- Nearly all mitochondrial proteins are synthesized in the cell's cytosol and must make their way into the mitochondria through connected complexes, first through TOM and then through TIM23. Scientists in the Youle lab at #NINDSIntramural have now developed a tool to study how quality is controlled as proteins travel from the cytosol into the mitochondria. Their tool clogs TOM, stalling protein import and leaving TIM23 unoccupied. They discovered that this clog activates a clean-up protein, YME1L1 that senses TIM23 complex is unoccupied and breaks down its components. This shows that mitochondrial quality control includes a breakdown system that prevents these channels from remaining unoccupied in the inner membrane.
