The potential of Kebar grass extract on histopathological changes in kidneys of mice offspring from the parent exposed to carbofuran during the lactation period

This research aims to know the potential of kebar grass extract (Biophytum petersianum) in reducing the impact of kidney damage in mice offspring during the lactation period from parents exposed to carbofuran. Carbofuran, kebar grass extract, and vitamin C are exposed orally postnatal day 1 st until the 14 th postnatal day after delivery. Forty-two mice adults in the lactation period were randomly into seven groups. This group consisted of K (aquadest control), P1 (carbofuran ¼ LD50 0.0125 mg/day), P2 (carbofuran 1/8 LD50 0.00625 mg/day), P3 (kebar grass extract 3.375 mg + carbofuran ¼ LD50), P4 (kebar grass extract 3.375 mg + carbofuran 1/8LD50), P5 (vitamin C 0.2 cc + carbofuran ¼ LD50), and P6 (vitamin C 0.2 cc + carbofuran 1/8 LD50). On the 15 th day of experimental, offspring were randomly selected and dissected, and the kidney was taken to make a histopathology sample. This study shows that carbofuran caused increased tubular degeneration, necrosis, and inflammation. Kebar grass extract could decrease tubular degeneration, necrosis, and inflammation. Kebar grass extract was more effective than vitamin C in reducing kidney damage in mice offspring during the lactation period from parents exposed to carbofuran.

Kebar grass contains active antioxidants and potential vitamins to neutralize TCDD toxicity. Prove that Kebar grass extract in various dosage can maintain viability, motility, and sperm concentration of male mice exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin. This study was an experimental laboratory study with five groups consists of Negative Control (C-), Positive Control (K +) with TCDD exposure of 0.7μg/Kg BW IP single dose. The acute toxicity (LD50) of insecticides carbofuran and its effects on mal e reproduction in rats were carried out. Cypermethrin is an insecticide synthetic pyrethroid type that can disrupt the reproductive system. Cypermethrin is also capable of producing reactive oxygene species (ROS) so it is responsible for causing oxidative stress. Green tea (Camellia sinensis) is containing polyphenols that inhibit hydroxyl radicals and prooksidan enzymes. This study aims to prove that green tea ethanolic extract can increase FSH level and antral follicle count in female rats exposed to cypermethrin. The study was true experimental on 25 wistar female rats (Rattus norvegicus) which were divided into 5 groups; KN: no treatment, KP: cypermethrin 20 mg/kgBW/day, P1: cypermethrin 20 mg/kgBW/day + green tea ethanolic extract 7 mg/kgBW/day, P2: cypermethrin 20 mg/kgBW/day + green tea ethanolic extract 14 mg/kgBW/day, P3: cypermethrin 20 mg/kgBW/day + green tea ethanolic extract 28 mg/kgBW/day orally for 28 days. FSH level was measured by ELISA method and ovarian organ were taken to check antral follicle count with hematoxylin eosin staining method. FSH and antral follicle count in female rats showed a positive and strong with correlation coefficient of 0.742, which means an increase in FSH followed by an increase antral follicle count. There was a significantly difference in the level of FSH (p=0.005) and antral follicle count (p=0.001) between the control group and the treatment group. The green tea ethanolic extract can increase the FSH level and antral follicle count in female rats exposed to cypermethrin.

Uterine Performance of Rats Exposed to Cigarette Post-Therapy Ethanol Extract of Kebar Grass (Biophytum petersianum Klotzsch)

This study aims to determine the effect of giving ethanol extract of kebar grass (Biophytum petersianum Klotzsch) on the uterine performance of Rattus norvegicus rats exposed to cigarette smoke. The experimental design used was a completely randomized design (CRD). The results obtained were analyzed by Analysis of Variance (ANOVA) then continued with Duncan's test at a significant level of α = 0.05 using SAS software. The sample consisted of 12 rats, which were divided into 4 groups. The rat group (+), 0.067, 0.0135, decreased uterine function by exposure to 10 cigarettes/head/day for 28 days. The aim of this study was to determine the efficiency of the progestagen containing intravaginal sponges used for 6 or 12 days at timed artificial insemination (TAI) in Hair Goats during the transition period, the effect of GnRH administrations at the time of TAI on fertility was also investigated. In this study 104 lactating hair goats aged between 2-5 years old minimally primiparous was used as animal materials. All of the goats received 30 mg fluorogestone acetate (FGA) impregnated vaginal sponges. The sponges were inserted into vagina for 6 days in the short-term (ST, n=52) group also for 12 days in the long-term (LT, n=52) group. The goats received 400 IU PMSG and 0.075 mg of cloprostenol at the time of sponge removal. The goats in both the ST and LT groups timed artificially inseminated 48 h after sponge withdrawal with fresh cooled semen. At the time of artificial insemination, the subgroups ST1 (n=24) and LT1 (n=22) were maintained as controls, the subgroups ST2 (n=24) and LT2 (n=23) received 5 mcg of buserelin acetate. Pregnancy and twinning rates of the ST1, ST2, LT1 and LT2 groups were 37.5%, 41.6%, 40.9%, 47.8% and 22%, 30%, 11%, 18%, respectively. The litter sizes were 122%, 130%,111% and 118% in the ST1, ST2, LT1 and LT2 groups, respectively. The parturition rates were 100% in all treatment groups. Pregnancy rates, twinning rates and litter sizes were not statistically different among the groups (P>0.05). The results of this study indicated that, a TAI protocol without detection estrus seems to be a suitable administration for short and long progesterone sponges aplications in lactating hair goats during the transition period. In addition, progesteron and AI without estrus detection procedure may be useful alternative to traditional synchronization program using progesteron and AI after estrus detection. Also, GnRH addition to progesterone sponges did not improve reproductive paramaters.

Effect of Kebar Grass (Biophytum petersianum) Extract on the Seminiferous Tubules in Male Mice (Mus musculus) Treated With 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)

The aim of this research was to analyze the effect of kebar grass on the seminiferous tubules diameter and epithelium thickness of mice exposed to 2,3,7,8 tetrachlorodibenzo-p-dioxin (TCDD). Twenty five BALB/C mice were used with five treatments (4 mice each group): Negative Control (C-), Positive Control (C+) with TCDD 7µg/kgBW IP, and treatment groups that treated with TCDD 7µg/kgBW IP and Kebar Grass orally for 53 days were presented T1 0,045mg/gBW/day, T2 0,080mg/gBW/day, and T3 0,135mg/gBW/day. Results showed that TCDD decreased the diameter and epithelium thickness of seminiferous tubules, while kebar grass extract was proven to maintain diameters but could not maintain epithelium thickness of of the seminiferous tubules.

Effect of Carbofuran Pesticide on Male reproductive organs, Semen Analysis and Thyroid

The present study aimed to investigate the effect of Carbofuran pesticide on male reproductive organs, semen analysis and thyroid gland in rats. Oral administration of Carbofuran in doses of 4.2 and 10.5 mg/kg. B.Wt. to male rats daily for 65 successive days significantly increased the weight of testes and decreased the weight of prostates and epididymis. Epididymal sperm characters such as sperm motility and concentration were reduced, while sperm abnormalities were increased. The activity of testicular enzymes such as lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and gamma glutamyltransferase (γ-GT) were increased, while the activity of acid phosphatase was decreased. Carbofuran increased the weight of thyroid gland and plasma level of thyroid stimulating hormones (TSH), while triiodothyronine (T3) and thyroxine (T4) levels were reduced. Biochemical analysis revealed that Carbofuran decreased serum glucose level, AST and ALT enzymes, while total cholesterol was increased. Thus Carbofuran has been shown to induce marked functional changes in male reproductive organs and thyroid gland of rats.
