Western blot using CPTC-MPO-2 as primary antibody against PBMC (lane 2), HeLa (lane 3), Jurkat (lane 4), A549 (lane 5), MCF7 (lane 6), and NCI-H226 (lane 7) whole cell lysates. Expected molecular weight - 83.9 kDa, 73.9 kDa, and 87.2 kDa. Molecular weight standards are also included (lane 1).
Western Blot - Tissue or Cell Lysate
Result: Presumed Positive (with additional bands)
Western blot using CPTC-MPO-2 as primary antibody against human lung (2), spleen (3), endometrium (4), breast (5), and ovary (6) tissue lysates. The expected molecular weight is 83.9 kDa, 73.9 kDa, and 87.2 kDa. Lung, spleen, and endometrium are presumed positive (MPO heavy chain). Breast and ovary are negative. Vinculin was used as a loading control. The immunoblot was conducted under reducing conditions.

Function:
Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of neutrophils.MPO (Myeloperoxidase) is a Protein Coding gene. Diseases associated with MPO include Myeloperoxidase Deficiency and Alzheimer Disease. Among its related pathways are Transcriptional misregulation in cancer and Folate Metabolism. Gene Ontology (GO) annotations related to this gene include chromatin binding and heme binding. An important paralog of this gene is EPX.Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity.
