Tumor Necrosis Factor-α Regulates Distinct Molecular Pathways and Gene Networks in Cultured Skeletal Muscle Cells (original) (raw)
Figure 5
Western blot analyses of genes differentially expressed by TNF-α in myotubes.
C2C12 myotubes were treated with 10 ng/ml of TNF for 18h followed by isolation of total protein and Western blotting. Equal amounts of proteins were loaded on 10% SDS-PAGE gel. A). Representative immunoblots presented here show that TNF-α increases the protein levels of NFKB1, NFKB2, MMP-9, and IκBα in myotubes. B). Densitometry analyses of bands from NFKB1, NFKB2, MMP-9, and IκBα immunoblots. C). Protein levels of Timp-2 and Notch1 were found to be reduced upon treatment with TNF-α. D). Densitometry analyses of bands from TIMP-2 and Notch1 immunoblots. *p<0.05, values significantly different from corresponding control myoblasts incubated without TNF-α (n = 4 in each group).
