Tracking antigen specific T cells with major histocompatibility complex (MHC) tetramers has provi... more Tracking antigen specific T cells with major histocompatibility complex (MHC) tetramers has provided us with insights into the dynamics of the adaptive immune system and holds great promise to aid in patient management and drug and vaccine development. Progress has been made primarily using MHC class I tetramers to monitor CD8+ T cells, whereas corresponding efforts to stain CD4+ T cells with class II tetramers have not been as successful. Two major reasons have been proposed for this lack of progress: (1) The frequency of antigen-specific CD4+ T cells is lower than the frequency of CD8+ T cells and (2) some, but not all, antigen- specific CD4+ T cells can bind tetramer because of low functional avidity. In this study, we asked if CD4+ T cells specific for common human viruses (e.g., influenza and Epstein-Barr) can be detected in healthy individuals previously exposed to them. We were able to clearly detect specific CD4+ T cells in all donors after in vitro expansion of peripheral blood mononuclear cells. Furthermore, we observe a clear separation of tetramer negative and tetramer positive CD4+ T cells in most samples similar to patterns commonly seen with class I tetramers. The data indicate that MHC class II tetramers can be used reliably for the identification of CD4+ T cells specific for ubiquitous infectious agents in normal donors.

The refractive index, absorption coefficient, complex real and imaginary permittivity, and loss t... more The refractive index, absorption coefficient, complex real and imaginary permittivity, and loss tangent of normal (non-tumorous) and tumorous breast tissues have been obtained from 0.2 to 0.9 THz. Dispersive Fourier transform spectroscopy was utilized to acquire the broadband millimeter and submillimeter wave dielectric properties and unique resonance signatures of breast tissues for the first time. Results indicate a significant difference in the electrical characteristics of tumorous and non-tumorous breast tissues.

We theoretically consider the effects of having unintentional charged impurities in laterally cou... more We theoretically consider the effects of having unintentional charged impurities in laterally coupled two-dimensional double (GaAs) quantum dot systems, where each dot contains one or two electrons and a single charged impurity. Using molecular orbital and configuration interaction method, we calculate the effect of the impurity on the 2-electron energy spectrum of each individual dot as well as on the spectrum of the coupled-double-dot 2-electron system. We find that the singlet-triplet exchange splitting between the two lowest energy states, both for the individual dots and the coupled dot system, depends sensitively on the location of the impurity and its coupling strength (i.e. the effective charge). A strong electron-impurity coupling breaks down equality of the two doublyoccupied singlets in the left and the right dot leading to a mixing between different spin singlets. As a result, the maximally entangled qubit states are no longer fully obtained in zero magnetic field case. Moreover, a repulsive impurity results in a triplet-singlet transition as the impurity effective charge increases or/and the impurity position changes. We comment on the impurity effect in spin qubit operations in the double dot system based on our numerical results.

More research is needed to understand the mechanisms underlying these relations and their potential relevance outside of the residential setting. This study integrates ecological momentary assessment (EMA) and geo-location tracking to explore real-time associations between exposure to TROs and smoking urges among 47 economically disadvantaged smokers in a cessation trial (59.6% female; 36.2% White). EMA data were collected for 1 week postquit via smartphone, which recorded smoking urge strength ≤ 4 random times daily along with real-time participant location data. For each assessment, the participants' proximity to the closest TRO and the density of TROs surrounding the participant were calculated. The broadband dielectric properties of tumorous and non-tumorous tissues were analyzed at the mic... more The broadband dielectric properties of tumorous and non-tumorous tissues were analyzed at the microwave, millimeter wave, and terahertz ranges. In-vitro measurements were obtained using the coaxial probe and dispersive Fourier transform spectroscopy. Both techniques were modified to yield the complex real and imaginary permittivity, absorption coefficient, and refractive index. The results reveal the characteristic signatures of breast tissues and display a significant difference in the electromagnetic response of tumorous and non-tumorous tissues.

Membrane-bound formate dehydrogenase (FDH) was purified to homogeneity from a facultative anaerob... more Membrane-bound formate dehydrogenase (FDH) was purified to homogeneity from a facultative anaerobic bacterium Citrobacter sp. S-77. The FDH from Citrobacter sp. S-77 (FDHS77) was a monomer with molecular mass of approximately 150 kDa. On SDS-PAGE, the purified FDHS77 showed as three different protein bands with molecular mass of approximately 95, 87, and 32 kDa, respectively. Based on the N-terminal amino acid sequence analysis, the sequence alignments observed for the 87 kDa protein band were identical to that of the large subunit of 95 kDa, indicating that the purified FDHS77 consisted of two subunits; a 95 kDa large subunit and a 32 kDa small subunit. The purified FDHS77 in this purification did not contain a heme b subunit, but the FDHS77 showed significant activity for formate oxidation, determined by the Vmax of 30.4 U/mg using benzyl viologen as an electron acceptor. The EPR and ICP-MS spectra indicate that the FDHS77 is a molybdenum-containing enzyme, displaying a remarkable O2-stability along with thermostability and pH resistance. This is the first report of the purification and characterization of a FDH from Citrobacter species.

For instance, prior research shows that both the Put-Call Ratio (P/C) and the Option-to-Stock Volume Ratio (O/S) predict negative future stock returns. In this paper, we compare the level of information contained in these two commonly used option volume ratios. Our comparison of the return predictability contained in these ratios yields some new results. First, we find that P/C ratios contain more predictability about future stock returns at the daily level than O/S ratios. Second, in contrast to our first set of results, O/S ratios contain more predictability about future returns at the weekly and monthly levels than P/C ratios. In fact, our tests show that while P/C ratios contain predictability about future daily returns and, to some extent, future weekly returns, the return predictability in P/C ratios is fleeting. O/S ratios, on the other hand, significantly predict negative returns at both the weekly and monthly levels, respectively.
