Research paper thumbnail of PiRNAs and PIWI proteins as new biomarkers for diagnosis and treatment of liver cancer

Gene Reports, 2021

Abstract Hepatocellular carcinoma (HCC) is one of the most common malignancies and a major cause ... more Abstract Hepatocellular carcinoma (HCC) is one of the most common malignancies and a major cause of cancer-related deaths worldwide due to the high mortality rate and unfavorable satisfaction treatment options available. However, a detailed understanding of the molecular mechanisms underlying liver cancer has not yet been established. piRNAs are RNA molecules with the estimated size of 26–31 nt binding to Piwi proteins. PIWI-interacting RNAs (piRNAs) are small non-coding RNAs (sncRNAs) that are composed of 24-32 nucleotides. These RNAs are responsible for epigenetic regulation and post-transcriptional regulation of gene expression. PIWI-piRNA pathway is the best characteristic of germ cells that have recently been identified in somatic tissues, including stem cells and cancer cells. Several studies have shown that this pathway plays an important role in individual cellular processes. Evidence is shown that abnormal piRNA expression is an important factor involved in cancer cell proliferation, apoptosis, invasion and migration in vitro and in vivo. Altered expression of piRNA in cancer is associated with clinical outcomes, highlighting the important role that they may serve as new diagnostic and prognostic biomarkers and as therapeutic targets for cancer treatment. Therefore, we have focused on PIWI proteins and piRNA functions and their effects on the progression of liver cancer.

Methods: The following databases were search up to March 2019: MEDLINE, EMBASE, Web of Science, and Cochrane Central Register of Controlled Trials. The quality of the relevant extracted data was assessed according to the Cochrane risk of bias tool. Data were pooled by the use of the inverse variance method and expressed as mean difference with 95% Confidence Intervals (95% CI). Results: Eleven effect sizes from nine studies were included in the final analyses. A pooled analysis of 9 effect sizes showed a significant reduction in Beck Depression Inventory (BDI) score following supplementation with vitamin D [weighted mean difference (WMD):-3.91; 95% CI:-5.15-2.66), I 2 = 85.9%]. Combining data from two available studies on the effects of vitamin D supplementation on Pittsburgh Sleep Quality Index (PSQI) also revealed a significant reduction in this score following the intervention (WMD:-1.78; 95% CI:-2.28,-1.28). In addition, there were significant increase in glutathione (GSH) through 3 studies (WMD: 180.70; 95% CI: 6.76, 354.64), and in total antioxidant capacity (TAC) through 3 studies (WMD: 90.09; 95% CI: 56.36, 123.82) after vitamin D supplementation. Combining data from five studies, we found a significant reduction in C-reactive protein (CRP) concentrations after vitamin D supplementation (WMD:-1.74; 95% CI:-2.82,-0.66). Conclusions: Overall, the current meta-analysis demonstrated that taking vitamin D supplements among patients with psychiatric disorders had beneficial effects on BDI, PSQI, GSH, TAC and CRP levels, but did not affect other biomarkers of inflammation and oxidative stress.

This purpose of this study was to compare of myo-inositol and metformin on mental health parameters and biomarkers of oxidative stress in subjects with PCOS. Methods: This randomized controlled trial was conducted among 60 subjects diagnosed with PCOS according to the Rotterdam criteria. Subjects were randomly assigned into two groups to intake either myo-inositol (n = 30) or metformin (n = 30) for 12 weeks. Parameters of mental health were recorded at baseline and after the 12-week intervention. Fasting blood samples were obtained at baseline and the end of the study to determine biomarkers of biomarkers of oxidative stress. Results: After the 12-week intervention, changes in beck depression inventory total score (−1.0 ± 1.7 vs. −0.3 ± 0.7, p = 0.03), general health questionnaire scores (−1.7 ± 2.9 vs. −0.5 ± 1.2, p = 0.02), depression anxiety and stress scale scores (−3.9 ± 6.4 vs. −0.9 ± 1.9, p = 0.01) and plasma total antioxidant capacity (TAC) concentrations (+106.1 ± 69.6 vs. +2.1 ± 132.4 mmol/L, p < 0.001) in the myo-inositol group were significantly different from the changes in these indicators in the metformin group. Myo-inositol supplementation for 12 weeks among patients with PCOS did not affect plasma glutathione and malondialdehyde levels. Conclusions: Overall, our data supported that myo-inositol supplementation for 12 weeks among patients with PCOS had favorable effects on parameters of mental health and plasma TAC levels.
