Chlorpyrifos (CPF) can induce ovarian damage and reproductive dysfunction through oxidative stres... more Chlorpyrifos (CPF) can induce ovarian damage and reproductive dysfunction through oxidative stress mechanisms. These can be reversed by antioxidants such as Epigallocatechin-3-gallate (EGCG) in green tea. This study aimed to investigate the toxicity effects of CPF on rat ovaries and the effectiveness of EGCG to protect against these effects. Fifty adult female albino rats were randomly assigned to five equal groups; negative and positive control groups, EGCG-treated group, CPFtreated group and CPF+EGCG-treated group. After four weeks the rat body weight and relative ovarian weight were estimated, and blood samples were collected to assess reproductive hormones (RHs) levels. Evaluation of ovarian oxidative stress indicators malondialdhyde (MDA), nitric oxide (NO), superoxide dismutase (SOD), reduced glutathione (GSH) and catalase (CAT) was performed. Ovarian sections were prepared and examined using the hematoxylin and eosin stain, proliferating cell nuclear antigen, and morphometric study to assess histopathologic changes. CPFtreated rats had significant lower body weight and relative ovarian weight compared to controls (P<0.001) and those treated with CPF+EGCG (P<0.001). CPF induced significant reduction in RHs levels and ovarian SOD, GSH and CAT activities when given alone, which were improved by the combined administration of CPF and EGCG. The levels of ovarian MDA and NO were significantly higher in CPF-treated rats than controls and rats treated with CPF+EGCG. The cellular proliferation in ovarian Graafian follicles was significantly suppressed in CPF-treated animals. Hence CPF can induce oxidative damage in rat ovaries, EGCG, a natural potent antioxidant, can be useful against the CPF toxicity effects in ovaries.

So, a rat model of chronic O3 inhalation was established to assess the biochemical and morphological alterations in the lung tissue and to investigate the ameliorative effects of bone marrow–derived mesenchymal stem cells (BMSCs) with or without hypoxia pre-treatment. Forty-two adult male albino rats were divided into four groups: control, ozone-exposed, normoxic BMSC-treated, and hypoxic BMSC-treated groups. METHODS Thirty rats were equally divided into three groups: sham-operated (SHAM), ovariectomized (OVX), and ovariectomized stem cells injected (OVX + hUCB-MSCs). Expressions of α-SMA, AQP1, Sca-1, PCNA, ssDNA, and caspase-3 were determined. Homing of hUCB-MSCs was detected by fluorescence microscopy and examination of immunostained sections for human CD105 and CD34 were performed. Thus, we evaluated the protective effect of hypoxia-preconditioned human umbilical cord blood-derived mesenchymal stem cells (hUCB-MSCs) on hypoglycemic testicular injury. We examined 56 testes from 28 animals: 7 rats with insulin-induced hypoglycemia (HG group), 7 hypoglycemic rats which received an intratesticular injection of hUCB-MSCs (HG-MSC group), and 14 untreated control rats. Testosterone level, testicular catalase (CAT) activity, and malondialdehyde (MDA) level were analyzed. Immunostaining for specific testicular germ and somatic cell markers was performed. Platelet-rich plasma (PRP) has been widely used for diabetic wound healing due to its high content of growth factors. Background: Corneal alkali burns are common ophthalmic emergencies which may lead to permanent vi... more Background: Corneal alkali burns are common ophthalmic emergencies which may lead to permanent visual impairment. In spite of great advances made in the treatment of these cases, the structural and functional restoration of corneal alkali burns remains challenging. Bone marrow-derived mesenchymal stem cells (BM-MSCs) have been demonstrated to enhance corneal healing. However the problem remains in finding a suitable vehicle to deliver BM-MSCs to the site of injury. Objectives: To evaluate the therapeutic potential of BM-MSCs loaded on two biological vehicles (amniotic membrane versus platelet-rich plasma gel) in corneal alkali burns. Materials & Methods: Fifty rats were randomly divided into five equal groups. Group I (control group), group II (corneal ulcer group) and group III (recovery group). Group IV rats with corneal ulcers were treated with amniotic membrane loaded with BM-MSCs while in the group V the rats with corneal ulcers were treated with platelet-rich plasma cells loaded with BM-MSCs. The corneal specimens were processed for the light microscope, transmission electron microscope examination, and RT qPCR assay for VEGF and iNOS. Results: Group II showed epithelial separation and denudation with disorganized stroma and disrupted Descemet's membrane and endothelium. Group III showed residual epithelial separation and vacuolation with neovascularization of the disorganized stroma. Group IV showed thinning of corneal epithelium with inflammatory infiltration of the stroma. Group V corneas revealed restoration of corneal epithelial thickness, organized stroma and continuous Descemet's membrane. Also, there was an upregulation of iNOS & VEGF gene expression in groups II and III which was downregulated in group IV and V. Conclusions: The combination of BM-MSCs with PRP gel was a promising treatment for corneal alkali burns and may be applicable for other types of corneal disorders.

In spite of great advances made in the treatment of these cases, the structural and functional restoration of corneal alkali burns remains challenging. Bone marrow-derived mesenchymal stem cells (BM-MSCs) have been demonstrated to enhance corneal healing. However the problem remains in finding a suitable vehicle to deliver BM-MSCs to the site of injury. Objectives: To evaluate the therapeutic potential of BM-MSCs loaded on two biological vehicles (amniotic membrane versus platelet-rich plasma gel) in corneal alkali burns. Materials & Methods: Fifty rats were randomly divided into five equal groups. Group I (control group), group II (corneal ulcer group) and group III (recovery group). Group IV rats with corneal ulcers were treated with amniotic membrane loaded with BM-MSCs while in the group V the rats with corneal ulcers were treated with platelet-rich plasma cells loaded with BM-MSCs. The corneal specimens were processed for the light microscope, transmission electron microscope examination, and RT qPCR assay for VEGF and iNOS. Results: Group II showed epithelial separation and denudation with disorganized stroma and disrupted Descemet's membrane and endothelium. Group III showed residual epithelial separation and vacuolation with neovascularization of the disorganized stroma. Group IV showed thinning of corneal epithelium with inflammatory infiltration of the stroma. Group V corneas revealed restoration of corneal epithelial thickness, organized stroma and continuous Descemet's membrane. Also, there was an upregulation of iNOS & VEGF gene expression in groups II and III which was downregulated in group IV and V. Conclusions: The combination of BM-MSCs with PRP gel was a promising treatment for corneal alkali burns and may be applicable for other types of corneal disorders.
