown in Citrus aurantifolia (Christm.) Swingle and Alpinia galangal (L.) Willd. at dosage 100 μg/mL. Moreover, the Orthosiphon aristatus (Blume) Miq (O. aristatus). showed the highest stimulatory activity at the concentration of 100 μg/mL. Other than that, four plant extracts significantly (P≤0.05) rose the phagocytosis activities of leukocytes in dose dependent manner. However, Annona muricata L. and O. aristatus showed the highest stimulated activities at the 100 μg/mL concentration.
Herbal Plants Used for Immunomodulatory Action: A Review
The immune system is one of our most complex biological system in the body. Immunization may be active or passive. The active immunization involves stimulation with an antigen to develop immunological defences against a future exposure and Passive immunization involves administration of preformed antibodies to an individual who is already exposed to be exposed to an antigen. The use of a variety of agents to enhance immunological and nonspecific host defences and thus to modify the favorably is an exciting development in immunopharmacology that is such agents may act by increase the humoral antibody responses, enhancing the phagocytic activity of macrophages or modifying the cell-mediated immune response. Immunomodulatory agents originate from both plant and animal which increases the immune responsiveness of the body against pathogens by activating the non specific immune system. The immune system dysfunction is responsible for various diseases like allergy, asthma, arthritis, cancer and other infectious diseases. So modulation of immune responses to much required to controlling the various infectious diseases. The texts of traditional Indian medicine literature consist of a number of plants reputed to promote physical and mental health, improve defence mechanisms of the body and enhance longevity, lesser number of side effects against synthetic immunomodulatory agent.
Effect of five medicinal plants used in Indian system of medicines on immune function in Wistar rats

Today, the use (which we can say 'return to nature') of traditional herbal medicines, herbal health products, pharmaceuticals food supplement, cosmetics, etc. is increasing due to the growing recognition that natural products are safe; have either no or negotiable side effects. The objective of this study was to investigate the immunomodulatory properties of five different medicinal plants used in Indian system of medicines on the Wistar rats. The activity was investigated by phagocytic carbon clearance, antibody titre and delayed type hypersensitivity test. The control group received 0.1% carboxyl methyl cellulose and other groups received the different doses of all the five plants extracts such as Tribulus terrestris, Cassia tora, Achyranthes aspera, Mucuna prurians and Abrus precatorius intraperitoneally for 7 consecutive days. All the five plants exhibited immunostimulatory activity and out of the five plants, T. terrestris showed significant dose dependent increase in the 'humoral antibody titre and DTH response' as indicated by increase in footpad thickness. It also showed significant increase in the 'phagocytic index' in rats. The investigation revealed that all the mentioned plants have immunomodulatory activities.
Immunomodulatory effect of medicinal plants, Kolli hills, Tamil Nadu
Four medicinal plant species were collected from the Kolli hills of Tamil Nadu and were screened for their immunosuppressive effect. The plants were shade dried and extracted with methanol. The crude methanol extracts were tested for inhibition of lymphocyte proliferation via lymphocyte proliferation assay by 3 thymidine uptake. The test plants were Justicia gendarussa, Plumbago indica, Aloe vera, and Aegle marmelos. Among the plants tested J. gendarussa (100 µg/ml) showed the highest lymphocyte inhibition (84%). Sequential extraction of J. gendarussa in various solvents (n-hexane, benzene, ethyl acetate, chloroform, acetone, ethanol, and water) confirmed that all of the above extracts at 50 µg/ml, aqueous extract inhibited lymphocyte proliferation. Further, 17 high performance liquid chromatography fractions were collected for the aqueous extract and fraction no. 15 showed maximum inhibition of lymphocyte proliferation. The present study indicates that these extracts should be investigated further for the possible presence of immunosuppressive components.
