This active optics system has allowed us to correct low-order aberrations identified by several wave-front analysis techniques.

Excitation-secretion uncoupling peptides (ESUPs) are inhibitors of Ca 2؉-dependent exocytosis in ... more Excitation-secretion uncoupling peptides (ESUPs) are inhibitors of Ca 2؉-dependent exocytosis in neural and endocrine cells. Their mechanism of action, however, remains elusive. We report that ESUP-A, a 20-mer peptide patterned after the C terminus of SNAP-25 (synaptosomal associated protein of 25 kDa) and containing the cleavage sequence for botulinum neurotoxin A (BoNT A), abrogates the slow, ATP-dependent component of the exocytotic pathway, without affecting the fast, ATP-independent, Ca 2؉-mediated fusion event. Ultrastructural analysis indicates that ESUP-A induces a drastic accumulation of dense-core vesicles near the plasma membrane, mimicking the effect of BoNT A. Together, these findings argue in favor of the notion that ESUP-A inhibits ATP-primed exocytosis by blocking vesicle docking. Identification of blocking peptides which mimic sequences that bind to complementary partner domains on interacting proteins of the exocytotic machinery provides new pharmacological tools to dissect the molecular and mechanistic details of neurosecretion. Our findings may assist in developing ESUPs as substitute drugs to BoNTs for the treatment of spasmodic disorders.

A Neospora caninum 17 kDa protein fraction (p17) has been described as an immunodominant antigen ... more A Neospora caninum 17 kDa protein fraction (p17) has been described as an immunodominant antigen (IDA) under reducing and non-reducing conditions. The aim of the present study was to investigate the diagnostic utility of p17 in cattle. In order to achieve this, p17 was purified by electroelution from whole N. caninum tachyzoite soluble extract and a p17-based Western blot (WB-p17) was developed. The p17 recognition was measured by densitometry and expressed as OD values to check the validity of the WB-p17. A total of 131 sera including sequential samples from naturally-and experimentally-infected calves and breeding cattle were analysed by WB-p17 and compared with IFAT using whole formalin-fixed tachyzoites as a reference test. The results obtained highlight the feasibility of using the N. caninum p17 in a diagnostic test in cattle. Firstly, the assay based on the p-17 antigen discriminated between known positive and negative sera from different cattle populations, breeding cattle and calves. Secondly, the p17 antigen detected fluctuations in the antibody levels and seroconversion in naturally-and experimentally-infected cattle. Significant differences in p-17 antigen recognition were observed between naturally infected aborting and non-aborting cattle, as well as significant antibody fluctuations over time in experimentally infected cattle, which varied between groups. Furthermore, the results obtained with WB-p17 are in accordance with the results obtained with the IFAT, as high agreement values were obtained when all bovine subpopulations were included (κ = 0.86).

Determinación de la confiabilidad de una prueba para la medición del dolor en adultas mayores. Revista de Ciencias del Ejercicio y la Salud, 1(1), 42-49. Se realizó un estudio para determinar la confiabilidad de una prueba para medir dolor en adultas mayores. Se utilizó un grupo de 12 adultas mayores (N = 12), con una edad promedio de 63.58 años (D.S. ± 10.08). Se realizó un pretest para observar el nivel de dolor inicial de las sujetos en las rodillas, el cual consistió en ejecutar cuatro movimientos (poniéndose de pie, caminando, subiendo gradas y bajando gradas). Después de realizar un solo movimiento, la ejecutante indicaba cuánto dolor sentía entre 0 (sin dolor) y 10 (dolor severo); posteriormente, después de estar en reposo durante dos horas, se les aplicó el post test, en donde se repitió todo el proceso anterior. Para observar si existieron diferencias estadísticamente significativas entre los promedios de ambas mediciones, fue aplicada una ANOVA de una vía de medidas repetidas y, para obtener la confiabilidad de la prueba, se aplicó el coeficiente r de Pearson. Los resultados no mostraron diferencias estadísticamente significativas entre los promedios de ambas mediciones para 11 g.l. y una p < .001. La prueba r de Pearson mostró que los coeficientes obtenidos para los cuatro movimientos (poniéndose de pie = .73, caminando = .77, subiendo gradas = .92 y bajando gradas = .75) fueron mayores que el valor crítico de la Tabla de Pearson (11 g.l., α = .01, valor crítico = .634). Se concluye que el dolor es medible bajo condiciones controladas y que la prueba de dolor es realmente confiable para cada uno de los cuatro movimientos ejecutados.
