Blood DNA analysis for Ehrlichia (Anaplasma) phagocytophila and Babesia spp. in Dogs from Northern Poland

The most common pathogen transmitted by the tick Ixodes ricinus is the spirochete Borrelia burgdorferi, rarely Ehrlichia bacteria and protozoans from the Babesia genus. The aim of this paper was determine if infested to ticks dogs are a reservoir for E. phagocytophila and Babesia spp. and examine the possibility of coinfection. Canine blood was sampled, part of the material originated from dogs exhibiting symptoms of borreliosis. In an earlier study, the samples were screened for DNA from B. burgdorferi sensu lato. In order to screen for E. phagocytophila and Babesia spp. DNA, a PCR-based method was used with the following primers: EHR521/EHR747 for Ehrlichia and FOR1/REV1 for Babesia. In 192 samples only two contained E. phagocytophila DNA. One of these samples originated from a healthy canine, the other from an individual with symptoms of borreliosis. The examined samples were not positive for Babesia spp DNA. Coinfection was not discovered. The low level of E. phagocytophila infection may indicate that the domestic dog is not a reservoir for Ehrlichia and Babesia in Szczecin and northwestern Poland. Moreover, this area does not have populations of the brown dog tick (Rhipicephalus sanguineus) or Dermacentor reticulatus-both of which are vectors of E. canis and B. canis and commonly induce ehrlichiosis and babesiosis in canines.

Occurrence of Ehrlichia canis and Anaplasma platys in household dogs from northern Parana

Canine monocytic ehrlichiosis caused primarily by Ehrlichia canis and canine thrombocytic anaplasmosis induced by Anaplasma platys are important emerging zoonotic tick-borne diseases of dogs. There is evidence that these pathogens can also affect humans. This study evaluated the presence of E. canis and A. platys in blood samples collected from 256 domiciled dogs in the municipality of Jataizinho, located in north region of the State of Parana, Brazil, by PCR assay. The occurrence of E. canis and A. platys was 16.4% (42/256) and 19.4% (49/256), respectively; while 5.47% (14/256) of the dogs evaluated were co-infected by these two organisms. The presence of E. canis and A. platys was not significantly associated with the variables evaluated (sex, age, outdoor access, and presence of ticks during blood collection). Occurrence of Babesia vogeli, Mycoplasma spp., Ehrlichia canis and Anaplasma spp. in a hospital dog population of western Paraná

Ehrlichia canis, Anaplasma platys, and Babesia vogeli are frequently detected in the veterinary routine with varied pathogenesis that can lead to death, mainly in co-infections. Although canine hemotropic mycoplasmas are considered of low clinical importance, they have recently gained prominence in molecular research. The present study aimed to analyze two hospital populations of dogs, randomly selected from the year 2013 and 2015, from the municipalities of Toledo (n=68) and Cascavel (n=107). Direct examination of blood smears, serology, and PCR were used to detect the presence of E. canis, A. platys, B. vogeli and Mycoplasma sp. Direct blood smear examination was negative for the investigated agents in all samples. First description of natural Ehrlichia canis and Anaplasma platys infections in dogs from Argentina

Bacteria belonging to the Anaplasmataceae family are vector transmitted agents that affect a variety of vertebrate hosts including the tick-borne pathogens Ehrlichia canis and Anaplasma platys, which cause canine monocytic ehrlichiosis and cyclic thrombocytopenia, respectively. These two infections, typically reported from tropical and sub-tropical regions, have not been previously reported in dogs from Argentina. A total of 86 blood samples from dogs with suspected rickettsial disease and 28 non-suspected dogs were studied. Analysis included evaluation of hematological findings, PCR for Ehrlichia and Anaplasma species and sequencing of the positive PCR products. E. canis was detected in the blood of six dogs and A. platys in eighteen. All the dogs categorized as non-suspected were negative by PCR. Co-infection with Hepatozoon canis and Babesia vogeli was documented. This first report of E. canis and A. platys infections in dogs from Argentina indicates that these tick-borne infections have a considerably broader range than previously recognized in South America.

Molecular evidence of Ehrlichia canis and Anaplasma platys and the association of infections with hematological responses in naturally infected dogs in Kalasin, Thailand

Background: Tick-borne bacteria, Anaplasma platys and Ehrlichia canis are well recognized as the etiology of anemia and thrombocytopenia in dogs. The clinical signs of anaplasmosis and ehrlichiosis range from asymptomatic to severe symptoms. There are insufficient studies about epidemiological surveys of these blood parasites, also the association of infections with the hematological study. Aim: This study aimed to screen A. platys and E. canis in naturally infected dogs and the effects of the infection on the levels of packed cell volume (PCV) and platelet count. Materials and Methods: A total of 68 blood samples were collected from free-roaming dogs at Nong Kung Sri district, Kalasin Province, Thailand, and examined for A. platys and E. canis infection by polymerase chain reaction (PCR) and measured PCV levels and platelet count. Results: Using nested PCR, 42.65% of dogs were infected with one or two pathogens. The molecular detection of anaplasmosis and ehrlichiosis in this population was 29.4% (95% confidence interval [CI]: 18.98-41.71) and 25% (95% CI: 14.4-35.3), respectively. Coinfection occurred at 11.8% (95% CI: 5.22-21.87). Infection with E. canis and coinfection showed significant association with PCV levels (p<0.05) while A. platys infection showed no statistical relationship. Infection with A. platys, E. canis, and coinfection had a non-significant correlation with platelet count (p>0.05). Conclusion: This study provides data of anaplasmosis and ehrlichiosis in free-roaming dogs which indicated that these zoonotic diseases are widespread and require for disease frequency determination, especially in Kalasin Province of Thailand where data of tick-borne infections in dogs have not been reported.

Molecular detection of Babesia vogeli, Ehrlichia canis and Anaplasma platys in a hospital population of dogs clinically diagnosed with hemoparasitosis

There is an increase in tick-borne diseases in dogs in urban and rural areas in Brazil and some of these are of public health importance. Rhipicephalus sanguineus-transmitted hemoparasitoses are the main causes of mortality in dogs. The present study investigated the molecular occurrence of Ehrlichia canis, Babesia vogeli and Anaplasma platys in dogs with clinical sings and hematological abnormalities suggestive of tick-borne diseases. These dogs were seen at a Veterinary Hospital of a Public University between January 2014 and December 2016, and were evaluated through anamnesis, clinical examination and complementary exams. Survey of Ehrlichia canis, Babesia spp. and Hepatozoon spp. in dogs from a semiarid region of Brazil

This study assessed the occurrence of Ehrlichia spp., Babesia spp. and Hepatozoon spp. infections in 100 tick-harboring dogs from a semiarid region of the State of Paraíba, Northeastern Brazil. Blood samples and ticks were collected from the animals, and a questionnaire was submitted to dog owners to obtain general data. Blood samples were used to perform hemogram, direct blood smear and immunological and molecular hemoparasite detection. The 1,151 ticks collected were identified as Rhipicephalus sanguineus; direct smears revealed E. canis-like morulae in the monocytes of 4% (4/100) of the non-vaccinated female dogs, and 34% and 25% of the dogs tested positive for Ehrlichia canis by indirect immunofluorescence assay (IFA) and polymerase chain reaction (PCR), respectively. HEMATOLOGICAL PARAMETERS AND SEROPREVALENCE OF Ehrlichia canis AND Babesia vogeli IN DOGS

The aim of this study was to determine the prevalence of anti-Ehrlichia canis and anti-Babesia vogeli IgG antibodies in dogs and correlate this prevalence with risk factors to evaluate the relation of serological status to hematological findings. Blood samples of dogs attended from September 2011 to March 2012 at the veterinary hospital of the Federal University of Lavras, Brazil, were analyzed using an indirect fluorescence antibody test (IFAT). Of the total 160 dog serum samples, 23.7% (38 dogs; CI95 17.7% - 30.7%) were seropositive for E. canis, 40.0% (64 dogs; CI95 40.0% - 59.2%) for B. vogeli, and 5.6% (9 dogs) for both hemoparasites. None of the epidemiological variables showed a significant association (P>0.05) with seropositivity to E. canis and B. vogeli. Detection and molecular characterization of Hepatozoon canis, Babesia vogeli, Ehrlichia canis, and Anaplasma platys in dogs from Metro Manila, Philippines

The study of canine vector-borne diseases in the Philippines started in the 1970s but only gained interest in the past decade. Characterization of such diseases in the Philippines remains incomplete, thus, it is necessary to obtain additional information on the prevalence and diversity of canine tick-borne diseases in the country. In this study, blood samples were obtained at two veterinary clinics in Metro Manila, Philippines from 114 dogs suspected of having canine tick-borne pathogens. Polymerase chain reaction (PCR) was performed on whole blood DNA extracts followed by sequencing, and the following pathogens were detected: Hepatozoon (H.) canis (5.26%), Babesia (B.) vogeli (5.26%), Ehrlichia (E.) canis (4.39%), and Anaplasma platys (3.51%). Additionally, a set of multiplex PCR primers were developed to detect H. canis, Babesia spp. (B. canis and B. vogeli), and E. canis in canine blood. Multiplex and conventional single-reaction PCR results for the 114 dog blood samples were similar, except for one H. canis sample. Multiplex PCR is, therefore, a useful tool in screening infected dogs in veterinary clinics. This study's results, together with those of previous studies in the country, show that canine vector-borne pathogens are an emerging veterinary concern in the Philippines.
