Abstract

OBJECTIVE: To determine ancestry informative markers, haplogroups and frequency of HLA-DRB1 alleles in patients with Multiple Sclerosis (MS) in Bogota, Colombia. BACKGROUND: In European-descended populations the HLA-DRB1*15 polymorphism has been identified as the strongest genetic risk factor for MS, whereas the HLA-DRB1*14 allele attenuates MS risk. However, the frequency and strength of association of these alleles have not been reported in patients from the low prevalence and highly admixed region of Bogota, Colombia. DESIGN/METHODS: Genomic DNA was isolated and purified from blood samples by a standard salting out procedure. HLA-DRB1 alleles genotyping was done using Single Specific Primer Polymerase Chain Reaction. Mitochondrial hypervariable region-1 was amplified and haplogroups were determined using HaploGrep software. Ancestry proportions were estimated genotyping a panel of ancestry informative markers.
