A Fast Track to Indoles and Annulated Indoles through ortho- vs ipso-Amination of Aryl Halides

A complementary site selective ortho-vs ipsoamination of aryl halides using non-electrophilic amine sources for construction of indole scaffolds is reported. A palladium-catalyzed alkyne insertion/C−H activation/palladacycle amination via merger of three easily diversified components including iodoarenes, alkynes, and amines delivers indoles with different substitution patterns even in gram scales. By employing ortho-bromoanilines, a consecutive annulative π-extension of indoles proceeds to construct indolo[1,2-f ]phenanthridine scaffolds via four C−C and C− N bond formations in one pot.
Metal-catalyzed privileged 2- and 3-functionalized indole synthesis*

This review summarizes the metal-catalyzed approaches for the synthesis of privileged 2-and 3-functionalized indoles via the Fischer indole synthesis, cycloaddition reaction, C-H activation reaction, and coupling reactions. Polycyclic derivatives of indole are also listed including triptamine-based pharmaceutical 5-HT1D agonist L 775,606. Some important mechanism studies are described in details in this review. The review covers literature for the last 20 years.
A Novel One-Pot Synthesis of N-Acylindoles from Primary Aromatic Amides

A novel one-pot synthesis of N-acylindoles via tandem cycloalkylation-annelation is described. This approach is based on the use of a strong solid-acid catalyst, montmorillonite K-10, and microwave irradiation under solvent-free conditions. The tandem cycloalkylation of amides and annelation of intermediate pyrroles were completed in minutes and provided good yields with high selectivities for the indoles. The safe, easy to handle catalyst, and the convenience of the product isolation make this process an attractive, environmentally benign alternative for the synthesis of N-acylindoles.

Synthesis of Indoles Derivatives Using Metal Free Catalyst in Organic Reactions

Objective: Indole derivatives are one of the most flexible and common nitrogen-based skeletons and often used in the synthesis of numerous heterocyclic compounds, having biological or medicinal importance. Many indole derivatives have been isolated naturally from plants, fungi, and marine organisms and are highly important due to their pharmaceutical activities. They also play a significant character in polymer and dye industries as well as in the agriculture sector. There exists an immense potential for designing and methodology development for the synthesis of indole based heterocyclic scaffold structure with desire chemical and biomedical relevance. Result and Discussion: In this present review, we outlined the recent applications of indoles in the onepot multi-component reaction using metal free catalysts for the preparation of various heterocyclic scaffolds and their corresponding biological activities are also discussed.
