It is well known that dopamine imbalances are associated with many psychiatric disorders and that... more It is well known that dopamine imbalances are associated with many psychiatric disorders and that the dopaminergic receptor D2 is the main target of antipsychotics. Recently it was shown that levels of two proteins implicated in dopaminergic signaling, Neuronal calcium sensor-1 (NCS-1) and DARPP-32, are altered in the prefrontal cortex (PFC) of both schizophrenic and bipolar disorder patients. NCS-1, which inhibits D2 internalization, is upregulated in the PFC of both patients. DARPP-32, which is a downstream effector of dopamine signaling, integrates the pathways of several neurotransmitters and is downregulated in the PFC of both patients. Here, we used PC12 cells stably overexpressing NCS-1 (PC12-NCS-1 cells) to address the function of this protein in DARPP-32 signaling pathway in vitro. PC12-NCS-1 cells displayed downregulation of the cAMP/PKA pathway, with decreased levels of cAMP and phosphorylation of CREB at Ser133. We also observed decreased levels of total and phosphorylated DARPP-32 at Thr34. However, these cells did not show alterations in the levels of D2 and phosphorylation of DARPP-32 at Thr75. These results indicate that NCS-1 modulates PKA/cAMP signaling pathway. Identification of the cellular mechanisms linking NCS-1 and DARPP-32 may help in the understanding the signaling machinery with potential to be turned into targets for the treatment of schizophrenia and other debilitating psychiatric disorders.

La thèse de Pierre Janet (1889), souvent présentée de manière restrictive sous l’angle de la psyc... more La thèse de Pierre Janet (1889), souvent présentée de manière restrictive sous l’angle de la psychopathologie, est ici replacée dans son contexte et dans sa portée problématique. Pierre Janet (1859–1947) impute les phénomènes d’automatisme à la faiblesse de la synthèse psychologique, qui entraîne une désagrégation mentale et un rétrécissement du champ de la conscience. Les actes automatiques résultent de montages psychiques anciens qui se conservent au niveau subconscient et se manifestent en diverses occasions: somnambulisme, distraction, suggestibilité, catalepsie, etc. Au lieu d’un mécanisme caché, c’est un fonctionnement dynamique que dévoile Janet, évaluant sur le terrain de la psychologie expérimentale la pertinence d’un thème kantien, la fonction unifiante du je, dont Émile Boutroux avait tiré une théorie de la puissance qualitative et créatrice de l’esprit en partie reprise par Bergson. La psychologie dynamique du premier Janet a servi de référence, au XXe siècle, à un courant de psychiatrie française marqué par Jean Delay, Henri Ey et Henri Baruk, dans un contexte intellectuel de résistance à la psychanalyse. On montre ici les raisons de cette continuité, en précisant la signification du subconscient et en interrogeant les conceptions épistémologiques du jeune Janet. Pierre Janet’s doctoral dissertation (1889), often narrowly discussed from a uniquely psychopathological perspective, is presented here within its context and intellectual framework. For Pierre Janet (1859–1947) automatism phenomena are due to a weakness in psychological synthesis, which leads to mental deterioration and a narrowing of the field of consciousness. Uncontrolled spontaneous acts pertain to previous psychic constructs that are stored on a subconscious level and expressed in various circumstances: somnambulism, distraction, suggestibility, catalepsy, etc. Instead of a hidden mechanism, Pierre Janet unfolds a dynamic function, assessing a Kantian theme-the unifying function of “I”—using experimental psychology, from which Emile Boutroux had drawn a theory of the qualitative and creative power of the mind, which partly inspired Bergson. The dynamic psychology of Pierre Janet served as a reference during the XXth century for a French psychiatric movement led by Jean Delay, Henri Ey and Henri Baruk in the context of resistance to psychoanalysis. Here, we trace the reasoning behind this continuum, by detailing the meaning of the subconscious and questioning the epistemological notions of the young Pierre Janet.
