Objectives: The aim of the present study was to investigate the misuse of levothyroxine treatment... more Objectives: The aim of the present study was to investigate the misuse of levothyroxine treatment in patients with hypothyroidism. Methods: Patients who received levothyroxine treatment were included in the study. Age, gender, duration of medication use, Thyrotropin (TSH) level, regular use of medication, time and manner of taking medication, drug storage conditions and use of additional medication were recorded. Taking the medication on a full stomach or in the evening, occasional use of medication, storing the medicine in the refrigerator, and the use of additional medication were classified as misuse of levothyroxine treatment. According to the levels of TSH: 0.4-4.5 mIU/L was considered a normal dose, while 4.5 mIU/L and above was considered insufficient, and 0.4 mIU/L and below was considered as high dose. Results: Patients who received levothyroxine pill were included in the study (average age: 53 ± 14 years, average duration of medication use: 48 ± 50 months and females/male ratio: 363/44). 323 (79.4%) of patients stated that they take the medicine regularly. Levothyroxine dosage was found to be sufficient in 235 patients (57.7%), insufficient in 101 patients (24.8%) and high in 71 patients (17.4%). Among the 84 patients which were identified as misusing, 66 (16.2%) patients were taking the medicine on a full stomach, 12 (2.9%) patients were taking the medicine in the evening, 11 patients (2.7%) were storing the medicine in the refrigerator, 17 (4.2%) patients were taking the medicine occasionally, and 21 (5.1%) patients were using one or more additional medications. Conclusions: Taking the medication on a full stomach or in the evening, occasional use of medication, storing the medicine in the refrigerator, and the use of additional medications were the major factors for insufficient TSH levels. Appropriate usage of levothyroxine pills is crucial for successful treatment.

Background: Inflammatory hematological parameters like the neutrophil/lymphocyte (N/L) ratio have... more Background: Inflammatory hematological parameters like the neutrophil/lymphocyte (N/L) ratio have been investigated in many cancer types and significant relationships found with prognosis, for example. The aim of this present study was to investigate the impact of hematological parameters notably on N/L ratio and mean platelet volume (MPV) in papillary thyroid cancer cases. Materials and Methods: A total of 79 patients who underwent a thyroidectomy operation in Fındıklı, Goiter Research and Treatment Center during 2011-2015 period were enrolled in the study, 41 with papillary thyroid cancer and 38 with benign goiter confirmed by pathological examination. We collected clinical and laboratory data for the patients from hospital records retrospectively. Blood samples taken at admission were assessed for parameters compared between the groups. Results: No significant differences between papillary thyroid cancer and benign goiter groups were apparent in terms of age, the N/L ratio, MPV, white blood cell count (WBC), red blood cell count (RBC), hemoglobin, hematocrit, platelet, neutrophil, lymphocyte, red blood cell distribution width (RDW) and platelet crit (PCT) levels (p>0.05). Only the level of platelet distribution width (PDW) significantly differed, being lower in the papillary cancer group (p<0.05). Conclusions: No significant relationship between papillary thyroid cancer and inflammatory hematological parameters including in particular the N/L ratio and MPV. The relevance of the PDW values remains unclear.

The quality and biochemical properties of honey are affected by heating or during storage period.... more The quality and biochemical properties of honey are affected by heating or during storage period. The most important biochemical reaction that occurs in this process is the Maillard reaction. HMF (5-hydroxymethylfurfural) is one of the major intermediate products in the Maillard reaction that can lead to quality reduction in heated honey. In this study, the effect of heating on the antioxidant activity, and colour values as Maillard reaction indicators of three different botanical honeys were investigated; the HMF formation was also determined. Temperatures of 50, 70 and 80 °C were applied on the honeys between 0 and 48 h. Total phenolic content, ferric reducing/antioxidant power and scavenging of 2,2-diphenyl-1-picrylhydrazyl free radical assays are used to determine the antioxidant capacity. Results showed that the formation of HMF and the antioxidant properties of honeys were significantly increased during the heating process. Pure HMF compound also showed lower antioxidant activity. The formation of HMF has higher degree of linearity in the fit of the zero order reaction and also it was the highest in the chestnut honey. Furthermore, it was found that as the biochemical value of the honeys increased, the HMF formation decelerated. Introduction: This study investigated the in vitro inhibition properties of newly synthesized cou... more Introduction: This study investigated the in vitro inhibition properties of newly synthesized coumarin derivates (C1-C5) against human carbonic anhdyrase I (hCA-I) and jack bean urease. Activities were expressed as IC50 (mg/mL), the concentration reducing 50% of the enzymes. The IC50 values for hCA-I ranged 5.20 µM from 12.10 µM, with compound C3 exhibiting the highest activity. The inhibition values for urease ranged 22.30 µM to 39.00 µM, the highest activity being observed in C5. Conclusion: Comparing the inhibitions with standard inhibitors of the enzymes, while the samples exhibited moderate inhibitions against hCA I, high inhibition was determined against urease.

The aim of this study was to investigate the inhibition effects of various phenolic standards, honey, propolis, and pollen species on human carbonic anhydrase I and II. The inhibition values (IC50) of the phenolics (gallic acid, protocatechuic acid, quercetin, catechin, tannic acid, and chrysin) ranged from 0.009 to 0.32 μg/mL, tannic acid emerging as the best inhibitor. The inhibition values of three different types of honey, heather, rhododendron, and chestnut ranged between 2.32 and 25.10 μg/mL, the chestnut honeys exhibiting the best inhibition. The ethanolic extracts of pollen and propolis exhibited good inhibitory properties, with IC50 values between 0.486 and 3.320 μg/mL. Honey adulteration is a serious ethical problem and results in many losses such as in nutrition, ... more Honey adulteration is a serious ethical problem and results in many losses such as in nutrition, health and economy. While adulteration of honey is very easy, it is difficult to determine it and requires troublesome techniques. The aim of the present study was to determine some physical and biochemical to differentiated parameters between the natural and adulterated with saccharose syrup honeys. Therefore, moisture, color, optical rotation, fructose, glucose, maltose, ribose, arabinose, proline, 5-hydroxymethlfurfural (HMF), total phenolic substances and total antioxidant capacities were measured to find any difference. Proline content, total amount of phenolic substances were found as important parameters that can be used to distinguish natural honey from that produced by over-feeding of bees with saccharine.

Monoamine oxidase (MAO) inhibitors are generally used in the treatment of depressive disorders an... more Monoamine oxidase (MAO) inhibitors are generally used in the treatment of depressive disorders and some neurodegenerative illnesses, such as Parkinson's disease and Alzheimer's disease. The aim of this preliminary study was to investigate the MAO [MAO (E.C.1.4.3.4)] inhibiting effect of various apitherapeutic products, such as chestnut honey, pollen and propolis. Extracts' MAO inhibition was measured using peroxidase-linked spectrophotometric assay in enzyme isolated from rat liver microsomes, and the values are expressed as the inhibition concentration (IC50) causing 50% inhibition of MAO. The antioxidant activity of the bee products was also determined in terms of total phenolic content (TPC) and ferric reducing/antioxidant power in aquatic extracts. All samples exhibited substantial inhibition of MAO, propolis having the highest. Inhibition was related to samples' TPCs and antioxidant capacities. These results show that bee products possess a sedative effect and may be effective in protecting humans against depression and similar diseases.

An α-carbonic anhydrase (CA, EC 4.2.1.1) was purified and characterized kinetically from erythroc... more An α-carbonic anhydrase (CA, EC 4.2.1.1) was purified and characterized kinetically from erythrocytes of the sturgeon Acipenser gueldenstaedti, an endangered species. The sturgeon enzyme (AgCA) showed kinetic parameters for the CO 2 hydration reaction comparable with those of the human erythrocytes enzyme hCA II, being a highly active enzyme, whereas its esterase activity with 4-nitrophenyl acetate as substrate was lower. Sulphonamide inhibitors (acetazolamide, sulphanilamide) strongly inhibited AgCA, whereas metal ions (Ag + , Zn 2+ , Cu 2+ and Co 2+) were weak, millimolar inhibitors. Several widely used pesticides (2,4-dichlorophenol, dithiocarbamates, parathion and carbaryl) were also assayed as inhibitors of this enzyme. The dithiocarbamates were low micromolar AgCA inhibitors (IC 50 of 16-18 µM), whereas the other pesticides inhibited the enzyme with IC 50 s in the range of 102-398 µM. The wide use of dithiocarbamate pesticides may be one of the factors enhancing the vulnerability of this sturgeon species to pollutants.

Arı sütü binlerce çiçekten özümsenen nektar ve çiçek polenleri ile bal arılarının enzimlerini de ... more Arı sütü binlerce çiçekten özümsenen nektar ve çiçek polenleri ile bal arılarının enzimlerini de içeren, akıcı jel halinde ve kremsi-beyaz renkteki etkili bir salgıdır. Besin değeri yüksek bu ürün amino asit, peptit, protein ve minerallerden oluşur. Arı sütünün en önemli özelliği diğer arı ürünlerinde bulunmayan kısa zincirli hidroksi yağ asitlerince zengin oluşudur. Antibakteriyel, anti-tümoral, ve immün sistemi uyarıcı özelliğe sahip arı sütünün majör yağ asidi trans-10-hidroksi-2-dekanoik asit (10-HDA)'dır. Yapılan bu çalışma ile farklı yıllarda toplanan arı sütü örneklerinin nem ve 10-HDA miktarı tayin edildi. HPLC-UV ile ölçülen 10-HDA miktarının yıllara göre ortalama %2.1 ile %2.6 arasında değiştiği tespit edildi. Arı sütü nem içeriğinin ise ortalama %62.6 ile %66.5 arasında değiştiği tespit edildi.

In this study, we investigated 17 different phenolic constituents and total antioxidant propertie... more In this study, we investigated 17 different phenolic constituents and total antioxidant properties of cherry laurel, Laurocerasus officinalis Roem (family Rosaceae), locally named karayemis or taflan, a summer fruit highly characteristic of the Black Sea region. Phenolic constituents were measured by reverse phase-high performance liquid chromatography (RP-HPLC). Total phenolic compounds, total flavonoids, ferric reducing /antioxidant power (FRAP), cupric ion reducing capacity (CUPRAC) and 2,2diphenly-1-picrylhydrazyl (DPPH) radical methods were used to evaluate the antioxidant capacity. The total phenolics and total flavonoids were found to be 1.094 g GAE/100 g DW and 0.080 g QUE/100 g DW, respectively. Chlorogenic acid was found, that is, the main phenolic component of the methanolic extract of the fruit. An α-carbonic anhydrase (CA, EC 4.2.1.1) was purified and characterized kinetically from erythroc... more An α-carbonic anhydrase (CA, EC 4.2.1.1) was purified and characterized kinetically from erythrocytes of the sturgeon Acipenser gueldenstaedti, an endangered species. The sturgeon enzyme (AgCA) showed kinetic parameters for the CO 2 hydration reaction comparable with those of the human erythrocytes enzyme hCA II, being a highly active enzyme, whereas its esterase activity with 4-nitrophenyl acetate as substrate was lower. Sulphonamide inhibitors (acetazolamide, sulphanilamide) strongly inhibited AgCA, whereas metal ions (Ag + , Zn 2+ , Cu 2+ and Co 2+) were weak, millimolar inhibitors. Several widely used pesticides (2,4-dichlorophenol, dithiocarbamates, parathion and carbaryl) were also assayed as inhibitors of this enzyme. The dithiocarbamates were low micromolar AgCA inhibitors (IC 50 of 16-18 µM), whereas the other pesticides inhibited the enzyme with IC 50 s in the range of 102-398 µM. The wide use of dithiocarbamate pesticides may be one of the factors enhancing the vulnerability of this sturgeon species to pollutants.

Arı sütü binlerce çiçekten özümsenen nektar ve çiçek polenleri ile bal arılarının enzimlerini de ... more Arı sütü binlerce çiçekten özümsenen nektar ve çiçek polenleri ile bal arılarının enzimlerini de içeren, akıcı jel halinde ve kremsi-beyaz renkteki etkili bir salgıdır. Besin değeri yüksek bu ürün amino asit, peptit, protein ve minerallerden oluşur. Arı sütünün en önemli özelliği diğer arı ürünlerinde bulunmayan kısa zincirli hidroksi yağ asitlerince zengin oluşudur. Antibakteriyel, anti-tümoral, ve immün sistemi uyarıcı özelliğe sahip arı sütünün majör yağ asidi trans-10-hidroksi-2-dekanoik asit (10-HDA)'dır. Yapılan bu çalışma ile farklı yıllarda toplanan arı sütü örneklerinin nem ve 10-HDA miktarı tayin edildi. HPLC-UV ile ölçülen 10-HDA miktarının yıllara göre ortalama %2.1 ile %2.6 arasında değiştiği tespit edildi. Arı sütü nem içeriğinin ise ortalama %62.6 ile %66.5 arasında değiştiği tespit edildi.

In this study, we investigated 17 different phenolic constituents and total antioxidant propertie... more In this study, we investigated 17 different phenolic constituents and total antioxidant properties of cherry laurel, Laurocerasus officinalis Roem (family Rosaceae), locally named karayemis or taflan, a summer fruit highly characteristic of the Black Sea region. Phenolic constituents were measured by reverse phase-high performance liquid chromatography (RP-HPLC). Total phenolic compounds, total flavonoids, ferric reducing /antioxidant power (FRAP), cupric ion reducing capacity (CUPRAC) and 2,2diphenly-1-picrylhydrazyl (DPPH) radical methods were used to evaluate the antioxidant capacity. The total phenolics and total flavonoids were found to be 1.094 g GAE/100 g DW and 0.080 g QUE/100 g DW, respectively. Chlorogenic acid was found, that is, the main phenolic component of the methanolic extract of the fruit.
