DMEM:Ham's F12 (1:1) + 2mM Glutamine + 5�g/ml Insulin + 10ng/ml EGF + 0.1�g/ml Choleratoxin + 0.5% Dimethyl Sulphoxide (DMSO) + 15% Fetal Bovine Serum (FBS) (Heat Inactivated). Split confluent cultures 1:4 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
Passages: +5; The RCE cell line, also known as K'RCE, was established after infection of primary cultured rabbit (New Zealand albino) corneal epithelial cells with a recombinant SV40 vector. The cells exhibit a cobblestone-like appearance and stratification, including desmosome formation and microvilli development. Expression of keratin AE1 was always detectable whereas AE5 was expressed only at confluency.
