In the context of multiplication and diversification of the requirements and social needs, public... more In the context of multiplication and diversification of the requirements and social needs, public administration, together with its authorities needs to operate through its entire activity to meet and resolve, under the best conditions, the public requirements and issues. Only by implementing the principles and the rules governing this matter and under the conditions in which the government deals with public affairs in an immediate and operational manner, efficiency will be ensured in meeting all interests, be they central or local. Constitutional principles, decentralization and local autonomy govern and underlie the formation, organization and functioning of local government, ensuring the interests and meet local needs, in concordance with the specifics of each administrative-territorial unit. Recognizing the legal personality of the administrative-territorial units and their right to self administrate in order to satisfy their own needs, the principle of decentralization and local autonomy gives the local government the mission to identify and to implement the optimal solutions for solving specific local administrative problems, and on behalf of the local interests which they represent. On the background of those exposed in the paper, analyzing the principles of decentralization and local autonomy, we can say that the exercise of this latter principle is possible only in the presence of the former, the basis for decentralization being the idea of local autonomy.

Protein tyrosine phosphatases (PTPs) play critical roles in signal transduction by dephosphorylat... more Protein tyrosine phosphatases (PTPs) play critical roles in signal transduction by dephosphorylating regulatory proteins involved in many signaling cascades. PTP inhibitors have been reported as potential drugs to treat diseases induced by PTP activity. In this study, we show that PTP inhibitor XIX (PI-19) inhibits dual-specificity phosphatase 22 (DUSP22). Kinetic analysis of PI-19 and DUSP22 suggests that PI-19 inhibits DUSP22 at the catalytic site. In addition, we performed drug affinity responsive targets stability (DARTS) analysis to reveal that the conformation of DUSP22 changed upon binding to PI-19. Moreover, PI-19 blocked DUSP22-mediated dephosphorylation of the signal transducer and activator of transcription 3 (STAT3) at Tyr-705, a critical site for STAT3 activity. Collectively, our results imply that PI-19 is an effective inhibitor of DUSP22 that regulates STAT3 activity.

(L. japonicum) have been used in traditional Chinese medicine for the treatment of various inflammatory diseases. However, the molecular mechanisms underlying their anti-inflammatory effects have yet to be elucidated. In the present study, we investigated the anti-inflammatory effects of ethanol extracts of L. japonicum spores (ELJ) by measuring the production of inflammatory mediators, and explored the molecular mechanisms underlying the effects of ELJ in murine macrophages in vitro using immunoblotting analyses. At non-cytotoxic concentrations of (50-300 µg/ml), ELJ was revealed to significantly suppress the production of nitric oxide (NO) and tumor necrosis factor (TNF)-α in lipopolysaccharide (LPS)-stimulated murine RAW 264.7 macrophages; ELJ repressed the production of interleukin (IL)-6 only at high concentrations (≥200 µg/ml). The ELJ-mediated decrease in NO production was demonstrated to depend on the downregulation of inducible NO synthase mRNA and protein expression. Conversely, the mRNA and protein expression of cyclooxygenase-2 were not affected by ELJ. In addition, ELJ was revealed to inhibit the mRNA expression of IL-6, IL-1β, and TNF-α in LPS-stimulated RAW 264.7 macrophages. The effects of ELJ on proinflammatory mediators may have been due to the stabilization of inhibitor of κBα and the inhibition of p38 mitogen-activated protein kinase (MAPK). These results suggested that ELJ may suppress LPS-induced inflammatory responses in murine macrophages in vitro, through the negative regulation of p38 MAPK and nuclear factor (NF)-κB. Therefore, ELJ may have potential as a novel candidate for the development of therapeutic strategies aimed at alleviating inflammation.

The rhizome of Anemarrhena asphodeloides Bunge (A. asphodeloides) has been used as a traditional ... more The rhizome of Anemarrhena asphodeloides Bunge (A. asphodeloides) has been used as a traditional East Asian medicine for the treatment of various types of inflammatory disease. However, to the best of our knowledge, there have been no systemic studies regarding the molecular mechanisms of action of the A. asphodeloides rhizome anti-inflammatory effects. The aim of the present study was to elucidate the anti-inflammatory effects and underlying mechanism of action of ethanol extracts of the rhizome of A. asphodeloides (EAA) in murine macrophages. Non-cytotoxic concentrations of EAA (10-100 µg/ml) significantly decreased the production of NO and interleukin (IL)-6 in lipopolysaccharide (LPS)-stimulated macrophages, while the production of tumor necrosis factor-α was not regulated by EAA. EAA-mediated reduction of nitric oxide (NO) was due to reduced expression levels of inducible NO synthase (iNOS). Furthermore, protein expression levels of LPS-induced cyclooxygenase-2, another inflammatory enzyme, were alleviated in the presence of EAA. EAA-mediated reduction of those proinflammatory mediators was due to inhibition of nuclear factor-κB (NF-κB) and activator protein 1 transcriptional activities followed by the stabilization of inhibitor of κ Bα and inhibition of p38, respectively. These results indicate that EAA suppresses LPS-induced inflammatory responses by negatively regulating p38 and NF-κB, indicating that EAA is a candidate treatment for alleviating inflammation.

To address the issue, this paper presents adaptive robust PID control subject to supervisory decoupled sliding mode control for an inverted pendulum system optimized by using a genetic algorithm optimization. Decoupled sliding mode control is a variable structure control method having appropriate features, such as good tracking performance and robustness with regard to disturbances. In the present study, Genetic Algorithm (GA) is employed as an effectual smart evolutionary algorithm to design optimally the control coefficients of an Inverted Pendulum (IP) and to eliminate tedious and repetitive trial-and-error process. When decoupled SMC is used to address the problem, there has to be a trade-off between the error of the position and the error of the angle. Hence, a Multi-objective Genetic algorithm is used to design optimally the Pareto front of the problem by regarding the errors as objective functions. Moreover, simulation has been done by considering the case with external disturbance and without external disturbance. Then, the results implemented in the MATLAB software environment are contrasted with the experimental results. Numerical results and comparison analysis elucidates the superiority of the optimal robust proposed controller over the traditional decoupled sliding mode controller in terms of attenuating the chattering and actuator variations.

In contrast to marine sediments, because of large electrical resistivity anomalies found in sulfi... more In contrast to marine sediments, because of large electrical resistivity anomalies found in sulfide deposits and methane hydrates, resistivity measurements such as marine towed electrical resistivity (MTER) might be a feasible method for discovering those natural minerals. To determine the feasibility of the MTER method we examined arrays consisting of a pole electrical dipole (PED), vertical electrical dipole (VED) and horizontal electrical dipole (HED). The VED array showed a maximum difference in electric fields of 36% and 105% in the resistive and conductive models, respectively, while the PED and HED arrays yielded worse results of around 13% to 19%, respectively. The VED array showed a higher difference in electric fields than both the HED and PED arrays in the two models. Therefore, we suggest that a VED array with a large electrical current would be most conducive leading to the discovery of such minerals during MTER surveys.

Signals from ten explosions were used to examine earthquake location uncertainty in Taiwan. Locat... more Signals from ten explosions were used to examine earthquake location uncertainty in Taiwan. Location errors for explosion sites determined using a relocation process were expressed in terms of statistical measurements for standard errors in the depth (ERZ), the epicenter (ERH), the root-mean-square of the travel time residuals (RMS), and the station coverage gap (GAP). In general, for this study, major factors in location errors resulted from the poor coverage of seismic stations (e.g., a large value of GAP). Using the relationship between location errors and parameters from the explosion location assessment, uncertainties for earthquake locations in the Taiwan region for a total of 384064 events from 1991 to 2011 were evaluated. Offshore regions in southwestern and northeastern Taiwan had larger location errors. For the inland locales, location errors in longitude, latitude, and depth were approximately 3.1 ± 2.7, 1.3 ± 1.6, and 4.6 ± 3.9 km, respectively. The uncertainties estimated from this study could offer a good reference for other related studies.
