Antitumor therapy, including adoptive immunotherapy, inevitably faces powerful counteraction from advanced cancer. If hematological malignancies are currently amenable to therapy with CAR-T lymphocytes (T-cells modified by the chimeric antigen receptor), solid tumors, unfortunately, show a significantly higher degree of resistance to this type of therapy. As recent studies show, the leading role in the escape of solid tumors from the cytotoxic activity of immune cells belongs to the tumor microenvironment (TME). TME consists of several types of cells, including neutrophils, the most numerous cells of the immune system. Recent studies show that the development of the tumor and its ability to metastasize directly affect the extracellular traps of neutrophils (neutrophil extracellular traps, NETs) formed as a result of the response to tumor stimuli. NAD(P)H: FMN-oxidoreductase functioning under macromolecular crowding: in vitro modeling

Изучено функционирование НАД(Ф)H : ФМН-оксидоредуктазы (Red) из Vibrio fischeri в условиях макромолекулярного краудинга (ММК), смоделированных in vitro добавлением биополимеров крахмала и желатина. Рассчитаны значения констант скорости и энергий активации диссоциации Red на субъединицы, проанализирован процесс денатурации Red. Показано, что функционирование Red в условиях ММК не отличается от такового в разбавленных растворах. Полученный результат опровергает распространённое убеждение в том, что в условиях in vivo благодаря ММК происходит стабилизация нативной конформации ферментов по сравнению с условиями in vitro.

Ca2+-transporting systems of exocrine glands secretory cells

In this rewiew we discuss present concepts of functioning of Ca2+-transporting systems in se-cretory cells of exocrine glands. In details are described voltage-operated, receptor-operated and store-operated Ca2+-channals of plasma membrane, ryanodine receptors, inositol-1,4,5-receptors of endoplasmic reticulum, Ca2+-pumps of plasma membrane and endoplasmic reticulum and Na+-Ca2+-exchanger of plasma membrane. Special attention is paid to cooperation of Ca2+-transporting sys-tems and to generation and propagation of calcium signals in secretory cells.

Synergetic inhibition of the brain mitochondrial NADH:ubiquinone oxidoreductase (complex I) by fatty acids and Ca2+

The NADH:ubiquinone oxidoreductase (respiratory complex I) activity of inside out pig brain submitochondri al particles is inhibited by endogenous or externally added free fatty acids in time dependent fashion. The rate and degree of the inhibition is dramatically increased by Ca 2+. The Ca 2+ promoted, fatty acid induced inhibition is pH dependent, this being particularly evident at pH > 8.0. The inhibition is completely reversed by either EGTA or by bovine serum albumin (BSA). BSA prevents previously described (Kotlyar, A. B., Sled, V. D., and Vinogradov, A. D. (1992) Biochim. Biophys. Acta, 1098, 144 150) inhibitory effect of Ca 2+ and alkaline pH on the de active to active form transition of complex I. A possi ble mechanism of synergetic inhibition on complex I by Ca 2+ and fatty acids is discussed.

Cа2+ TRANSPORTING SYSTEMS IN SECRETORY CELLS OF THE RAT EXORBITAL LACRIMAL GLAND І. Ca2+ PUMPS OF PLASMA MEMBRANE AND ENDOPLASMIC RETICULUM

The mechanism of secretion by lacrimal glands, the innervation and orientationeffects, and the mechanism of transduction of primary agonists are similar to the major salivary glands. However, unlike the major salivary glands, about organization of Ca2+ signalling and Ca2+-transporting systems contribution to Ca2+ homeostasis maintenance in secretory cells of the lacrimal glands is not totally clear yet. The purpose of the work was to investigate the role of plasma membrane (PMCA) and endoplasmic reticulum (SERCA) Ca2+ pumps in maintenance of Ca2+ homeostasis in the secretory cells of the lacrimal glands. The study was performed on intact and digitonin-permeabilized secretory cells of exorbital lacrimal glands of rats. The functioning of the Ca2+-transporting systems was estimated by changes of Ca2+ content in cells, after incubation with agonists or antagonists.
