DMEM + 2mM Glutamine + 5% Fetal Bovine Serum (FBS). Split confluent cultures 1:6 to 1:8 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25% trypsin; 5% CO2; 37C. Care is required during subculture as cells have a tendency to easily detach from the tissue culture flask.
Passages: 7; The cells do not produce virus particles. In serum-free media the cells release a specific growth factor. This cell line is of Swiss 3T3 origin, transformed by the Moloney murine sarcoma virus. It is a non-producer of the virus particles. In serum-free media the line will release a growth factor which can be used to stimulate quiescent cells to grow in monolayer as well as in soft agar.
