AIM: The present study was undertaken to isolate and standardize the various active phytochemical constituents present in the fruit rinds of Punica granatum. METHODS: Fruit rinds of Punica granatum were dried and extracted with methanol in a static extractor; the percentage yield of the methanolic extract (MEPG) was found to be 26%; the methanolic extract was partitioned using n-butanol, ethyl acetate and water; the percentage yield of the fractions were found to be 17.16%, 26.88% and 47.72% respectively. HPLC was carried out for all the fractions and the markers corresponding to the major peaks in HPLC were isolated using column chromatography. The isolated markers were characterized using UV, IR, Mass and NMR spectroscopy. RESULTS & CONCLUSION: Totally four compounds were isolated and identified as punicalin, punicalagin, gallic acid and ellagic acid based on UV, IR, Mass and NMR data, they were standardized by HPLC. Herbal medicines as the major remedy in traditional system of medicine have been used in medical practices since antiquity. In addition to its ancient historical uses, pomegranate is used in several systems of medicine for a variety of ailments. The objective of the present study was to investigate the presence of various phytochemicals from the ethanolic, aqueous and chloroform extracts of Punica granatum peel, whole fruit and seeds. The three different extracts from peel were found to contain Triterpenoids, Steroids, Glycosides, Flavonoids, Tannins, Carbohydrate & Vitamin C. The three different extracts from whole fruit were found to contain Triterpenoids, Steroids, Glycosides, Saponins, Alkaloids, Flavonoids, Tannins, Carbohydrate & Vitamin C. The three different extracts from seeds were found to contain Triterpenoids, Steroids, Glycosides, Saponins, Alkaloids, Tannins, Carbohydrate & Vitamin C. The generated data from the three different extracts of Punica granatum peel, whole fruit & seeds provided the basis for its wide uses in the traditional & folk medicines.

Punica granatum L. is a native plant that is greatly dispensed in south of Iran. The massive pomace of Punica exhibits a challenging losses exposure difficulty for the processing industries. The resent study was aimed to investigate the composition and quality of P. granatum pomace and understand its bioactive compounds to introduce it to different industries such as pharmaceutical, food, medicinal, agricultural etcetera for optimum use. The SHS (static headspace) GC-MS on the Combi PAL system method was used to identify and analyze the volatile components (VCs). The massive pomaces of Punica granatum L. exhibit a challenging losses exposure difficulty for the processing industries. The resent study was aimed to investigate the bioactive compounds of pomace extracts to introduce it to different industries such as pharmaceutical, food, medicinal, agricultural etcetera for optimum use. Four different extracts were prepared and the phenolic compounds were quantified using HPLC-DAD. Different amounts of phenolic compounds were detected in the samples including gallic acid, catechin, ellagic acid, rosmarinic acid, hesperidin, p-coumaric acid and chlorogenic acid. Gallic acid was major compound in all studied extracts of pomaces, with the maximum amount belonging to water extract (at 60°C). The average amount of gallic acid detected in water extract (at 60°C) of Punica granatum L. was 11.25 mg g-1 dry weight, while it was 3.24 3.02 and 1.09 mg g-1 dry weight for the extracts obtained by distilled water, methanol and methanol 80%, respectively.

Two new compounds, coniferyl 9-O- [ -D-apiofuranosyl(1f6)]-O--D-glucopyranoside (1) and sinapyl 9-O- [ -D-apiofuranosyl(1f6)]-O--D-glucopyranoside (2), were isolated from the seeds of Punica granatum (pomegranate), together with five known compounds, 3,3′-di-O-methylellagic acid (3), 3,3′,4′-tri-Omethylellagic acid (4), phenethyl rutinoside, icariside D 1 , and daucosterol. The structures of 1 and 2 were elucidated by spectroscopic data analysis. Compounds 1-4 exhibited antioxidant activity, which was evaluated by measurement of low-density lipoprotein (LDL) susceptibility to oxidation and by determination in vitro of malondialdehyde (MDA) levels in the rat brain.
