Sponges are a species of marine biota that can be used as a producer of antimicrobial compounds. The bacteria found in sponges have an important role in the continuity of life in the sea. The symbiotic lifestyle that occurs in bacteria and sponges has the opportunity to form substitutions for the content of secondary metabolites and enzymes, especially extracellular enzymes (amylase, protease, cellulose and lipase). This study aims to determine how to isolate sponge symbiotic bacteria and identify spongy symbiotic bacteria. The method used is purposive to take sponges. Characterization of bacteria was carried out based on morphology and gram staining. Enzymatic bacterial selection was carried out by testing the activity of the amylase enzyme (soluble starch), lipase enzyme (Teen 80), protease enzyme (skim milk) and cellulose enzyme (carboxyl methyl cellulose). Data analysis was performed in a qualitative descriptive manner by measuring the clear zone in the extracellular enzyme test results. Meanwhile, other research parameters measured in this study were temperature, salinity and pH. The results showed that out of 47 isolated bacteria and 33 of them had extracellular enzymes with 10 bacteria had amylase enzymes, 27 bacteria had lipase enzymes, 2 bacteria had protease enzymes and 1 isolate had cellulose enzymes

Everyone needs to be given understanding in choosing healthy foods for the body. β-carotene is one of the good compounds contained in vegetables and fruits. This research was conducted to determine the content of β-carotene compounds in fruit (Arenga pinata). Samples were obtained from Baumata village. This research uses thin layer chromatography (TLC) and spectrophotometry methods. The research data were analyzed based on linear regression equations and relevant reference studies. The results of the study using the TLC method showed that the Rf value for palm fruit was 0.943. Spectrophotometric examination showed the content of β-carotene compounds from palm fruit extract at a wavelength of 470 nm had a concentration of 26.18 ppm. Based on the results of these studies it can be concluded that the palm fruit contains β-carotene. Thus palm fruit can be used as one of the choices of healthy food for the community and also as a choice of sources of vitamin A.

Palm oil (Arenga pinnata Merr.) is a plantation plant that has the potential to be cultivated in ... more Palm oil (Arenga pinnata Merr.) is a plantation plant that has the potential to be cultivated in the future, because it has high economic value and is prospective for commercial cultivation considering its various uses. Enau is a plant that has been developed from generation to generation on the island of Timor. The community in Nekmese Village manages enau still in the traditional way. Vitamin C or ascorbic acid is one of the vitamins made from hexose derivatives that are soluble in water and easily oxidized. The purpose of this study was to determine the content of vitamin C in the flesh of palm fruit. This research was carried out in May 2019 at the Kupang State Agricultural Polytechnic TPH Laboratory. Determination of the vitamin C content in the sample was determined by titration using an iodine solution. The content of vitamin C in palm fruit flesh based on the temperature treatment of 5°C 10°C and 15°C showed that storage treatment at 5°C had the highest vitamin C content with an average of 4.14 mg followed by storage at 10°C with an average of 3.33 mg and storage at a temperature of 15°C with an average vitamin C content of 3.02 mg. This study also showed that the longer the palm fruit is stored, the lower the vitamin C content in the palm fruit.

The prevalence of fungal infections and the occurrence of fungi in mangrove leaves from in natura... more The prevalence of fungal infections and the occurrence of fungi in mangrove leaves from in natural forests and reforestation sites were compared in three areas from Philippines: Central Visayas-Bais, Negros Oriental, Alcantara, Cebu and Pangangan Island, Bohol. Three diseases were identified, namely the Brown Leaf Spot disease (BRS), Black Leaf Spot disease (BLS) and White Leaf Spot disease (WLS). BRS was found in Sonneratia alba and Avicennia marina in natural forests and reforestation sites in all areas, while BLS and WLS were found only in Bais, with BLS occurring in Ceriops decandra and Osbornia octodonta in natural forests and in Rhizophora stylosa in reforested sites. WLS occurred only in A. marina in natural forests. Disease prevalence for BRS was significantly higher in reforested areas (F=8.477; p=0.001). 4 fungi genera were consistently found in infected leaves: Aspergillus, Penicillium, Pseudocercosporella and Rhizopus. The occurrence percentage was higher in natural forests than in reforestation sites for all fungi.

Tumbuhan mangrove merupakan salah satu ekosistem pantai yang mendominasi wilayah pantai suatu dae... more Tumbuhan mangrove merupakan salah satu ekosistem pantai yang mendominasi wilayah pantai suatu daerah dan berpotensi untuk dikembangkan dalam penanggulangan berbagai penyakit baik infeksi pada ikan budidaya maupun sebagai medi pengobatan alami bagi manusia, karena senyawa metaboit sekunder yang terkandung didalamnya mampu berkhasiat sebagai antibakteri. Tujuan dari penelitian ini untuk mengetahui pengaruh ekstrak kasar kulit batang mangrove A. marina terhadap pertumbuhan bakteri E. coli dan konsentrasi optimum dari estrak kasar kulit batang mangrove A. marina dalam menghambat pertumbuhan bakteri E. coli. Metode yang digunakan dalam penelitian ini metode eksperimen laboratorium dengan 4 perlakuan yaitu konsentrasi 55 mg/mL, 60 mg/mL, 65 mg/mL, 70 mg/mL dan 1 kontrol tanpa pemberian ekstrak dengan 3 kali ulangan. Hasil pengamatan diameter zona hambat dianalisis secara statistik menggunakan analisis variansi (ANOVA) satu jalur dengan signifikansi 5%. Analisis statistik dilakukan menggunakan program SPSS versi 16.0. Hasil penelitian menunjukkan bahwa ekstrak kasar kulit batang mangrove A. marina berpengaruh terhadap pertumbuhan bakteri E. coli. Hal ini dapat dilihat dar konsentrasi yang digunakan mulai dari 55 mg/mL, 60 mg/mL, 65 mg/mL, 70 mg/mL memiliki rata-rata zona hambat yang berkisar antara 26 mm, 27.33 mm, 27.66 mm dan 28.66 mm. Dengan demikian semakin tinggi konsentrasi ekstrak yang digunakan, maka semakin besar daya hambat yang terbentuk.

Poir) merupakan salah satu sayuran yang kaya akan βkaroten sebagai prekursor vitamin A dan memiliki kandungan karbohidrat yang cukup tinggi. Pengolahan labu kuning dapat dilakukan untuk menghasilkan berbagai macam produk salah satunya cake. Cake merupakan adonan panggang dan adonan liquid yang terbuat dari tepung terigu, gula, garam, bahan pengembang, shortening, susu, telur, dan penambah aroma. Penelitian ini bertujuan untuk mengetahui kadar β-karoten yang terdapat pada cake labu kuning dan untuk mengetahui daya terima β-karoten pada cake labu kuning. Penelitian ini merupakan penelitian eksperimen. Rancangan percobaan yang digunakan yaitu Rancangan Acak Lengkap (RAL) dengan 1 kontrol 4 perlakuan dan masing-masing perlakuan diulang sebanyak 3 kali, selain β-Karoten yang diuji 2 kali. Tahap penelitian terdiri dari pembuatan tepung dan pembuatan cake. Metode pengumpulan data meliputi uji organoleptik dan uji β-karoten. Data dianalisis menggunakan analisa varian/ANOVA untuk mengetahui perbedaan kualitas, dan apabila terdapat pengaruh maka dilanjutkan dengan uji Tukey. Hasil penelitian uji organoleptik menunjukan bahwa pada parameter warna tidak memberikan pengaruh yang nyata, sehingga tidak dilanjutkan dengan uji Tukey, dari segi aroma yaitu A1 merupakan perlakuan terbaik dengan nilai 3,56, dari segi tekstur perlakuan terbaik adalah A1 dengan nilai 4,04 dan dari segi rasa perlakuan terbaik adalah A1 dengan nilai 3,80. Untuk uji β-karoten, perlakuan cake labu kuning dengan kadar β-karoten tertinggi terdapat pada A4 dengan rata-rata 0,262 dan kadar terendah terdapat pada A0 dengan rata-rata 0,018. Berdasarkan data hasil penelitian, tepung labu kuning layak dijadikan salah satu sumber tepung alternatif karena cake yang dihasilkan memenuhi standar mutu SNI baik dari segi warna, rasa, aroma, tekstur maupun kadar β-karoten.

Watumbolo forest is located in South West Sumba, with the average temperature ranges from 21-34°C, therefore the varians types of plants grow well in this forest, including ferns. Sponges are a species of marine biota that can be used as a producer of antimicrobial compounds. The bacteria found in sponges have an important role in the continuity of life in the sea. The symbiotic lifestyle that occurs in bacteria and sponges has the opportunity to form substitutions for the content of secondary metabolites and enzymes, especially extracellular enzymes (amylase, protease, cellulose and lipase). This study aims to determine how to isolate sponge symbiotic bacteria and identify spongy symbiotic bacteria. The method used is purposive to take sponges. Characterization of bacteria was carried out based on morphology and gram staining. Enzymatic bacterial selection was carried out by testing the activity of the amylase enzyme (soluble starch), lipase enzyme (Teen 80), protease enzyme (skim milk) and cellulose enzyme (carboxyl methyl cellulose). Data analysis was performed in a qualitative descriptive manner by measuring the clear zone in the extracellular enzyme test results. Meanwhile, other research parameters measured in this study were temperature, salinity and pH. Everyone needs to be given understanding in choosing healthy foods for the body. β-carotene is one of the good compounds contained in vegetables and fruits. This research was conducted to determine the content of β-carotene compounds in fruit (Arenga pinata). Samples were obtained from Baumata village. This research uses thin layer chromatography (TLC) and spectrophotometry methods. The research data were analyzed based on linear regression equations and relevant reference studies. The results of the study using the TLC method showed that the Rf value for palm fruit was 0.943. Spectrophotometric examination showed the content of β-carotene compounds from palm fruit extract at a wavelength of 470 nm had a concentration of 26.18 ppm. Based on the results of these studies it can be concluded that the palm fruit contains β-carotene. Thus palm fruit can be used as one of the choices of healthy food for the community and also as a choice of sources of vitamin A.

Enau is a plant that has been developed from generation to generation on the island of Timor. The community in Nekmese Village manages enau still in the traditional way. Vitamin C or ascorbic acid is one of the vitamins made from hexose derivatives that are soluble in water and easily oxidized. The purpose of this study was to determine the content of vitamin C in the flesh of palm fruit. This research was carried out in May 2019 at the Kupang State Agricultural Polytechnic TPH Laboratory. Determination of the vitamin C content in the sample was determined by titration using an iodine solution. The content of vitamin C in palm fruit flesh based on the temperature treatment of 5°C 10°C and 15°C showed that storage treatment at 5°C had the highest vitamin C content with an average of 4.14 mg followed by storage at 10°C with an average of 3.33 mg and storage at a temperature of 15°C with an average vitamin C content of 3.02 mg. This study also showed that the longer the palm fruit is stored, the lower the vitamin C content in the palm fruit.

The prevalence of fungal infections and the occurrence of fungi in mangrove leaves from in natura... more The prevalence of fungal infections and the occurrence of fungi in mangrove leaves from in natural forests and reforestation sites were compared in three areas from Philippines: Central Visayas-Bais, Negros Oriental, Alcantara, Cebu and Pangangan Island, Bohol. Three diseases were identified, namely the Brown Leaf Spot disease (BRS), Black Leaf Spot disease (BLS) and White Leaf Spot disease (WLS). BRS was found in Sonneratia alba and Avicennia marina in natural forests and reforestation sites in all areas, while BLS and WLS were found only in Bais, with BLS occurring in Ceriops decandra and Osbornia octodonta in natural forests and in Rhizophora stylosa in reforested sites. WLS occurred only in A. marina in natural forests. Disease prevalence for BRS was significantly higher in reforested areas (F=8.477; p=0.001). 4 fungi genera were consistently found in infected leaves: Aspergillus, Penicillium, Pseudocercosporella and Rhizopus. The occurrence percentage was higher in natural forests than in reforestation sites for all fungi.

Tumbuhan mangrove merupakan salah satu ekosistem pantai yang mendominasi wilayah pantai suatu dae... more Tumbuhan mangrove merupakan salah satu ekosistem pantai yang mendominasi wilayah pantai suatu daerah dan berpotensi untuk dikembangkan dalam penanggulangan berbagai penyakit baik infeksi pada ikan budidaya maupun sebagai medi pengobatan alami bagi manusia, karena senyawa metaboit sekunder yang terkandung didalamnya mampu berkhasiat sebagai antibakteri. Tujuan dari penelitian ini untuk mengetahui pengaruh ekstrak kasar kulit batang mangrove A. marina terhadap pertumbuhan bakteri E. coli dan konsentrasi optimum dari estrak kasar kulit batang mangrove A. marina dalam menghambat pertumbuhan bakteri E. coli. Metode yang digunakan dalam penelitian ini metode eksperimen laboratorium dengan 4 perlakuan yaitu konsentrasi 55 mg/mL, 60 mg/mL, 65 mg/mL, 70 mg/mL dan 1 kontrol tanpa pemberian ekstrak dengan 3 kali ulangan. Hasil pengamatan diameter zona hambat dianalisis secara statistik menggunakan analisis variansi (ANOVA) satu jalur dengan signifikansi 5%. Analisis statistik dilakukan menggunakan program SPSS versi 16.0. Hasil penelitian menunjukkan bahwa ekstrak kasar kulit batang mangrove A. marina berpengaruh terhadap pertumbuhan bakteri E. coli. Hal ini dapat dilihat dar konsentrasi yang digunakan mulai dari 55 mg/mL, 60 mg/mL, 65 mg/mL, 70 mg/mL memiliki rata-rata zona hambat yang berkisar antara 26 mm, 27.33 mm, 27.66 mm dan 28.66 mm. Dengan demikian semakin tinggi konsentrasi ekstrak yang digunakan, maka semakin besar daya hambat yang terbentuk.

Poir) merupakan salah satu sayuran yang kaya akan βkaroten sebagai prekursor vitamin A dan memiliki kandungan karbohidrat yang cukup tinggi. Pengolahan labu kuning dapat dilakukan untuk menghasilkan berbagai macam produk salah satunya cake. Cake merupakan adonan panggang dan adonan liquid yang terbuat dari tepung terigu, gula, garam, bahan pengembang, shortening, susu, telur, dan penambah aroma. Penelitian ini bertujuan untuk mengetahui kadar β-karoten yang terdapat pada cake labu kuning dan untuk mengetahui daya terima β-karoten pada cake labu kuning. Penelitian ini merupakan penelitian eksperimen. Rancangan percobaan yang digunakan yaitu Rancangan Acak Lengkap (RAL) dengan 1 kontrol 4 perlakuan dan masing-masing perlakuan diulang sebanyak 3 kali, selain β-Karoten yang diuji 2 kali. Tahap penelitian terdiri dari pembuatan tepung dan pembuatan cake. Metode pengumpulan data meliputi uji organoleptik dan uji β-karoten. Data dianalisis menggunakan analisa varian/ANOVA untuk mengetahui perbedaan kualitas, dan apabila terdapat pengaruh maka dilanjutkan dengan uji Tukey. Hasil penelitian uji organoleptik menunjukan bahwa pada parameter warna tidak memberikan pengaruh yang nyata, sehingga tidak dilanjutkan dengan uji Tukey, dari segi aroma yaitu A1 merupakan perlakuan terbaik dengan nilai 3,56, dari segi tekstur perlakuan terbaik adalah A1 dengan nilai 4,04 dan dari segi rasa perlakuan terbaik adalah A1 dengan nilai 3,80. Untuk uji β-karoten, perlakuan cake labu kuning dengan kadar β-karoten tertinggi terdapat pada A4 dengan rata-rata 0,262 dan kadar terendah terdapat pada A0 dengan rata-rata 0,018. Berdasarkan data hasil penelitian, tepung labu kuning layak dijadikan salah satu sumber tepung alternatif karena cake yang dihasilkan memenuhi standar mutu SNI baik dari segi warna, rasa, aroma, tekstur maupun kadar β-karoten.
