We report the synthesis of a series of new functionalized bisphosphonates and bisphosphonic acids with an alkoxy group fixed at the geminal carbon, which is proposed to increase their lipophilicity and so their bioavailability. Subsequently, the alkylation of these alkoxy bisphosphonates with allyl bromide is reported. The reactivity of the allyl group has been studied to give access to alkoxy bisphosphonates functionalized by diverse groups including alcohol, aldehyde, carboxylic acid, epoxide and amine.
Synthesis and stereochemistry of 6-membered ring phosphonates

Background: Organophosphorus compounds have important industrial and biomedical applications as pharmaceutical and agrochemical agents, as well as transition state analogs for the production of monoclonal antibodies. Methods: Two diastereomers of a 6-membered ring, cyclic phenyl phosphonate were synthesized in 8 steps from 1,3-butanediol. Results: The stereochemistry of the diastereomers was elucidated on the basis of 3 1 P NMR signals, together with 1 H NMR nuclear Overhauser effects (NOE) difference experiments. Conclusions: Such cyclic phosphonates may have utility serving as transition state analogs for the production of monoclonal antibodies.

A new efficient and practical synthesis of syn-and antib,g-dihydroxyphosphonates 1 and 2 was developed in high diastereoselectivity via reduction of b-ketophosphonates readily obtained from (S)-mandelic acid. An example of "diastereoselective 1,2-induction" is showed and a mechanistic model to explain the stereochemical outcome is proposed. Assignment of the configuration at the new stereogenic centers was achieved by 1 H NMR spectral data of their corresponding acetonides.

The synthesis of a new class of epoxide derivatives from prochiral vinylidene bisphosphonate (VBP) precursors is reported using hydrogen peroxide as the terminal oxidant. The reaction is carried out using a series of possible organic activators having a basic character, with the best results being observed using quinine and sparteine. These activators not only provide from good to excellent epoxide yields with a large variety of VBPs, but also interesting enantioselectivities in the 67%-96% ee range, at least in the case of the Ph and m-MeO-Ph VBP derivatives, opening the way to a number of chiral anti-osteoporosis potentially active pharmaceutical ingredients.

Bisphosphonates (BPs), well-known medicinal compounds used for osteoporosis management, are currently the target of intensive research, from basic pre-formulation studies to more advanced stages of clinical practice. The high demand by the pharmaceutical industry inherently requires an easy, efficient and quick preparation of BPs. Current synthetic procedures are, however, still far from ideal. This work presents a comprehensive compilation of reports on the synthesis of the commercially available bisphosphonates that are pharmaceutical active ingredients. Current limitations to the conventional synthesis are assessed, and paths towards their improvement are described, either through the use of alternative solvents and/or by selecting appropriate ratios of the reactants. Innovative processes, such as microwave-assisted synthesis, are presented as more environmental-friendly and effective methods. Two simple and efficient one-pot procedures for the synthesis of a series of -branched N-heterocycle-substituted methane-1,1-bisphosphonates are outlined. In the first method, the parent halosubstrates were reacted with cyanomethylphosphonate followed by reaction with dialkyl phosphonates to give asymmetrical or symmetrical bisphosphonates (BPs). In the second approach, the same halocompounds were reacted with tetraethyl methyl-1,1-bisphosphonate to give the requisite BPs. Partial and complete hydrolysis of the prepared BPs were also investigated. Recent Synthesis and Transformation of Vinylphosphonates
Vinylphosphonates are an important group of compounds. This review summarizes recent developments in the synthesis and transformations of vinylphosphonates. Owing to the importance of vinylphosphonates, many different synthetic methods have been developed. Especially useful are various organometallic synthetic methods utilizing the chemistry of copper, titanium, zirconium, and lithium, etc. The transformations of vinylphosphonates is also described and include expoxidation, cycloaddition, Suzuki coupling, and Diels-Alder reactions.
