Fifty-one Lathyrus sativus L. landraces and one L. clymenum L. landrace collected from Turkey and... more Fifty-one Lathyrus sativus L. landraces and one L. clymenum L. landrace collected from Turkey and one L. sativus cultivar, G?rb?z, were evaluated with ISSR markers in this study to molecular characterization. Three ISSR primers were used and 45 DNA fragment were evaluated, of which 44 bands were polymorphic. The frequencies of scored bands ranged from 0.009 to 0.888 and averaged 0.363. The genotypes MLT04 and NEV02 had high similarity with 0.825 according to pairwise grouping. The furthest pairwise group was G?rb?z and MLT02 with 0.244. The nearest genotype to G?rb?z was CNR03 with 0.577. The pairwise genetic distance between L. clymenum and L. sativus ranged from 0.353 (accession NEV01) to 0.637 (accession DEN04) and pairwise genetic distance to the cultivar G?rb?z was 0.375. Art is one of the most powerful ways of expressing an individual's internal state, expectations a... more Art is one of the most powerful ways of expressing an individual's internal state, expectations and needs with an aesthetic attitude. Different materials can be used during the expression process including colour, line, sound, movement, word, object and so on. Although the materials used in these branches of art are different from each other, the path followed in the application process and the final results are the same "to reveal an aesthetic product". In order for an artistic practice to mature and reach the beautiful one, the practitioner must have the art education discipline and acquire certain knowledge, skills and understanding of the art field concerned. The art of painting, which is one of the plastic arts, provides the ability to search for beauty through lines and colours on two dimensional surfaces, to create compositions and express the individual's self with this composition, also has its own beauty value criteria. From this point of view, it was aimed to determine the opinions of the prospective teachers studying in the Department of Painting Education in the study about the beauty criteria that should be in the picture. In this study, survey method which is one of the quantitative research methods was preferred. As a data collection tool, a five-point Likert-type scale was developed by the researchers, and 25 items were used. 110 prospective teachers from various grades participated in the study. The results of the research showed that the prospective teachers greatly contributed to the beauty criteria that should be in the picture.

Within the present study, it was conducted a genetic diversity analysis using ISSR markers for so... more Within the present study, it was conducted a genetic diversity analysis using ISSR markers for some apple genotypes grown in Ardahan region, Turkey. Total genomic DNA (gDNA) isolation from apple leaves was performed using commercial kits. Five ISSR primers were used to determine the genetic diversity among the genotypes studied. Polymerase Chain Reaction (PCR) was performed with all gDNA samples to produce bands to score. PCR products were run in agarose gel and visualized under UV light. Bands on the gels were scored as “1”, while no bands at the corresponding positions were scored as “0”, to generate the matrix file. Five ISSR primers produced a total of 35 bands, and 20 of them were polymorphic. The polymorphic bands rated approximately 57%. Phylogenetic relationships and genetic distances between the genotypes were calculated by using the PAUP [Phylogenetic Analysis Using Parsimony (and Other Methods)] program. Hayat Bilgisi sadece bir ders olmaktan da öte toplumun sahip olduğu ahlaki ve kültürel alt yapıyı ve bu alt yapıya dahil olan değerleri ve becerileri de bireylere aktarma noktasında önemli bir paya sahiptir. Bu görevler ve sorumluluklar, Hayat Bilgisi dersine farklı ve önemli bir misyon yüklemiştir. Bu misyon çerçevesinde Hayat Bilgisi dersini en iyi yansıtacak öğrenme araçlardan olan Hayat Bilgisi ders kitaplarının da elbette güncel ve işlevsel olması beklenmektedir. Ders kitaplarının güncel hayatı takip eder nitelikte canlı ve dinamik bir alt yapının eseri olması gerektiği söylenebilir. Hayat Bilgisi ders kitaplarında tasarım, dizayn, resimler ile metinler arasındaki uyum da oldukça önemlidir. Çünkü belirlenen belli başlı temalar altında bireylere kazandırılacak olan kazanımlar, değerler ve beceriler uygun etkinlikler, örnekler ile ders kitabında sunulmaktadır. Bu sunum esnasında da kullanılan etkinlikler, örnek olaylar ya da metinlerin, ilgili olarak birlikte kullanıldıkları görseller ile de örtüşmesi beklenmektedir. Ancak bu şekilde etkili bir öğrenme sürecinden bahsedilebilecektir. Bu çalışmada, Türkiye ve Almanya'daki Hayat Bilgisi ders kitaplarındaki resim-metin uyumu incelenmiş olup, ders kitaplarındaki görsel ve metinlerin kullanımları arasındaki benzerlik ve farklılıklar değerlendirilmiştir.

Malpighiales, containing approximately 16000 species, is one of the most diverse and largest angi... more Malpighiales, containing approximately 16000 species, is one of the most diverse and largest angiosprem orders in plants. Growth-regulating factor (GRF) protein is a putative transcription factor and plays a regulatory role during plant growth and development processes such as stem elongation and cell expansion. The latest available protein data provides an opportunity to compare and understand the critical similarities and differentiations of GRF proteins among Malpighiales members. In the present study, using different bioinformatic tools, we conducted domain, two-and three-dimensional (2D, 3D) comparative, physichochemical, subcellular prediction and sequence analysis of 87 putative GRF proteins belonging to 4 genera in Malpighiales. Results showed that QLQ, WRC, TQL, GGPL, RSRKPVE, FDV, and GFG domain motifs are conserved in different ratios between 34-82%. Comparative analysis proved that there is a common triangle-like shape among GRF proteins and the conserved motifs are localized approximately in similar regions of this triangle. The sequence lengths of GRFs changes between 93-639 aa and pH values differ between 5.28 and 10.50. According to subcellular prediction and domain analysis, only 9-11% GRF are localized out of nuclear region. Additionally, it was determined that 87 GRF proteins have monophyletic phylogeny. The avarage values of pairwise distance and nucleotide diversity (π) was found to be 2.78 and 0.727, respectively. These results could make a contribution to understand how structurally similar but functionally diverse GRF proteins execute various functions in different plant parts or in different plant species.

A novel amperometric glucose biosensor was proposed in flow injection analysis (FIA) system using... more A novel amperometric glucose biosensor was proposed in flow injection analysis (FIA) system using glucose oxidase (GOD) and Quantum dot (ZnS-CdS) modified Pencil Graphite Electrode (PGE). After ZnS-CdS film was electrochemically deposited onto PGE surface, GOD was immobilized on the surface of ZnS-CdS/PGE through crosslinking with chitosan (CT). A pair of well-defined reversible redox peak of GOD was observed at GOD/CT/ZnS-CdS/PGE based on enzyme electrode by direct electron transfer between the protein and electrode. Further, obtained GOD/CT/ZnS-CdS/PGE offers a disposable, low cost, selective and sensitive electrochemical biosensing of glucose in FIA system based on the decrease of the electrocatalytic response of the reduced form of GOD to dissolved oxygen. Under optimum conditions (flow rate, 1.3mLmin(-1); transmission tubing length, 10cm; injection volume, 100μL; and constant applied potential, -500mV vs. Ag/AgCl), the proposed method displayed a linear response to glucose in the range of 0.01-1.0mM with detection limit of 3.0µM. The results obtained from this study would provide the basis for further development of the biosensing using PGE based FIA systems.

The results indicated that plant highest varies between 54.6-67.2 cm, technical stem length 29.4-61.7 cm, number of branches 4.4-7.4, number of capsule per plant 25.6-47.1, number of seeds per capsule 6.4-8.1, number of seeds per plant 163.84-381.51, 1000 seeds weight 6.853-9.242 g and seed yield per plant 1.123-3.526 g. On the basis of these results finally, it was decided to continuation of selection by using the well performed lines (such as 1, 4, 5, 7 and 8 lines in WxN combination, 14 and 15 lines in NxA combination, 21 line in BxN combination, 22, 23, 24, 25 and 27 lines in BxS combination, 29 line in BxA combination and 30 line in SxA combination) for breeding suitable linseed varieties due to yield and yield components for Samsun ecological condition.
In this research, a model for predicting the leaf area was developed for flax by using Linda, Ant... more In this research, a model for predicting the leaf area was developed for flax by using Linda, Antares, Bionda, Avangard, Atalanta, Flanders, Dakota, Sari-85, Ayancik and Windemore cultivars by measuring lamina width, length and leaf area without destroying in 2004. Two hundred leaves were collected from each line and an allometric relationship was derived between actual leaf area (ALA) measured using the Placom Digital Planimeter (Sokkisha Planimeter Inc., Model KP-90), leaf length (LL) and leaf width. Multiple regression analysis for the cultivars was performed. The proposed leaf area (LA) prediction model is LA (cm 2 )=-0,7796+0,2678*L+2,2652*W +0,0120*α- 0,0454*α*W, R 2 = 9697.

This regulator protein behaves as a transition state regulator and governs many adaptations of Bacillus subtilis cells during the transition from exponential growth to stationary phase. In this study, we conducted a comparative in silico analysis considering the primer structure of Bacillus subtilis 168 LutR protein against to the sequences and 3-D conformational situations of the well-known HTH-type transcriptional factors FadR (PDB ID: 1HW1, 1HW2, 1HT9) and YvoA (PDB ID: 2WV0) using PyMOL. As a result, we conclude that the critical residues for DNA-recognition and DNA-binding of LutR are highly conserved and conformationally correspond to the same positions in the three-dimentional structure as those in FadR and YvoA. (pale flax) genotypes which were collected from 34 locations of 16 different Turkey's provinces at the University of Ondokuz Mayıs, Faculty of Agriculture, Department of Field Crops. Results of the research indicated that; the percentage of palmitic acid was changed between 5.47-8.58%, the percentage of stearic acid was changed between 0.00-6.51%; the percentage of oleic acid was changed 12.72-37.13%; the percentage of linoleic acid was changed between 5.22-13.25% and the percentage of linolenic acid was changed between 42.4-70.85%. Based on these results the population, were studied, cannot be used in order to improve linseed species have edible oil quality, however the genotypes of, have 0.00% linolenik acid, Kastamonu (12, 13), Karabük (14) and Bolu (18, 19) can be used in order to reduce saturated fatty acid content in linseed.

Monoclonal anti-collagen type II antibody CIIC1 is an arthritogenic autoantibody, which induces a... more Monoclonal anti-collagen type II antibody CIIC1 is an arthritogenic autoantibody, which induces arthritis in mice. We crystallized and solved the structure of CIIC1 Fab molecule. Analysis of structure revealed an interaction between the CDR regions of one Fab to the CH1 domain of another Fab, which resembles an antibody-antigen interaction. ELISA experiments confirmed the cross-reactivity of both the full CIIC1 antibody and a single chain Fv fragment to other anti-collagen antibodies which are of different isotypes and epitope specificity. The rheumatoid factor like reactivity of CIIC1 antibody together with its collagen type II specificity may explain the pathogenicity of this antibody.

Introduction: Antibodies towards type II collagen (CII) are detected in patients with rheumatoid ... more Introduction: Antibodies towards type II collagen (CII) are detected in patients with rheumatoid arthritis (RA) and in non-human primates and rodents with collagen induced arthritis (CIA). We have previously shown that antibodies specific for several CII-epitopes are pathogenic using monoclonal antibodies from arthritic mice, although the role of different anti-CII epitopes has not been investigated in detail in other species. We therefore performed an inter-species comparative study of the autoantibody response to CII in patients with RA versus monkeys and mice with CIA. Methods: Analysis of the full epitope repertoire along the disease course of CIA was performed using a library of CII triple-helical peptides. The antibody responses to the major CII epitopes were analyzed in sera and synovial fluid from RA patients, and in sera from rhesus monkeys (Macaca mulatta), common marmosets (Callithrix jacchus) and mice. Results: Many CII epitopes including the major C1, U1, and J1 were associated with established CIA and arginine residues played an important role in the anti-CII antibody interactions. The major epitopes were also recognized in RA patients, both in sera and even more pronounced in synovial fluid: 77% of the patients had antibodies to the U1 epitope. The anti-CII immune response was not restricted to the anti-citrulline protein antibodies (ACPA) positive RA group. Conclusion: CII conformational dependent antibody responses are common in RA and are likely to originate from rheumatoid joints but did not show a correlation with ACPA response. Importantly, the fine specificity of the anti-CII response is similar with CIA in monkeys and rodents where the recognized epitopes are conserved and have a major pathogenic role. Thus, anti-CII antibodies may both contribute to, as well as be the consequence of, local joint inflammation.

The paper deals with the measurement of the dynamic stability derivatives of a generic combat air... more The paper deals with the measurement of the dynamic stability derivatives of a generic combat aircraft model undergoing forced oscillation motion in roll. The experiments are performed in the 8' x 10' low-speed, closed circuit Ankara Wind Tunnel (AWT). The model used for measurements is the so called "Standard Dynamic Model", which is used for wind-tunnel tests in various NATO countries. The model was designed and manufactured within the scope of a NATO-AGARD Working Group (WG16) project. The aerodynamic loads acting on the model are measured by means of a five-component internal strain gauge balance. The elaboration of the signal output of the balance is analyzed, according to the small-amplitude direct forced oscillation technique. This technique is a reliable method used to obtain basic dynamic stability derivatives without solving the equations of motion. The paper presents the test rig used to perform the oscillatory tests in the roll plane, the motion control and the data acquisition units that are interfaced to the test rig to measure the aerodynamic loads. A brief review of the forced oscillation technique is also given and the results of the measurements are briefly discussed.

This regulator protein behaves as a transition state regulator and governs many adaptations of Bacillus subtilis cells during the transition from exponential growth to stationary phase. In this study, we conducted a comparative in silico analysis considering the primer structure of Bacillus subtilis 168 LutR protein against to the sequences and 3-D conformational situations of the well-known HTH-type transcriptional factors FadR (PDB ID: 1HW1, 1HW2, 1HT9) and YvoA (PDB ID: 2WV0) using PyMOL. As a result, we conclude that the critical residues for DNA-recognition and DNA-binding of LutR are highly conserved and conformationally correspond to the same positions in the three-dimentional structure as those in FadR and YvoA. (pale flax) genotypes which were collected from 34 locations of 16 different Turkey's provinces at the University of Ondokuz Mayıs, Faculty of Agriculture, Department of Field Crops. Results of the research indicated that; the percentage of palmitic acid was changed between 5.47-8.58%, the percentage of stearic acid was changed between 0.00-6.51%; the percentage of oleic acid was changed 12.72-37.13%; the percentage of linoleic acid was changed between 5.22-13.25% and the percentage of linolenic acid was changed between 42.4-70.85%. Based on these results the population, were studied, cannot be used in order to improve linseed species have edible oil quality, however the genotypes of, have 0.00% linolenik acid, Kastamonu (12, 13), Karabük (14) and Bolu (18, 19) can be used in order to reduce saturated fatty acid content in linseed.

Monoclonal anti-collagen type II antibody CIIC1 is an arthritogenic autoantibody, which induces a... more Monoclonal anti-collagen type II antibody CIIC1 is an arthritogenic autoantibody, which induces arthritis in mice. We crystallized and solved the structure of CIIC1 Fab molecule. Analysis of structure revealed an interaction between the CDR regions of one Fab to the CH1 domain of another Fab, which resembles an antibody-antigen interaction. ELISA experiments confirmed the cross-reactivity of both the full CIIC1 antibody and a single chain Fv fragment to other anti-collagen antibodies which are of different isotypes and epitope specificity. The rheumatoid factor like reactivity of CIIC1 antibody together with its collagen type II specificity may explain the pathogenicity of this antibody.

Introduction: Antibodies towards type II collagen (CII) are detected in patients with rheumatoid ... more Introduction: Antibodies towards type II collagen (CII) are detected in patients with rheumatoid arthritis (RA) and in non-human primates and rodents with collagen induced arthritis (CIA). We have previously shown that antibodies specific for several CII-epitopes are pathogenic using monoclonal antibodies from arthritic mice, although the role of different anti-CII epitopes has not been investigated in detail in other species. We therefore performed an inter-species comparative study of the autoantibody response to CII in patients with RA versus monkeys and mice with CIA. Methods: Analysis of the full epitope repertoire along the disease course of CIA was performed using a library of CII triple-helical peptides. The antibody responses to the major CII epitopes were analyzed in sera and synovial fluid from RA patients, and in sera from rhesus monkeys (Macaca mulatta), common marmosets (Callithrix jacchus) and mice. Results: Many CII epitopes including the major C1, U1, and J1 were associated with established CIA and arginine residues played an important role in the anti-CII antibody interactions. The major epitopes were also recognized in RA patients, both in sera and even more pronounced in synovial fluid: 77% of the patients had antibodies to the U1 epitope. The anti-CII immune response was not restricted to the anti-citrulline protein antibodies (ACPA) positive RA group. Conclusion: CII conformational dependent antibody responses are common in RA and are likely to originate from rheumatoid joints but did not show a correlation with ACPA response. Importantly, the fine specificity of the anti-CII response is similar with CIA in monkeys and rodents where the recognized epitopes are conserved and have a major pathogenic role. Thus, anti-CII antibodies may both contribute to, as well as be the consequence of, local joint inflammation.

The paper deals with the measurement of the dynamic stability derivatives of a generic combat air... more The paper deals with the measurement of the dynamic stability derivatives of a generic combat aircraft model undergoing forced oscillation motion in roll. The experiments are performed in the 8' x 10' low-speed, closed circuit Ankara Wind Tunnel (AWT). The model used for measurements is the so called "Standard Dynamic Model", which is used for wind-tunnel tests in various NATO countries. The model was designed and manufactured within the scope of a NATO-AGARD Working Group (WG16) project. The aerodynamic loads acting on the model are measured by means of a five-component internal strain gauge balance. The elaboration of the signal output of the balance is analyzed, according to the small-amplitude direct forced oscillation technique. This technique is a reliable method used to obtain basic dynamic stability derivatives without solving the equations of motion. The paper presents the test rig used to perform the oscillatory tests in the roll plane, the motion control and the data acquisition units that are interfaced to the test rig to measure the aerodynamic loads. A brief review of the forced oscillation technique is also given and the results of the measurements are briefly discussed.
