DMEM + 2mM Glutamine + 100-200ng/ml/day Fibroblast Growth Factor (FGF) + 10% Fetal Bovine Serum (FBS). Pre-soak flasks in 0.1% gelatin for over 2 hrs and wash with PBS. Split confluent cultures 1:2 to 1:3 i.e. seeding at 3-5x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2 37C.
Passages: 12; The cells have an absolute dependency on fibroblast growth factor for survival. The cells are BVDV negative. Fresh Fibroblast growth factor (FGF) should be added daily.
