RPMI 1640 + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:8 i.e. seeding at 1x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
Passages: 14; The cell line OE50, also known as JROECL50, was established in 1994 from the adenocarcinoma of lower oesophagus of a 71 year old female patient. The tumour was identified as pathological stage IIA (UICC) and showed moderate to poor differentiation. OE50 express HLA-A,-B and -C (MHC class I) constitutively. ICAM-1 expression can be induced by interferon-gamma treatment. HLA-DR (MHC class II) is only expressed in a sub-population after cytokine addition. The cell line is nearly diploid with frequent loss of chromosome 3,5 and 17. Expression of epithelial cytokeratins has been reported. OE50 cells are tumourigenic in nude mice. OE47 and OE50 have been demonstrated, by DNA fingerprint analysis at the ECACC, to represent cell lines from the same individual. However, these cell lines have different morphology and differentiation patterns. The original tumour of origin was a carcinoma of mixed cell type: exhibiting a focal glandular pattern consistent with differentiation towards adenocarcinoma. OE50 is more pleomorphic than OE47 which does not stain by D/PAS (consistent with its squamous origin). OE50 is D/PAS positive. The two cell lines probably represent independent sub-lines of a mixed tumour which show karyological differences (Dr. A. Morris personal communication).
