McCoy's 5a + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:4 to 1:8 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C.
Passages: +4; A bone marrow-derived stromal cell line established by A Pessina in 1987 from a normal female 5 month old mouse. A bone marrow culture was treated with the supernatant from Y1 cells which actively produce viral C-particles (MuLV). This fibroblast-like cell line produces macrophage-colony stimulating factor (M-MCF) and exhibits a high sensitivity to fibroblast growth factor (FGF) in clonogenic assays. The cells produce sarcomas in syngenic mice and have a reported doubling time of 14.7 hours.
