Intense exercise is a physiological stress capable of inducing damage in the exercised muscle gro... more Intense exercise is a physiological stress capable of inducing damage in the exercised muscle group. As a consequence of this exercise-induced micro lesion, there is an increase in the interaction of leukocytes (neutrophils and monocytes/macrophages) with muscle endothelial cells and their transmigration into skeletal muscle tissue. Mechanisms driving this physiological response are not known, but exercise-induced muscle damage may be associated with a local inflammation involving leukocyte accumulation in damaged muscle tissue. Important recent findings show that this infiltration of leukocytes plays an important role in the skeletal muscle remodeling process by activating angiogenesis and hypertrophic pathways inside the skeletal muscle tissue. There are some speculations suggesting that this inflammatory process should be controlled by anti-inflammation drugs in athletes to accelerate the recovery time. Investigations of the literature in this field up to this point do not offer a sufficient body of evidence confirming the need for an anti-inflammatory drug administration to control the exercise-induced inflammation. On the other hand, anti-inflammatory drug administration as a supplemental diet could inhibit important pathways of tissue adaptative responses and block the benefits of exercise to the skeletal muscle tissue.

Reactive species and free radicals are unstable molecules that oxidize other molecules in order to become stable. Although they play important roles in our body, they can also lead to oxidative stress impairing diverse cellular functions. During exercise, reactive species can be produced mainly, but not exclusively, by the following mechanisms: electron leak at the mitochondrial electron transport chain, ischemia/reperfusion and activation of endothelial xanthine oxidase, inflammatory response, and autooxidation of catecholamines. Objective Interleukin-4 (IL-4) is a multifunctional cytokine involved in many diseases such as au... more Objective Interleukin-4 (IL-4) is a multifunctional cytokine involved in many diseases such as autoimmune hepatitis and idiosyncratic drug reactions. However, its role in acetaminophen (APAP)-induced liver injury remains unclear. Our objective was to evaluate the contribution of IL-4 to the pathogenesis of APAP-induced liver injury. Methods Balb/C (WT) and IL-4 knockout (IL-4-/-) mice were orally overdosed with APAP. After 24 h, survival percentage, biochemical and morphological markers of liver injury, and tissue inflammation were assessed. Results IL-4-/mice were protected from APAP toxicity. Intravital confocal microscopy, tissue histology and serum ALT levels showed significantly less liver injury and inflammation than in the WT group, which may explain the increased survival rate of IL-4-/mice. In addition, IL-4-/mice had decreased production of tumor necrosis factor a, CXCL1 and interleukin-1b in the liver, but not in a remote site such as the lungs. Hepatic macrophage activation was markedly reduced in IL-4-deficient mice. In addition, glutathione depletion-a primary cause of APAPmediated injury-was significantly attenuated in IL-4-/mice. Conclusions Taken together, our data demonstrate that IL-4-/mice are protected from APAP-induced liver injury due to reduced depletion of glutathione, which prevented liver damage and tissue inflammation.

Intense exercise is a physiological stress capable of inducing the interaction of neutrophils wit... more Intense exercise is a physiological stress capable of inducing the interaction of neutrophils with muscle endothelial cells and their transmigration into tissue. Mechanisms driving this physiological inflammatory response are not known. Here, we investigate whether production of reactive oxygen species is relevant for neutrophil interaction with endothelial cells and recruitment into the quadriceps muscle in mice subjected to the treadmill fatiguing exercise protocol. Mice exercised until fatigue by running for 56.366.8 min on an electric treadmill. Skeletal muscle was evaluated by intravital microscopy at different time points after exercise, and then removed to assess local oxidative stress and histopathological analysis. We observed an increase in plasma lactate and creatine kinase (CK) concentrations after exercise. The numbers of monocytes, neutrophils, and lymphocytes in blood increased 12 and 24 hours after the exercise. Numbers of rolling and adherent leukocytes increased 3, 6, 12, and 24 hours post-exercise, as assessed by intravital microscopy. Using LysM-eGFP mice and confocal intravital microscopy technology, we show that the number of transmigrating neutrophils increased 12 hours post-exercise. Mutant gp91 phox-/-(non-functional NADPH oxidase) mice and mice treated with apocynin showed diminished neutrophil recruitment. SOD treatment promoted further adhesion and transmigration of leukocytes 12 hours after the exercise. These findings confirm our hypothesis that treadmill exercise increases the recruitment of leukocytes to the postcapillary venules, and NADPH oxidase-induced ROS plays an important role in this process.

Purpose We investigated lower limb strength training (LLST)’s effect on the levels of renin angio... more Purpose We investigated lower limb strength training (LLST)’s effect on the levels of renin angiotensin system (RAS) molecules in urine and serum. Methods Twelve male volunteers with 23.4 ± 2.7 years old performed our LLST protocol for 10 weeks. The effect of LLST was evaluated based on urine and serum levels of RAS molecules, including Angiotensin II, Ang-(1–7), angiotensin converting enzyme ACE 1 and ACE 2, before and immediately after the first (S1) and the last (S30) training sessions. Results The training protocol significantly reduced the percentage of fat and increased the total lean mass. Serum and urinary levels of Ang II were higher after the last session in comparison to levels before this session as well as if compared to values after the first exercise session. Levels of Ang-(1–7) significantly increased in serum and urine after first and last sessions and in the comparison to values before and after last session to those before and after first session, respectively. Levels of ACE1 and ACE2 did not modify in any condition. In addition, LLST seems to chronically shift the balance between both RAS axes toward the alternative axis based on the analysis of Ang-(1–7)/Ang II ratio. Conclusion LLST acutely and chronically modified serum and urinary concentrations of Ang II and Ang-(1–7).

This study aimed to determine if the level of hepatic oxidative stress resulting from high-intensity interval training (HIIT) is affected by the frequency pattern (consecutive vs. non-consecutive) of the training sessions. Thirty-two Wistar rats were divided equally into four groups: two control groups, (CS1) and (CS2), remained sedentary throughout the experiment, and two test groups, (CT1) and (CT2), and were submitted to HIIT for 12 consecutive and non-consecutive (12 sessions, 3 times/wk over four weeks) days, respectively. However, it remains unclear how physical training regulates exercise-induced inflammation and performance. Here, we demonstrate that acute high intensity exercise promotes an inflammatory profile characterized by increased blood IL-6 levels, neutrophil migratory capacity, and leukocyte recruitment to skeletal muscle vessels. Moreover, we found that physical training amplified leukocyte–endothelial cell interaction induced by acute exercise in skeletal muscle vessels and diminished exercise-induced inflammation in skeletal muscle tissue. Furthermore, we verified that disruption of the gp-91 subunit of NADPH-oxidase inhibited exercise-induced leukocyte recruitment on skeletal muscle after training with enhanced exercise time until fatigue. Realizaram-se 15 entrevistas semiestruturadas com 12 mentores e três discentes que ocupam representações estudantis. Utilizou-se análise de conteúdo, possibilitando delineamento de quatro categorias: a) resultados preliminares obtidos; b) potencialidades do instrumento; c) barreiras e enfrentamentos; d) propostas de aperfeiçoamento. Observou-se que a maioria das metas teve ações norteadoras iniciadas, mas nenhuma concluída. Ressalta-se que o contexto da pandemia mundial (covid-19) exerceu significativo impacto, interferindo diretamente nos resultados. Purpose We investigated lower limb strength training (LLST)’s effect on the levels of renin angio... more Purpose We investigated lower limb strength training (LLST)’s effect on the levels of renin angiotensin system (RAS) molecules in urine and serum. Methods Twelve male volunteers with 23.4 ± 2.7 years old performed our LLST protocol for 10 weeks. The effect of LLST was evaluated based on urine and serum levels of RAS molecules, including Angiotensin II, Ang-(1–7), angiotensin converting enzyme ACE 1 and ACE 2, before and immediately after the first (S1) and the last (S30) training sessions. Results The training protocol significantly reduced the percentage of fat and increased the total lean mass. Serum and urinary levels of Ang II were higher after the last session in comparison to levels before this session as well as if compared to values after the first exercise session. Levels of Ang-(1–7) significantly increased in serum and urine after first and last sessions and in the comparison to values before and after last session to those before and after first session, respectively. Levels of ACE1 and ACE2 did not modify in any condition. In addition, LLST seems to chronically shift the balance between both RAS axes toward the alternative axis based on the analysis of Ang-(1–7)/Ang II ratio. Conclusion LLST acutely and chronically modified serum and urinary concentrations of Ang II and Ang-(1–7).

This study aimed to determine if the level of hepatic oxidative stress resulting from high-intensity interval training (HIIT) is affected by the frequency pattern (consecutive vs. non-consecutive) of the training sessions. Thirty-two Wistar rats were divided equally into four groups: two control groups, (CS1) and (CS2), remained sedentary throughout the experiment, and two test groups, (CT1) and (CT2), and were submitted to HIIT for 12 consecutive and non-consecutive (12 sessions, 3 times/wk over four weeks) days, respectively. However, it remains unclear how physical training regulates exercise-induced inflammation and performance. Here, we demonstrate that acute high intensity exercise promotes an inflammatory profile characterized by increased blood IL-6 levels, neutrophil migratory capacity, and leukocyte recruitment to skeletal muscle vessels. Moreover, we found that physical training amplified leukocyte–endothelial cell interaction induced by acute exercise in skeletal muscle vessels and diminished exercise-induced inflammation in skeletal muscle tissue. Furthermore, we verified that disruption of the gp-91 subunit of NADPH-oxidase inhibited exercise-induced leukocyte recruitment on skeletal muscle after training with enhanced exercise time until fatigue. Monitorar e analisar a execução das metas pactuadas no Planejamento Estratégico da Escola de Educ... more Monitorar e analisar a execução das metas pactuadas no Planejamento Estratégico da Escola de Educação Física da Universidade Federal de Ouro Preto, nos 18 primeiros meses de implementação. Realizaram-se 15 entrevistas semiestruturadas com 12 mentores e três discentes que ocupam representações estudantis. Utilizou-se análise de conteúdo, possibilitando delineamento de quatro categorias: a) resultados preliminares obtidos; b) potencialidades do instrumento; c) barreiras e enfrentamentos; d) propostas de aperfeiçoamento. Observou-se que a maioria das metas teve ações norteadoras iniciadas, mas nenhuma concluída. Ressalta-se que o contexto da pandemia mundial (covid-19) exerceu significativo impacto, interferindo diretamente nos resultados. O estudo apresenta caráter de monitoramento em relação à ferramenta de gestão, tendo em vista a relevância de implementar mecanismos para avaliação sistematizada dos indicadores de desempenho institucionais.
