For safe consumption of raw milk products, ripening is a critical period for reducing populations of pathogenic and spoilage microorganism and, then, the knowledge on inactivation kinetics of microorganisms is necessary to guarantee food safety and quality. In the present work, inactivation kinetics of foodborne bacterial pathogens required by Brazilian regulations in serrano artisanal cheeses were statistically evaluated during ripening. The Salmonella spp., Listeria monocytogenes, coagulase-positive strain of Staphylococcus aureus and fecal coliforms were analyzed for up to 60 days. Experimental data were fitted using first-order model, Weibull and log-linear + shoulder models. Neste relato descreve-se um caso de tétano em um canino, com ênfase em seus aspectos clínicos e terapêuticos, além da sua recuperação clínica. Foi atendido no Hospital Veterinário da Universidade de Passo Fundo, um canino, fêmea, da raça Pitbull, com cinco meses, em status epilepticus. Após a estabilização da paciente, constataram-se animal em posição de cavalete, com paralisia espástica generalizada, risus sardonicus, trismo, cauda e orelhas eretas e dificuldade em expandir o tórax. O diagnóstico presuntivo de tétano foi firmado a partir da anamnese, sinais clínicos e achados laboratoriais. The disease can occur early in newborn piglets (EITP) or late in 2- to 3-week old piglets (LITP). In this study, we analysed the clinical, pathological, and hematological aspects of 391 ITP cases (312 with EITP and 79 with LITP). In LIPT cases, morbidity and mortality rates were higher, with rates of 60% (morbidity) and 53% (mortality). The main clinicopathological findings in ITP cases were different patterns of hemorrhages organs and tissues. In EITP, clinical signs were characterized by extensive subcutaneous hemorrhages and death occurred within a few days; however, in LITP, often sudden death occurred. In macroscopic analysis, hemorrhagic diathesis was observed in all affected animals. In EITP, the most severe hemorrhagic lesions were integumentary, mainly in the dermis and epidermis. Nos animais, especialmente equinos, a doença se apresenta frequentemente de forma assintomática, entretanto, são considerados uma fonte de infecção para humanos e outras espécies. O presente estudo objetivou avaliar padrões do metabolismo oxidativo eritrocitário (glutationa e malondialdeído), fragilidade osmótica eritrocitária e hematobioquímicos em equinos com leptospirose subclínica para auxiliar no diagnóstico e prognóstico da infecção. Para tal foram avaliados 207 equinos, positivos (n=94) e negativos (n= 113) para leptospirose. The purpose of this research was to study the management, production of animal health services re... more The purpose of this research was to study the management, production of animal health services residues (AHSR) in swine farms in the state of Santa Catarina, Brazil and identify, quantify possible agents bacterial indicators of environmental contamination and to determine the resistance of bacteria more commonly isolated to the main antimicrobial used in swine breeding. In order to realize this work 131 swine production unities were studied, divided in 78 piglets production unities (PPU), 18 growing unities (GU) and 38 finishing unities (FU). In 54.96% (72/ 131) of the swine farms they accomplished the separation among the different classes of AHSR . Comparing the AHSR stored, residue group B and E and AHSR burning places, the bacterial counting was superior in AHSR (P<0.05). Surra is an infectious disease caused by Trypanosoma evansi, which affects a large number of dome... more Surra is an infectious disease caused by Trypanosoma evansi, which affects a large number of domestic and wild animal species. Infection control is based on rapid diagnosis followed by treatment of sick animals. This study aimed to evaluate a buffered T. evansi antigen and rapid serum agglutination test (BA/Te) for the detection of anti-T. evansi antibodies in serum samples of horses. For this purpose, 445 serum samples from horses were evaluated and the results compared with the diagnosis by CATT/T. evansi. Our data show a sensitivity of 92%, specificity of 91% and a degree of agreement kappa (κ) of 0.82 (95% CI: 0.771–0.877, P < 0.01) between BA/Te and CATT/T. evansi. Antigen specificity was also evaluated against reactive serum for other infectious agents circulating in equine herds. In conclusion, our findings show that BA/Te has the potential to be a practical and quick screening method for the detection of anti-T. evansi antibodies in horses.

This study reports the first autochthonous "surra" outbreak in horses in the State of S... more This study reports the first autochthonous "surra" outbreak in horses in the State of Santa Catarina, southern Brazil. Six horses with clinical suspicion of trypanosomosis had the natural infection by T. evansi confirmed by PCR and rapid serum agglutination test. Clinical, parasitological, and hematology evaluations were performed at initial observation (T0) and 90 days after (T1). At T0, all animals that tested positive for T. evansi in PCR presented with severe clinical signs and out of normal range hematological hematological (hematocrit, leukocytes, platelets, hemoglobin) and serum biochemical parameters (alanine aminotransferase (ALT), creatinine phosphokinase (CPK), lactate dehydrogenase (LDH), creatinine, bilirubin, and glucose). At T1, the progressive clinical recovery of animals, normalization of hemato-biochemical parameters, and negative PCR results for T. evansi were observed. It was not possible to identify the vector/mechanism of transmission through which animals were infected; therefore, the implementation of surveillance and control measures is essential to prevent the spread of this disease in horse herds, as well as to other animal species.

This study aimed to evaluate the seropositivity of unvaccinated horses in the Serra Catarinense region and the possible risk factors associated with the infection. We used 207 horses from 26 properties, with no clinical history of infection and unvaccinated for leptospirosis. We submitted serum samples to the microscopic agglutination serum (SAM) to obtain the main reference serogroups. We considered animals with titre ≥100 as infected. The results of the serology revealed seropositivity in 45.4% (94/207) of the animals studied. The most common causative agents of avian infectious arthritis are the bacterium Mycoplasma synoviae and avian reoviruses (ARVs) (family Reoviridae, genus Orthoreovirus). In this study, we evaluated the occurrence of these two pathogens in arthritis or tenosynovitis lesions of broilers and breeder flocks in southern Brazil using molecular detection. Tissue sections from tibiotarsal joints with visible lesions from 719 broilers and 505 breeders were analysed using pathogen-specific polymerase chain reaction (PCR) assays. Os distúrbios gastrintestinais e toxi-infeciosos causadas por Escherichia coli são uma importante... more Os distúrbios gastrintestinais e toxi-infeciosos causadas por Escherichia coli são uma importante causa de mortalidade, atrasos no crescimento e decréscimo na taxa de conversão alimentar em leitões no período pós-desmame, gerando perdas econômicas severas. Dentre as estratégias de combate a estas formas de apresentação estão à utilização de antimicrobianos, probióticos e aditivos na dieta, como o óxido de zinco. O zinco tem efeito promotor de crescimento, diminui a incidência de diarréia e mortalidade de leitões desmamados. O objetivo da realização desse trabalho foi verificar, in vitro, o papel de diferentes concentrações de zinco na adesão da E. coli aos enterócitos de suínos. O ensaio avaliou a eficácia de quatro suspensões: zero (controle negativo), 1200, 2400 e 3000 ppm de óxido de zinco. O teste de adesão fundamentou-se na exposição de enterócitos suínos, a uma cepa patogênica de E. coli e ao zinco, simulando o que ocorreria in vivo. Nas condições in vitro em que foi desenvolvido o experimento, o zinco não foi capaz de inibir a adesão de E.coli aos enterócitos em nenhuma das concentrações utilizadas.

RESUMO O objetivo deste estudo é determinar os aspectos microbiológicos e físico-químicos da urin... more RESUMO O objetivo deste estudo é determinar os aspectos microbiológicos e físico-químicos da urina de 922 fêmeas suínas com suspeita clínica de infecção urinária (IU) em diversas fases de produção em granjas comerciais no Sul do Brasil e estabelecer o perfil de sensibilidade a antibóticos dos agentes bacterianos mais freqüentemente isolados. Confirmou-se o diagnóstico de infecção urinária em 886 (93,98%) das 922 fêmeas com sinais clínicos de IU. Os agentes bacterianos isolados em cultura pura com maior freqüência foram Escherichia coli (48,13%) e Streptococcus sp (27,58%). As fêmeas apresentaram, com maior freqüência, no exame físico da urina, coloração amarelo-clara (32,3%), aspecto turvo (86,6%) e odor amoniacal (66,04%). Quanto aos valores de pH, as fêmeas com e sem IU apresentaram valores médios de 6,42 e 6,29, respectivamente. Os parâmetros clínicos dos animais e aspectos físico-químicos da urina devem sempre ser levados em consideração na avaliação da ocorrência de IU, no entanto, não devem ser considerados isoladamente como indicadores de IU, uma vez que somente a análise conjunta torna-os representativos e significantes. Os antimicrobianos mais eficientes no controle de IU foram ceftiofur, norfloxacina e enrofloxacina.

In contrast, some reports have suggested rhBMP-2 does not provide advantages over autogenous bone grafting owing to the undesirable postoperative symptoms of this growth factor. Because the undesirable symptoms of rhBMP-2 are usually promoted by inflammation, this study evaluated the in vivo effect of human adipose-derived stem cells (ASCs) incorporated into polylactic co-glycolic acid (PLGA) scaffolds in decreasing the inflammatory response induced by a low dose of rhBMP-2. Materials and Methods: PLGA scaffolds were characterized and loaded with rhBMP-2 1, 2.5, or 5 mg per scaffold (n = 6) and the in vitro released protein amounts were quantified at 7 hours and 1, 7, and 21 days after loading (n = 3). The muscle tissue of 6 beagles received the following treatments: PLGA, PLGA plus rhBMP-2 (2.5 mg), and PLGA plus rhBMP-2 plus ASCs (1 Â 10 6 ASCs). The samples were evaluated 45 days after surgery. Statistical analyses were performed and the P value was set at .05. Results: PLGA plus rhBMP-2 plus ASCs yielded the smallest number of inflammatory foci (P < .001) and giant cells (P < .001) and the largest number of angiogenesis sites (P < .001). Conclusions: Human ASCs administered in vivo into PLGA scaffolds with a low dose of rhBMP-2 decrease tissue inflammation and increase angiogenesis in muscular sites.

Although it has been demonstrated that mucosal immunization using vectors such as simian adenovir... more Although it has been demonstrated that mucosal immunization using vectors such as simian adenovirus (AdC) stimulates robust adaptive immune responses, there remains a paucity of information on the modulation of innate immune responses by such vectors. Using an established murine model of intravaginal immunization (Ivag), we have investigated whether mucosal ␥␦ T cells participate in immune responses induced by AdC vectors. ␥␦ T cell numbers were found to be increased in the vaginal tract. Moreover, ␥␦ T cells isolated from the genital tract showed an activated phenotype and enhanced expression of cytokine gene. Altogether, our results demonstrate that AdC modulates ␥␦ T cell responses and suggest that this cell population may influence immune responses following vaginal immunization.

O presente estudo objetivou avaliar padrões do metabolismo oxidativo eritrocitário (glutationa e malondialdeído), fragilidade osmótica eritrocitária e hematobioquímicos em equinos com leptospirose subclínica para auxiliar no diagnóstico e prognóstico da infecção. Infection control is based on rapid diagnosis followed by treatment of sick animals. This study aimed to evaluate a buffered T. evansi antigen and rapid serum agglutination test (BA/Te) for the detection of anti-T. evansi antibodies in serum samples of horses. For this purpose, 445 serum samples from horses were evaluated and the results compared with the diagnosis by CATT/T. evansi. Our data show a sensitivity of 92%, specificity of 91% and a degree of agreement kappa (κ) of 0.82 (95% CI: 0.771–0.877, P < 0.01) between BA/Te and CATT/T. evansi. Antigen specificity was also evaluated against reactive serum for other infectious agents circulating in equine herds. In conclusion, our findings show that BA/Te has the potential to be a practical and quick screening method for the detection of anti-T. evansi antibodies in horses.

This study reports the first autochthonous "surra" outbreak in horses in the State of S... more This study reports the first autochthonous "surra" outbreak in horses in the State of Santa Catarina, southern Brazil. Six horses with clinical suspicion of trypanosomosis had the natural infection by T. evansi confirmed by PCR and rapid serum agglutination test. Clinical, parasitological, and hematology evaluations were performed at initial observation (T0) and 90 days after (T1). At T0, all animals that tested positive for T. evansi in PCR presented with severe clinical signs and out of normal range hematological hematological (hematocrit, leukocytes, platelets, hemoglobin) and serum biochemical parameters (alanine aminotransferase (ALT), creatinine phosphokinase (CPK), lactate dehydrogenase (LDH), creatinine, bilirubin, and glucose). At T1, the progressive clinical recovery of animals, normalization of hemato-biochemical parameters, and negative PCR results for T. evansi were observed. It was not possible to identify the vector/mechanism of transmission through which animals were infected; therefore, the implementation of surveillance and control measures is essential to prevent the spread of this disease in horse herds, as well as to other animal species.

This study aimed to evaluate the seropositivity of unvaccinated horses in the Serra Catarinense region and the possible risk factors associated with the infection. We used 207 horses from 26 properties, with no clinical history of infection and unvaccinated for leptospirosis. We submitted serum samples to the microscopic agglutination serum (SAM) to obtain the main reference serogroups. We considered animals with titre ≥100 as infected. The most common causative agents of avian infectious arthritis are the bacterium Mycoplasma synoviae and avian reoviruses (ARVs) (family Reoviridae, genus Orthoreovirus). In this study, we evaluated the occurrence of these two pathogens in arthritis or tenosynovitis lesions of broilers and breeder flocks in southern Brazil using molecular detection. Tissue sections from tibiotarsal joints with visible lesions from 719 broilers and 505 breeders were analysed using pathogen-specific polymerase chain reaction (PCR) assays. Os distúrbios gastrintestinais e toxi-infeciosos causadas por Escherichia coli são uma importante... more Os distúrbios gastrintestinais e toxi-infeciosos causadas por Escherichia coli são uma importante causa de mortalidade, atrasos no crescimento e decréscimo na taxa de conversão alimentar em leitões no período pós-desmame, gerando perdas econômicas severas. Dentre as estratégias de combate a estas formas de apresentação estão à utilização de antimicrobianos, probióticos e aditivos na dieta, como o óxido de zinco. O zinco tem efeito promotor de crescimento, diminui a incidência de diarréia e mortalidade de leitões desmamados. O objetivo da realização desse trabalho foi verificar, in vitro, o papel de diferentes concentrações de zinco na adesão da E. coli aos enterócitos de suínos. O ensaio avaliou a eficácia de quatro suspensões: zero (controle negativo), 1200, 2400 e 3000 ppm de óxido de zinco. O teste de adesão fundamentou-se na exposição de enterócitos suínos, a uma cepa patogênica de E. coli e ao zinco, simulando o que ocorreria in vivo. Nas condições in vitro em que foi desenvolvido o experimento, o zinco não foi capaz de inibir a adesão de E.coli aos enterócitos em nenhuma das concentrações utilizadas.

RESUMO O objetivo deste estudo é determinar os aspectos microbiológicos e físico-químicos da urin... more RESUMO O objetivo deste estudo é determinar os aspectos microbiológicos e físico-químicos da urina de 922 fêmeas suínas com suspeita clínica de infecção urinária (IU) em diversas fases de produção em granjas comerciais no Sul do Brasil e estabelecer o perfil de sensibilidade a antibóticos dos agentes bacterianos mais freqüentemente isolados. Confirmou-se o diagnóstico de infecção urinária em 886 (93,98%) das 922 fêmeas com sinais clínicos de IU. Os agentes bacterianos isolados em cultura pura com maior freqüência foram Escherichia coli (48,13%) e Streptococcus sp (27,58%). As fêmeas apresentaram, com maior freqüência, no exame físico da urina, coloração amarelo-clara (32,3%), aspecto turvo (86,6%) e odor amoniacal (66,04%). Quanto aos valores de pH, as fêmeas com e sem IU apresentaram valores médios de 6,42 e 6,29, respectivamente. Os parâmetros clínicos dos animais e aspectos físico-químicos da urina devem sempre ser levados em consideração na avaliação da ocorrência de IU, no entanto, não devem ser considerados isoladamente como indicadores de IU, uma vez que somente a análise conjunta torna-os representativos e significantes. Os antimicrobianos mais eficientes no controle de IU foram ceftiofur, norfloxacina e enrofloxacina.

In contrast, some reports have suggested rhBMP-2 does not provide advantages over autogenous bone grafting owing to the undesirable postoperative symptoms of this growth factor. Because the undesirable symptoms of rhBMP-2 are usually promoted by inflammation, this study evaluated the in vivo effect of human adipose-derived stem cells (ASCs) incorporated into polylactic co-glycolic acid (PLGA) scaffolds in decreasing the inflammatory response induced by a low dose of rhBMP-2. Materials and Methods: PLGA scaffolds were characterized and loaded with rhBMP-2 1, 2.5, or 5 mg per scaffold (n = 6) and the in vitro released protein amounts were quantified at 7 hours and 1, 7, and 21 days after loading (n = 3). The muscle tissue of 6 beagles received the following treatments: PLGA, PLGA plus rhBMP-2 (2.5 mg), and PLGA plus rhBMP-2 plus ASCs (1 Â 10 6 ASCs). The samples were evaluated 45 days after surgery. Statistical analyses were performed and the P value was set at .05. Results: PLGA plus rhBMP-2 plus ASCs yielded the smallest number of inflammatory foci (P < .001) and giant cells (P < .001) and the largest number of angiogenesis sites (P < .001). Conclusions: Human ASCs administered in vivo into PLGA scaffolds with a low dose of rhBMP-2 decrease tissue inflammation and increase angiogenesis in muscular sites.

Although it has been demonstrated that mucosal immunization using vectors such as simian adenovir... more Although it has been demonstrated that mucosal immunization using vectors such as simian adenovirus (AdC) stimulates robust adaptive immune responses, there remains a paucity of information on the modulation of innate immune responses by such vectors. Using an established murine model of intravaginal immunization (Ivag), we have investigated whether mucosal ␥␦ T cells participate in immune responses induced by AdC vectors. ␥␦ T cell numbers were found to be increased in the vaginal tract. Moreover, ␥␦ T cells isolated from the genital tract showed an activated phenotype and enhanced expression of cytokine gene. Altogether, our results demonstrate that AdC modulates ␥␦ T cell responses and suggest that this cell population may influence immune responses following vaginal immunization.
