The cytotoxicity of the methanol extract of Origanum syriacum against human cerivical adenocarcinoma, Hela cells, was evaluated by the MTT assay. From the crude extract, a new flavone glycoside, acacetin-7-O-[2-O-α-L-rhamnopyranosyl-6-O-β-D-glucopyranosyl]-β-D-glucopyranoside (1), together with the ten known flavonoids luteolin (2), apigenin (3), luteolin-6-C-glucoside (4), luteolin-3-methylether-6-C-glucoside (5), luteolin-7,4-dimethyether-6-C-glucoside (6), apigenin-7methylether-6-C-glucoside (7), apigenin-7-O-glucoside (8), diosmetin-7-O-glucoside (9), acacetin-7-O-glucoside (10) and acacetin-7-O-rutinoside (11), was isolated from the methanol extract of the aerial parts of Origanum syriacum. The structure elucidation of the isolated compounds was performed by spectroscopic and chromatographic methods.

Polyphenolics of Plumeria rubra L.and Cytotoxic Activities against Lung, Colon, and Breast Carcinoma
Numerous polyphenolic compounds originating from plants are thought to have anticancer and apoptosis-inducing effects on cancer cells. Plumeria rubra (family Apocyanaceae) is utilized as traditional medicine to cure a variety of diseases. It is well known for being a decorative tree, broad cultivated, very fragrant, and having a wide range of secondary metabolites, which inspired us to learn more about its possible medical uses. In this work, the total phenolic content of the methanol extract of P. rubra was established spectrophotometrically and found to be 11.05 mg/g, while the total flavonoid content was found to be 29.22 mg/g. Fifteen polyphenolic compounds were identified by the HPLC analysis of P. rubra methanol leaves extract. Additionally, an in vitro cell viability test was used to assess the cytotoxic activity of the methanol leaves extract of P. rubra. The findings of the cytotoxicity test revealed substantial anticancer activity against the human lung carcinoma A-549 cell line with an IC 50 of 41.72 μg/ml and less activity against the human colon carcinoma HCT-116 and the human breast carcinoma MCF-7 cell line with IC 50 of 57.62 and 75.8 μg/ml, respectively. Results of this work demonstrate that methanol leaves extract of P. rubra is a valuable source of phenolics and flavonoids that that can substantially slow the spread of cancer.
A New Hepatoprotective Flavone Glycoside from the Flowers of Onopordum alexandrinum Growing in Egypt

A bioactivity-guided fractionation of the ethyl acetate fraction of the fl owers of Onopordum alexandrinum L. (Asteraceae) yielded a new fl avonoidal glycoside designated as acacetin-7-O-galacturonide (9), alongside with nine known fl avonoids; 6-methoxy-apigenin (hispidulin) (1), acacetin (2), apigenin (3), luteolin (4), kaempferol (5), eriodictyol , apigenin-7-O-glucoside , luteolin-7-O-glucoside (8), and kaempferol-3-O-rutinoside (10). The compounds were assayed for their hepatoprotective activity against CCl 4 -induced hepatic cell damage in rats and free radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH). Compounds 4, 6, 9, and 10 have not been previously reported from fl owers of O. alexandrinum L., and this is the fi rst report of acacetin-7-O-galacturonide (9) in nature which has also shown signifi cant hepatoprotective and free radical scavenging effects. The isolated compounds were identifi ed using different spectroscopic methods (UV, 1 H NMR, 13 C NMR, HMQC, HMBC, and COSY).
Antimicrobial Activities and Some Flavonoids in Extracts of Some Medicinal Plants
Origanum majorana, Melissa officinalis, Anthemis cotula and Avena sativa were extracted by using 65% ethanol to isolate their active constituents. The antimicrobial activities of extracts were investigated against 15 microorganisms by using the disk diffusion method, MIC (Minimum Inhibitory Concentration), MBC (Minimum Bactericidal Concentration) and MFC (Minimal Fungicidal Concentration) tests. Furthermore, the presence of some flavonoids were analyzed by using HPLC. It was determined flavonoids in the extracts of O. majorana, M. officinalis, A. cotula and A. sativa. As a results it was observed that O. majorana was active against Staphylococcus aureus, Enterococcus faecium, Enterococcus faecalis, Salmonella typhimurium and Pseudomonas aeruginosa, where M. officinalis was active against Enterococcus faecium and Enterococcus faecalis. On the other hand, both A. cotula and A. sativa were observed to be active against Enterococcus faecium. The extracts of plant samples showed antibacterial activity against tested microorganisms at different levels.

BACKGROUND: A detailed phytochemical analysis of Origanum vulgare L. ssp. viride (Boiss.) Hayek was carried out and the antioxidant activities of five different crude extracts were determined. The antiproliferative activities of the extracts were determined using the xCELLigence system (Real Time Cell Analyzer). RESULTS: Differences between the essential oil and volatile organic compound profiles of the plant were shown. The main component of the essential oil was caryophyllene oxide, while the main volatile organic compounds were sabinene and eucalyptol as determined by HS-GC/MS. Phenolic contents of the extracts were determined qualitatively and quantitatively by HPLC/TOF-MS. Ten phenolic compounds were found in the extracts from O. vulgare and Origanum acutidens: rosmarinic acid (in highest abundance), chicoric acid, caffeic acid, p-coumaric acid, gallic acid, quercetin, apigenin-7-glucoside, kaempferol, naringenin and 4-hydroxybenzaldehyde. CONCLUSION: This study provides first results on the antiproliferative and antioxidant properties and detailed phytochemical screening of O. vulgare ssp. viride (Boiss.) Hayek.
Cytotoxic flavonoids from Diplotaxis harra (Forssk.) Boiss. growing in Sinai

As part of our ongoing collaborative effort to discover the anticancer activity of the phenolics isolated from terrestrial plant sources, the EtOH extract of the aerial parts of the Egyptian medicinal plant Diplotaxis harra (Forssk.) Boiss. was in vitro investigated for cytotoxicity against HCT116, HepG2 and MCF-7 cell lines, and resulted with IC 50 = 4.65, 12.60 and 17.90 µg/ml, respectively. Doxorubicin (+ve control) showed in vitro cytotoxic activity with IC 50 = 3.64, 4.57 and 2.97 µg/ml, respectively. The phenolic-rich fraction of the EtOH extract was subjected to further fractionation, which led to the isolation of five flavonoids; identified as quercetin, quercetin 3-O-β-glucoside, isorhamnetin 7-O-β-glucoside, apigenin 3-O-β-rhamnoside and kaempferol 3-O-β-glucoside, according to its' spectral data and comparison with the literature. An investigation of Leptadenia pyrotechnica (Forsk.) Decne (Asclepiadaceace) chemical constituents led to the isolation of six flavonoids, kaempferol-3-O-α-l-rhamnopyranosyl (1999→699)-O-β-d-glucopyranoside (E-I.1), kaempferol-3-O-β-d-rhamnopyranosyl (1999→699)-O-β-d-glucopyranoside (E-I.2), texasin-7-O-β-dglucopyranoside E-II.2, kaempferol-3-O-β-d-glucopyranoside (E-III.1), kaempferol (E-IV.1) and kaempferide-3-O-α-l-rhamnopyranosyl (1999→699)-O-β-d-glucopyranoside (E-I.1a). The isolation of these compounds was carried out using Sephadex LH-20 low pressure liquid chromatography (LPLC), preparative paper chromatography (PC), and high performance liquid chromatography (HPLC). The chemical structures of the isolated compounds were established by mass spectrometry (FAB-and EI-techniques), nuclear magnetic resonance NMR (1 H-, 13 C-and COSY) spectral data and ultraviolet (UV) spectroscopic techniques. The acute toxicity of total alcoholic and total flavonoid extracts were examined by brine shrimp. The LC 50 values were 11.89 and 84.14 ppm for the total alcoholic and total flavonoid extracts, respectively. The mortality rates of the isolated flavonoid fractions of E-I, E-I.1, E-I.2 represent the higher percentages of mortality compared with the rest of the flavonoid fractions. The plant exhibited activity as an antitumor agent in the initial potato disc screen.

Rumex dentatus L. and Rumex vesicarius L., of the family Polygonaceae, are edible herbs growing wild in Egypt. Their lipoid constituents were examined by both liquid chromatography/mass spectrometry (LC/MS) and by gas chromatography/mass spectrometry (GC/MS). Their essential oil compositions consisted mainly of thujene, limonene, fenchon, estragole, and anethole but at largely different concentration. Fatty acid compositions were similar among the two species and consisting of palmitic, oleic, linoleic and linolenic acids, with R. vesicarius containing much higher level of omega-3-fatty acids. Both of the crude lipid extracts of the two species showed strong antioxidant activity as a radical quenching agent against 2,2-diphenyl-1-picrylhydrazyl (DPPH) systems. Origanum species (Lamiaceae) are commenly used as herbal tea, flavoring agents and medicinal plants all over the world. Rosmarinic acid, lithospermic acid, vicenin-2, betulalbuside A, 8-OH-linaloyl glycoside, ursolic acid, oleanolic acid were isolated from the aerial parts of Origanum acutidens in this study. Their structures were identified by means of spectroscopic methods (1D-and 2D-NMR).
