In gallbladder cancer (GBC), SERPINB5's aberrant expression is reported but its role in genetic predisposition is not known. We enrolled 270 cases and 296 controls and genotyped them for single nucleotide polymorphisms (SNPs) using direct DNA sequencing, followed by genotype-phenotype analysis in GBC and other cancer cell lines. Luciferase assay was done to determine the role of rs2289521 SNP on expression regulation. We found that two SERPINB5 variants rs2289519 and rs2289521 are significantly associated with GBC and contribute to genetic predisposition. The TT genotype of variant rs2289519 was found to be significantly associated (p = 0.008) with GBC in a recessive model. C allele of rs2289521 increased the risk for GBC significantly at genotypic (CT, p = 0.026) and allelic (p = 0.04) levels. In silico analysis and luciferase assay uncovered the probable regulatory role of the rs2289521 variant on expression. Genotype-phenotype correlation in GBC and breast cancer cell lines showed reduced expression of SERPINB5 in the presence of C allele that was consistent with the result of luciferase assay. Overall, our study reveals the genetic association of two SERPINB5 variants with GBC and rs2289521's possible role in the regulation of expression.

The aberrant expression of SERPINB5 is reported in many cancers along with GBC but limited information is available about its role in genetic predisposition for GBC. We carried out case-control study in 206 cases and 219 controls. Promoter SNPs were genotyped by Sanger's sequencing. In-silico promoter analysis and luciferase reporter assay were done to elucidate the role of promoter variants in regulation of SERPINB5 expression. Out of four SNPs, three SERPINB5 promoter variants showed association with GBC in different models. The 'C' allele of variant rs17071138 was found to be significantly associated with GBC (p = 0.017). The 'T' allele of rs3744940 significantly increased the risk for GBC in dominant (p = 0.035) and additive models (p = 0.005). Also, rs3744941 'T' allele increased the risk for GBC by dominant (p = 0.042) as well as additive models (p = 0.016). In-silico promoter analysis and luciferase reporter assay revealed the probable regulatory role of the SERPINB5 promoter variant rs17071138 on the expression. Overall, our study reveals the genetic association of SERPINB5 promoter variants with GBC and possible role of rs17071138 in the regulation of expression.

Gallbladder cancer (GBC) is relatively rare but shows high frequency in certain geographical regi... more Gallbladder cancer (GBC) is relatively rare but shows high frequency in certain geographical regions and ethnic groups, which include Northern and Eastern states of India. Previous studies in India have indicated the possible role of genetic predisposition in GBC pathogenesis. Although matrix metalloproteinase-14 (MMP14) is known modulator of tumour microenvironment and tumorigenesis and TCGA data also suggests its upregulation yet, its role in genetic predisposition for GBC is completely unknown. We explored MMP14 promoter genetic variants as risk factors and their implication in expression modulation and the pathogenesis of GBC. We genotyped all single nucleotide polymorphisms of MMP14 promoter by Sanger's sequencing in approximately 300 GBC and 300 control study subjects of Indian ethnicity and, in 26 GBC tissue samples. Protein expression of MMP14 in GBC tissue samples was checked by immunohistochemistry. In vitro luciferase reporter assay was carried out to elucidate role of promoter genetic variants on expression levels in two different cell lines. MMP14 promoter variants, rs1003349 (p value = 0.0008) and rs1004030 (p value = 0.0001) were significantly associated with GBC. Luciferase reporter assay showed high expression for risk alleles of both the SNPs. Genotype-phenotype correlation for rs1003349 and rs1004030, in patient sample, confirmed that risk allele carriers had higher expression levels of MMP14; moreover, the correlation pattern matched with genetic association models. Overall, this study unravels the association of MMP14 promoter SNPs with GBC which contribute to pathogenesis by increasing its expression.

Introduction: Domestic violence against women is a global problem and present in every country cu... more Introduction: Domestic violence against women is a global problem and present in every country cutting across the boundaries of culture, class, education, income, ethnicity and age. It has serious consequences on the women's mental, physical, reproductive and sexual health. Materials and Methods: A community based, cross-sectional study was conducted from 200 women of reproductive age (18-49 years) residing in rural field practice area of Adichunchanagiri Institute of Medical Sciences (AIMS), Mandya, Karnataka, with a predesigned, pretested, semi-structured questionnaire, pertaining to the socio-demographic parameters, type, frequency, modes, reasons for domestic violence and legal support seeking behaviour in them. Data was analysed using descriptive and inferential statistics. Results: The prevalence of domestic violence was found to be 39.5%. The common type of domestic violence was psychological violence 59(74.7%), followed by 35(44.3%) had physical violence and 15(18.9%) sexual violence. Insult (84.7%), beating and slapping and pressure for sex 12(80%), were the most common types of violence’s observed respectively. Husband was the 48(60.8%) most common perpetrator of domestic violence. Alcohol addiction of husband was the leading cause for domestic violence. The common reason to stay in violent relationship was for children 55 (69.6%), the common response to violence was tolerating 57(72.2%). Domestic violence significantly associated with factors such as occupational status of the women includes agriculture, business & government service, husband’s educational status such as graduation & primary school education and with their habits of smoking & alcohol consumption. The legal help seeking behaviour was very less 7(8.9%) and most common reason cited was social stigma 22 (27.8%) %). Conclusion: About one third of the women in the reproductive age group were exposing to domestic violence and psychological violence was the most common type. The occupational status of the women, such as agriculture, business & government service, the husband’s, educational status including graduation & primary school education, and alcohol & smoking habits were independent risk factors for developing domestic violence. The legal help seeking behaviour of the women was very low.

Disfunción eréctil; Alteración en andrógenos; Acumulación de adipocitos Resumen Objetivo: Modelos... more Disfunción eréctil; Alteración en andrógenos; Acumulación de adipocitos Resumen Objetivo: Modelos animales han demostrado que existe una asociación entre disfunción eréctil y acumulación de grasa bajo la albugínea del pene, pudiendo provocar fuga venosa y pérdida de rigidez del pene. En este estudio se llevó a cabo una comparación de la histología de los cuerpos cavernosos bajo la albugínea de pacientes con disfunción eréctil refractarios a tratamiento médico sometidos a implante de prótesis de pene, y pacientes con enfermedad de Peyronie, sin disfunción eréctil, sometidos a corporoplastia. Materiales y métodos: Se incluyeron 17 pacientes con disfunción eréctil y 14 pacientes potentes con enfermedad de Peyronie. Se recolectaron muestras de tejido cavernoso bajo la túnica albugínea en cada cirugía, las cuales fueron analizadas por un uropatólogo en búsqueda de adipocitos subalbugíneos. Se llevó a cabo un análisis bivariado para comparar características de ambos grupos. Se calcularon las odds ratio con un modelo multivariado de regresión logística. Un valor de p < 0,05 fue considerado significativo. Resultados: Once pacientes (11/17) en el grupo de disfunción eréctil presentaron adipocitos en la histología, mientras solo un paciente (1/14) lo presentó en el grupo control (p < 0,05). El análisis multivariado mostró una odds ratio de 40,72; IC 95%: 2,28-727,29 (p = 0,012). Conclusiones: Alteraciones en los andrógenos provocan cambios estructurales en el pene, llevando a apoptosis y desdiferenciación de músculo trabecular hacia adipocitos. Este es el primer estudio prospectivo en humanos que muestra una asociación entre grasa subalbugínea y disfunción eréctil. La fuga venosa secundaria a este fenómeno podría influir en la disfunción eréctil, especialmente en pacientes que no responden a tratamiento médico.

The aim of this study is to determine the presence of adipocyte accumulation under penile tunica ... more The aim of this study is to determine the presence of adipocyte accumulation under penile tunica albuginea in a group of refractory erectile dysfunction patients. Nineerectile dysfunction patients (case group) and eleven potent patients with Peyronie's disease (control group) underwent penile prosthesis implantation and curvature correction surgeries, respectively. In both groups, sub-albuginean tissue samples were taken within the operative time. Groups were compared in terms of clinical characteristics, co-morbidities and presence of sub-albuginean adipocyte accumulation. Of the nine patients in the case group, eight presented cavernous fat cell accumulation, while only one patient in the control group presented this finding (p<0.05). A significant association (p<0.05) was found between adipocyte accumulation and erectile dysfunction (OR 35 CI 95% 1.98-1727.62). A similar association with chronic arterial hypertension was also found (OR 20 CI 95% 1.29-1008.46). This is the first human study to report an association between erectile dysfunction and penile sub-albuginean fat accumulation. Metabolic syndrome-related conditions could cause disruption in androgen homeostasis, leading to adipocyte accumulation. Venous leakage secondary to accumulation of fat under tunica albuginea could be an important element in the pathophysiology of erectile dysfunction, especially in metabolic syndrome patients that do not respond to medical therapy.

Un valor de p < 0,05 fue considerado significativo. Resultados: Once pacientes (11/17) en el grupo de disfunción eréctil presentaron adipocitos en la histología, mientras solo un paciente (1/14) lo presentó en el grupo control (p < 0,05). El análisis multivariado mostró una odds ratio de 40,72; IC 95%: 2,28-727,29 (p = 0,012). Conclusiones: Alteraciones en los andrógenos provocan cambios estructurales en el pene, llevando a apoptosis y desdiferenciación de músculo trabecular hacia adipocitos. Este es el primer estudio prospectivo en humanos que muestra una asociación entre grasa subalbugínea y disfunción eréctil. La fuga venosa secundaria a este fenómeno podría influir en la disfunción eréctil, especialmente en pacientes que no responden a tratamiento médico.

The aim of this study is to determine the presence of adipocyte accumulation under penile tunica ... more The aim of this study is to determine the presence of adipocyte accumulation under penile tunica albuginea in a group of refractory erectile dysfunction patients. Nineerectile dysfunction patients (case group) and eleven potent patients with Peyronie's disease (control group) underwent penile prosthesis implantation and curvature correction surgeries, respectively. In both groups, sub-albuginean tissue samples were taken within the operative time. Groups were compared in terms of clinical characteristics, co-morbidities and presence of sub-albuginean adipocyte accumulation. Of the nine patients in the case group, eight presented cavernous fat cell accumulation, while only one patient in the control group presented this finding (p<0.05). A significant association (p<0.05) was found between adipocyte accumulation and erectile dysfunction (OR 35 CI 95% 1.98-1727.62). A similar association with chronic arterial hypertension was also found (OR 20 CI 95% 1.29-1008.46). This is the first human study to report an association between erectile dysfunction and penile sub-albuginean fat accumulation. Metabolic syndrome-related conditions could cause disruption in androgen homeostasis, leading to adipocyte accumulation. Venous leakage secondary to accumulation of fat under tunica albuginea could be an important element in the pathophysiology of erectile dysfunction, especially in metabolic syndrome patients that do not respond to medical therapy.
