International Journal of Impotence Research, 2012

Sustained hyperglycemia is considered as a major cause of sexual and erectile dysfunction in huma... more Sustained hyperglycemia is considered as a major cause of sexual and erectile dysfunction in human population. Curculigo orchioides (CO) is considered as a sexual tonic in Ayurvedic system of medicine with potent antioxidant and adaptogenic properties. The aqueous extract of the herb was evaluated for its effectiveness against streptozotocin-induced hyperglycemic stress and subsequent sexual dysfunction due to hyperglycemia in male rats. Six groups with eight male rats in each group were used for this study and the study was carried out for 28 days. The body and organ weights of the animals were recorded. Behavioral analysis of rats was undertaken to observe the effect on mount, ejaculation and intromission (latencies and frequencies) and hesitation time. Blood glucose and serum testosterone levels were determined 28 days past treatment with CO at 100 and 200 mg kg(-1) doses. Glibenclamide and sildenafil citrate were used as positive controls. This deleterious effect of sustained hyperglycemia and associated stress was prominently ameliorated in animals treated with aqueous extract of CO. CO treatment was helpful in ameliorating the damage caused by sustained hyperglycemia evidenced in the principle parameters viz. male sexual behavior, sperm count, penile erection index and seminal fructose content Antioxidant and anabolic activities of the extract under investigation could be a major attribute in preserving the sexual functions in hyperglycemic male rats. The study validates the use of CO in traditional medicine for curing diabetes-induced sexual dysfunction and compromised sexual potency.

The developed method is also utilized to determine purity and quality of the market formulation drug by dissolution studies. The mobile phase consisted of methanol and water (89:11 v/v) showed retention time of 2.99 min with a flow rate of 1 mL/min at detection wavelength 266 nm. Linear regression analysis data for the calibration plot showed a good linear relationship between response and concentration in the range 25–350 µg mL−1; the regression coefficient was 0.9995 and the linear regression equation was y = 19.75x + 57.73. The detection (LOD) and quantification (LOQ) limits were 2.92 and 8.87 µg mL−1, respectively. The method was validated for accuracy, precision, specificity, robustness, and detection and quantification limits, in accordance with ICH guidelines. Statistical analysis proved the method was precise, reproducible, selective, specific, and accurate for analysis of Zidovudine. The wide linearity range, sensitivity, accuracy, short retention time, and simple mobile phase imply the method is suitable for routine quantification of Zidovudine with high precision and accuracy.

The changes in sexual behaviour, reproductive organ weights, histology of testis and epididymis, epididymal sperm density, and androgenic hormone levels were evaluated. The sexual behaviour parameters studied such as mount frequency, intromission frequency, mount latency, intromission latency were significantly affected. Increase in body weight as well as weight of testis, prostate, seminal vesicle, and epididymis was noticed. Transverse sections of testis exhibited increased spermatogenesis and a significant increase in sperm count in epididymis. The fructose content of seminal vesicle was also increased. The extract treatment also brought a significant increase in serum testosterone and luteinizing hormone levels. The studies clearly reflect androgenic activity of the extract and its effects on hypothalamic pituitary gonadal axis.

The herbs have been traditionally used as Vajikaran Rasayana herbs because of their putative positive influence on sexual performance in humans. Lyophilized aqueous extracts obtained from the roots of A. racemosus, C. borivilianum, and rhizomes of C. orchioides were studied for sexual behavior effects in male albino rats and compared with untreated control group animals (total N = 60). The rats were evaluated for effect of treatments on anabolic effect. Seven measures of sexual behavior were evaluated. Administration of 200 mg/kg body weight of the aqueous extracts had pronounced anabolic effect in treated animals as evidenced by weight gains in the body and reproductive organs. There was a significant variation in the sexual behavior of animals as reflected by reduction of mount latency, ejaculation latency, post ejaculatory latency, intromission latency, and an increase of mount frequency. Penile erection (indicated by Penile Erection Index) was also considerably enhanced. Reduced hesitation time (an indicator of attraction towards female in treated rats) also indicated an improvement in sexual behavior of extract treated animals. The observed effects appear to be attributable to the testosterone-like effects of the extracts. Nitric oxide based intervention may also be involved as observable from the improved penile erection. The present results, therefore, support the folklore claim for the usefulness of these herbs and provide a scientific basis for their purported traditional usage.

The ethanolic extract of seeds of A. longifolia was administered to groups of rats in 100, 150 and 200 mg kg⁻¹ doses for a period of 28 days, and the action compared with control rats. The changes in body and organ weight, sexual behaviour, histo-architecture and fructose levels of seminal vesicles were observed. The sexual behaviour was assessed by determining parameters such as mount frequency (MF), intromission latency, mount latency (ML) and post-ejaculatory latency. The ethanolic extract exhibited pronounced anabolic effects in treated animals, as evidenced by gains in the body and reproductive organ weights. Increased spermatogenesis due to treatment with extracts was also witnessed in transverse section. The treatment further markedly affected sexual behaviour of the animals, as reflected by the reduction of ML, increase in MF and enhanced attractability towards females. A significant increase in the sperm count as well as fructose levels of seminal vesicles was noted.

Androgenetic alopecia (AGA) is the most common form of alopecia, which affects millions of men and women worldwide, and is an androgen driven disorder. Here, the Citrullus colocynthis Schrad fruit is evaluated for hair growth activity in androgen-induced alopecia. Petroleum ether extract of C. colocynthis was applied topically for its hair growth-promoting activity. Alopecia was induced in albino mice by testosterone administration intramuscularly for 21 days. Its inhibition by simultaneous administration of extract was evaluated using follicular density, anagen/telogen (A/T) ratio and microscopic observation of skin sections. Finasteride (5α-reductase inhibitor) solution was applied topically and served as positive control. Petroleum ether extract of C. colocynthis exhibited promising hair growth-promoting activity, as reflected from follicular density, A/T ratio and skin sections. The treatment was also successful in bringing a greater number of hair follicles in anagenic phase than the standard finasteride. The result of treatment with 2 and 5% petroleum ether extracts were comparable to the positive control finasteride. The petroleum ether extract of C. colocynthis and its isolate is useful in the treatment of androgen-induced alopecia.

They are believed to have aphrodisiac action. The present investigation was undertaken to evaluate their effects on sexual behavior in male rats. Thirty-two male Wistar rats were divided into control group, testosterone group, low-dose (50 mg/kg) petroleum ether extract (PEE) group and high-dose (100 mg/kg) PEE group. PEE obtained from the roots of Anacyclus pyrethrum was administered orally to albino rats once daily, and 0.5 mg/kg (body weight) of testosterone was given intramuscularly twice weekly and served as positive control. The course of treatment was 28 days. The effects of PEE and testosterone on changes in body and accessory sexual organ weights, sexual behavior, penile erection and sexual performance were studied before treatment, after 15 and 28 days of treatment and 7 and 15 days after treatment. After 28 days of treatment, PEE and testosterone had a marked influence on body and accessory sexual organ weights as compared with arachis oil. The treated male rats were more receptive and oriented towards female rats and increased precopulatory activities like licking and sniffing of female anogenital were observed. The penile erection index was significantly increased with reduction in mount latency and intromission latency period. There were four-fold increase in mount and three-fold increase in intromission frequency in treated rats reflecting improved sexual performance. The behavioral and sexual parameters were also observed after a lapse of 7 and 15 days of discontinuance of drug treatment. Unlike testosterone, the PEE of Anacyclus pyrethrum shows efficacy in rats tested after the lapse of 7 and 15 days of discontinuation of treatment. This suggests that the drug has prolonged effect and capacitate the treated rats for improved sexual potential.
