Unani System of medicine is one among the oldest systems that prevails till date with its efficient drugs derived from animal, plant and mineral resources. Over 2400 years ago the father of medicine, Hippocrates practiced it, however His medicine included a great deal of ancient Egyptian medicine as well as important components of the ancient Mesopotamian traditions. In Unani system of medicine, arthritis is described under a broad term Waja-ul-Mafasil which encompasses entire joint disorders like inflammatory, non-inflammatory, infectious, metabolic and other musculoskeletal disorders. Background: The conventional drugs used for gout are full of adverse effects (hepatotoxicity, nephrotoxicity, haematological toxicity). The need of the time is to develop evidenced based safe and \cost effective hypouricemic agent which can easily counteract treatment-failure gout and minimize the suffering of the patients and improve their quality of life. Antioxidant and Anti-inflammatory Properties of a Nontoxic Herbal formulation – GSTC
Abstract: Chronic inflammation is an etiological factor in most human degenerative diseases and oxidative stress, secondary to the inflammation is thought to contribute to the severity. Therefore, molecules having antioxidant and anti-inflammatory properties with lesser side effects are indeed essential. It has been recommended that combinations of different antioxidants may have a better biological efficacy than a single drug. Present study envisions a synergistic or additive effect of four medicinal plants, Commiphora mukul (Hook. ex Stocks.) Eng. (Burseraceae), Salacia reticulata Wight (Celastraceae),Terminalia arjuna (Roxb.) Wight & Arn (Combretaceae) and Curcuma longa Linn (Zingiberaceae) in a formulation (GSTC) at equimolar concentrations. In the preliminary analysis, GSTC was found to scavenge superoxide, hydroxyl, DPPH radicals and inhibited lipoxygenase mediated oxygenation of linoleic acid and peroxyl radical mediated peroxidation of human erythrocyte membrane lipids in vitro. In rats, acute oral dose of GSTC upto 2 g/kg bwt was nontoxic and at a concentration of 100 mg/kg bwt, reversed sodium fluoride mediated decrease in antioxidant molecules and enzymes activity. Further, GSTC at doses of 250 and 500 mg/kg bwt orally, reduced carrageenan induced paw oedema in mice.Overall, GSTC having considerably high antioxidant and anti-inflammatory potential together with its nontoxic nature finds promise as a therapeutic drug candidate in various chronic inflammatory diseases. Wajaul Mafasil (joint pain) is the most prevalent and common cause of morbidity and disability not only in India but also throughout the world. In spite of various scientific developments in conventional medicine, there is no specific therapy or cure available for joints pain particularly when the disease has progressed to chronic stage. Therefore, it is the need of the hour to explore new formulations from herbal sources which are safe and effective. The present trial was undertaken to scientifically validate the Unani pharmacopoeial compound formulations of Habb-e-Suranjan and Raughan-e-Suranjan for its safety and efficacy in patients with joints pain. The awareness and use of Traditional Siddha Indian Medicine is on the increase in this globalized world. The specialty lies in its therapeutic potential in curing chronic diseases such as Arthritis, dermatological conditions, life style disorders etc. The usage of NSAID in the treatment of painful musculo-skeletal conditions often results in adverse effects such as gastric irritation, renal damage etc. On the other hand, poly herbal medicines are safe, effective, time-tested and devoid of drastic side-effects. Siddha Medicine has many such herbal medicines indicated for the treatment of Vatha (arthritis). This research paper deals with the In-vitro Anti-inflammatory screening of such a medicine documented in Classic Siddha text, ‘Yugi Muni Vaithiya Kaaviyam’ and specially indicated for ‘Kudaithiri Vaatham’ – which can be correlated to a kind of Rheumatism attended with pain all over the body especially in the loins. In our study we reproduced an inflammatory state by treating THP-1 cells (human myelomonocytic leukemia) with pro-inflammatory stimuli, such as LPS obtaining an up-regulation in the expression and in the activity of nitrate level. Our results show a significant increase in the expression and activity of Tissue nitrite level when cells were treated with the Test drug in different concentrations.
