Fluorescent nanoparticles as selective Cu(ii) sensors

Latex nanoparticles functionalized with cyclam (1,4,8,11-tetraazacyclotetradecane), a copper chelator, have been doped with a fluorescent dye 1 (BODIPY derivative: 4,4-difluoro-8-(2 ,4 ,6-trimethyl)phenyl-2,6-diethyl-1,3,5,7-tetramethyl-4-bora-3a,4a-diaza-s-indacene). The bulky, hydrophobic fluorophore 1 dissolves within the nanoparticles' polymer core up to a concentration of about 88.4 lmol g −1. At this concentration the fluorescence yield is about 0.80. Adding Cu 2+ ions to the solution decreases the fluorescence because of the energy transfer between the dye and the violet copper cyclam complexes. The response is fast: 90% of the quenching occurs within 1 s. The Cu 2+ detection threshold is of 1 nanomolar. No interferences were observed with zinc and nickel ions.

This paper describes the preparation of silica nanoparticles doped with a molecularly engineered luminescent Ru(II) complex for the selective determination of Cu(II) ions in aqueous media. Initially, three novel indicator dyes, namely [Ru(phen)2(L)] 2+ , where L is either hmpi (2-(1H-imidazo[4,5-f]-1,10-phenanthrolin-2-yl)-4-methoxyphenol), haip (1-(4-hydroxy-3-(1H-imidazo[4,5-f]-1,10-phenanthrolin-2-yl)phenyl)ethanone), or btym (N,N'-(2,2'bipyridine-4,4'-diyl)bis(2-(thymin-1-yl)acetamide)), were synthesised, characterised and their 2 response to metal ions was evaluated. The [Ru(phen)2(haip)] 2+ complex demonstrated to be selective to Cu(II), so that it was embedded in 20-nm silica nanoparticles, synthesised by the Stöber method. The maximum sensitivity of the luminescent nanosensor was obtained when the indicator dye was incorporated 8 h after the beginning of the 10-h silane condensation process, because interaction with the metal ions improves when the encapsulated luminophore molecules are closer to the nanoparticle outermost surface The nanosensor showed selective response to Cu(II) in the presence of Zn(II), Hg(II), Pb(II), Ni(II), Cd(II) and Fe(II) ions in 50 mM phosphate buffer at pH 8.0, with a limit of detection of 0.30 mol L-1 , RSD values lower than 3.0%, and a response time < 30 s. The nanosensor was applied to the determination of Cu(II) in mineral and tap waters spiked with 5.0 and 7.0 mol L-1 of the analyte, with recoveries from 88 to 110 %.

A luminescent cadmium-pamoate metal-organic framework, [Cd 2 (PAM) 2 (dpe) 2 (H 2 O) 2 ]•0.5(dpe) (1), has been synthesized under hydrothermal conditions by using p-electron-rich ligands 4,4'-methylenebis(3-hydroxy-2-naphthalenecarboxylic acid) (H 2 PAM) and 1,2-di(4-pyridyl)ethylene (dpe). Its structure is composed of both mononuclear and dinuclear Cd II building units, which are linked by the PAM and dpe ligands, resulting in a (4,8)-connected 3D framework. The p-conjugated dpe guests are located in a 1D channel of 1. The strong emission of 1 could be quenched efficiently by trace amounts of 2,4,6-trinitrophenol (TNP), even in the presence of other competing analogues such as 4-nitrophenol, 2,6-dinitrotoluene, 2,4-dinitrotoluene, nitrobenzene, 1,3-dinitrobenzene, hydroquinone, dimethylbenzene, and bromobenzene. The high sensitivity and selectivity of the fluorescence response of 1 to TNP shows that this framework could be used as an excellent sensor for identifying and quantifying TNP. A facile and label-free detection method based on the formation of the G-quadruplex structure of Thioflavin T and a DNA sequence was developed for rapid and sensitive detection of copper (Cu 2+) ion. The DNA sequence with many guanine (G) repeats has the potential to form a G-quadruplex structure and exhibits high selectivity to Cu 2+. ThT promotes the formation of the G-quadruplex structure resulting in high fluorescence intensity because of its fluorescence molecular rotor characteristic. As opposed to, when the DNA sequence binds to its target in the presence of Cu 2+ , conformational changes occur, resulting in ThT dissociation and a substantial decrease in fluorescence intensity. By virtue of this change, unlabeled, low-cost, sensitive, and selective detection of Cu 2+ was achieved and not interfered by many other ions. The proposed method has high sensitivity and selectivity, with a detection limit of 30.7 nM, allowing it to directly measure Cu 2+ in contaminated water samples in a wide linear range from 0.1 to 100 µM with a high recovery rate.
Fluorescent peptide sensor for the selective detection of Cu2+

A new fluorescent peptidyl chemosensor for Cu(2+) ions with fluorescence resonance energy transfer (FRET) capabilities has been synthesized via Fmoc solid-phase peptide synthesis. The metal chelating unit, which is flanked by the fluorophores tryptophan (donor) and dansyl chloride (acceptor), consists of the amino acids glycine and aspartic acid (Gly-Gly-Asp-Gly-Gly-Asp-Gly-Gly-Asp-Gly-Gly-Asp-Gly-Gly). Coordination of the Cu(2+) ions to the metal chelating unit results in fluorescent quenching of both the donor and acceptor fluorophores. Although it was determined that Cu(2+) binding causes no change in FRET efficiency, emission and Cu(2+)-induced quenching of the acceptor dye can be used to monitor the concentration of the copper ions, with a detection limit of 32microgL(-1). The sensor also demonstrated sensitivity, reversibility and selectivity towards Cu(2+) in a transition metal matrix at pH 7.0.

A new fluorescent electrospun fiber polymer 1-(2-Pyridylazo)-2-naphthol (PAN) − poly(acrylic acid) (PAA) previously synthesized by covalent functionalization of PAN into PAA and electrospinning technique was applied for Cd 2+ probe via fluorescence enhancement due to coordination of the metal ion. The fluorescent indicator molecule (PAN−PAA) exhibits distinct and well-defined emission peak. Emission maximum of the functionalized polymer molecule appeared at 593 nm which is indicative of a good Stokes’ shift. In order to get clearer insight into the florescence of the functionalized polymer, the fluorescence behavior was measured with addition of various metal (Cd 2+ ,Cr 3+ ,Co 2+ ,Fe 2+ andNi 2+ ). The fluorescence “turn-on” effect of PAN−PAA upon addition of Cd 2+ was predominant. A simple, environment friendly fluorescent turn-off and colorimetric sensor for the rapid detection of Cu 2+ ions in aqueous solution by using organic Schiff base ligand benzildihydrazone-N,N'-bis(2-hydroxy-4-diethylamino-1-formylbenzene) (L) has been developed. L exhibited binding-induced color change from yellow to light red. The obtained L-Cu 2+ complex could be used as a new cascade sensor for detecting cyanide anion (CN-). The corresponding detection limits were found to be 7.3×10 −10 M for Cu 2+ and 1.5×10 −7 M for CN-.
