EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:2 i.e. seeding at 4x10,000 cells/cm2 using 0.25% trypsin; 5% CO2; 37C. Limited lifespan; up to 35 passages possible.

From 6 year old Caucasian male with Fanconi's anaemia which is associated with high incidence of leukaemia. Chromosomes show a tendency to spontaneous abnormalities including breaks and complex rearrangement. Have a higher proportion of transformed colonies with SV40 compared with normal diploid cells.
