RPMI 1640 + 2mM Glutamine + 0.05mM 2-Mercaptoethanol (2ME) + 10% Fetal Bovine Serum (FBS). Maintain cultures between 2-4 x 100,000 cells/ml; 5% CO2; 37C.
Karyotype: Near-tetraploid (8q+)
This IgA-producing cell line has been established from a malignant effusion in a 62-year-old Caucasian woman with myeloma. They are positive for plasma cell antigen 1 (PCA-1), transferrin receptor and CD38 but negative for early B cell developmental markers. HLA DR and CALLA. NCI-H929 is negative for Epstein-Barr virus nuclear antigen. A rearrangement of the c-myc proto-oncogene has been observed as well as the expression of c-myc RNA and an activated ras allele.
