Abstract: Most of the women take either prescribed or over-the-counter medication during pregnanc... more Abstract: Most of the women take either prescribed or over-the-counter medication during pregnancy. Pregnant women often make independent decisions about their treatment and require careful counseling that allows them to balance the risk of taking a drug against the risk of not taking a drug and leaving a medical condition untreated. Most drugs are safe in pregnancy but some have unproven safety and may adversely affect the growing fetus. The safety profile of some of the medication may change according to the gestational age of the fetus. An estimated 10 % or more of the birth defects results from maternal drug exposure. The development of knowledge in understanding the use of drugs during pregnancy has been in stalemate in comparison to other areas of therapeutics, mainly due to difficulties in testing new products in pregnant women and paucity of good quality research.

A new, simple, precise, rapid and selective high-performance thin-layer chromatographic (HPTLC) m... more A new, simple, precise, rapid and selective high-performance thin-layer chromatographic (HPTLC) method for the simultaneous quantification of Metronidazole (MTZ) and Miconazole nitrate (MCZ) in gel has been developed. It was performed on silica gel 60 GF254 Thin Layer Chromatographic plates using mobile phase comprising of Toluene: Chloroform: Methanol (3.0:2.0:0.6 v/v) and the detection was carried out at 240 nm using densitometer. The retention factors of MTZ and MCZ were 0.34 and 0.55 respectively. Calibration curves were linear in the range of 300-700 ng/spot of MTZ and 600-1400 ng/spot of MCZ both by height and by area. The percent recovery of the drugs from gel carried out by standard addition method was found to be 100.13+/-1.59 (by height) and 98.92+/-0.76 (by area) for MTZ and 99.49+/-1.58 (by height) and 99.63+/-1.46 (by area) for MCZ indicative of accuracy and precision of simultaneous determination of MTZ and MCZ nitrate.

A rapid and sensitive reversed-phase high-performance liquid chromatographic method is developed ... more A rapid and sensitive reversed-phase high-performance liquid chromatographic method is developed for simultaneous estimation of fluconazole, an orally active triazole anti-fungal agent, and tinidazole, which belongs to the group of 5-nitroimidazoles in combined dose tablet. Chromatographic separation was on an ODS Hypersil C(18) column using 0.05 M potassium dihydrogen phosphate buffer (pH 3.25, adjusted with orthophosphoric acid) and acetonitrile (82:18, v/v) as the mobile phase at a flow rate of 1.5 mL/min with detection at 210 nm. The asymmetry factors are 1.36 +/- 0.04 for fluconazole and 1.26 +/- 0.07 for tinidazole with a total run time of less than 7 min. The calibration curves were linear in the range 6-14 microg/mL for fluconazole and 80-190 microg/mL for tinidazole. The method was validated with respect to linearity, precision, accuracy, and specificity. The mean recovery for fluconazole and tinidazole is 99.65 +/- 0.84 and 99.34 +/- 0.70, respectively. Guanine rich sequence have the ability to fold into stable 4 stranded structures called G-quadrup... more Guanine rich sequence have the ability to fold into stable 4 stranded structures called G-quadruplex under physiological concentrations of Na+ or K+. G-quadruplexes are found in telomeres, being stable structures under the control of telomerase binding proteins. They are also identified throughout the genome and are enriched in promoter regions of protein coding genes, upstream and downstream of the transcription initiation sites. A number of these promoter quadruplexes have been investigated for several proto-oncogenes. The formation of these quadruplexes can lead to chemical intervention of gene expression using a G-quadruplex binding ligand. We review location, configuration, and stabilization of these quadruplexes in some of the important promoters with regards to their potential as anticancer target.

The biological significance of G-quadruplex has been recognized by numerous research efforts. Wit... more The biological significance of G-quadruplex has been recognized by numerous research efforts. With the advancement of x-ray crystallography, CD and NMR spectroscopy studies, the structure and topology of G-quadruplex has become clear than before. As there are many G-rich regions along chromosomes and some of these are associated with the promoter of oncogenes. This provided us an excellent opportunity to perform structure based drug design to target telomeric and oncogene promoter sequences using series of meso- substituted porphyrins. In the proposed research work ligand based docking was carried out on Schrödinger 2011 suite using maestro elements. The docking was carried out on two oncogene promoter sequences with PDB id 2A5R and 2HRI. Among the number of inhibitors four legends shows promising binding ability in comparison with known inhibitor of G-quadruplex TMPyP4.
