The effects of extra virgin olive oil (EVOO) and carbon tetrachloride (CCl4) induced oxidative st... more The effects of extra virgin olive oil (EVOO) and carbon tetrachloride (CCl4) induced oxidative stress in rats were determined by the generation of isoprostanoids. These are known to be robust biomarkers to evaluate nonenzymatic and free radical related oxidation. Other oxidative stress biomarkers such as hydroxyeicosatetraenoic acid products (HETEs) and cholesterol oxidation products (COPs) were also determined. The rodents received a control diet, high-fat diet (20% w/w) composed of extra virgin olive oil (EVOO), corn oil (CO), or lard, and high-fat diets with CCl4 insult throughout the experimental period. The EVOO diet was found to suppress the formation of isoprostanoids and COPs compared to that of the control. EVOO also had a high total phenolic content and antioxidant activity compared to those of CO and lard and may be contributed to by the hydroxytyrosol component conjugated to fatty acids (HT-FA). This is the first study to identify HT-FA in EVOO, and it was 4-fold higher than that of olive oil, whereas none was found in corn oil. Furthermore, the EVOO diet showed reduced liver lipid vesicles in CCl4 treated rats compared to that of the control. However, liver toxicity measurements of AST (aspartate transaminase) and ALT (alanine transaminase) activities showed augmentation with CCl4 treatment but were not alleviated by the diets given. Our findings suggest that EVOO is a daily functional food capable of enhancing the antioxidant system for liver protection; the effect is potentially attributed to the phenolic and lipophenolic (phenol conjugated by fatty acids) content.

The present work reports a new mode of action of the naturally occurring flavanols catechin and e... more The present work reports a new mode of action of the naturally occurring flavanols catechin and epicatechin and their dimers B2 and B5, in the breast cancer T47D cell line, namely, their interaction with membrane androgen receptors. We show that monomeric and dimeric flavanols are complete (B2) or partial displacers of radiolabeled testosterone bound on T47D membranes, with affinities ranging from 1.7 (B5) to 82.2 nM (B2). In addition, they trigger the phosphorylation of the same signaling molecules (FAK, PI3K) as testosterone-BSA, minutes after binding to membrane receptors, leading to actin cytoskeleton polymerization and redistribution, with formation of filopodia and lamellipodia. The PI3K inhibitor wortmannin reverts the effect of polyphenols and testosterone-BSA, providing additional evidence about activation of a similar signaling cascade. Incubation of T47D cells for more than 2 h with polyphenols or testosterone-BSA induces apoptosis, which follows the same time-dependent pattern. We conclude that flavanols (monomers or dimers) are agonists of membrane androgen receptors and could be used as testosterone -protein conjugates for the management of tumors, in which, application of testosterone-BSA induces regression, providing additional data about the mechanism of their antiproliferative action. Based on the network weights obtained from the training set, prediction can be made for the unknown objects. In this paper, discrimination of wines based on 2D NMR spectra is performed using LVQ neural networks with orthogonal signal correction (OSC). OSC has been proposed as a data preprocessing method that removes from X information not correlated to Y. Moreover, the partial least squares discriminant analysis (PLS-DA) method has also been used to treat the same data set. It has been found that the OSC-LVQ neural networks method gives slightly better prediction results than OSC-PLS-DA

The diastereoisomer of the natural series 6 was separated by crystallization, the other one being an oil. This process allowed us to prepare enantiomerically pure (+)-catechin 8 in high yield. The pure isomer in the ent-series 9 could be obtained, following the same scheme of reactions.

Polyphenols from the ethyl acetate extracts of red wine were successfully fractionated using a fo... more Polyphenols from the ethyl acetate extracts of red wine were successfully fractionated using a fourstep process (solvent extraction, ion-exchange column chromatography, centrifugal partition chromatography, and semipreparative HPLC), which resulted in the isolation of 22 compounds belonging to different classes of polyphenols (stilbenes, cinnamic acids, flavonoids). Five of them are red wine constituents reported for the first time. The newly isolated compounds include resveratrol dimers, dihydroflavonols, and a cinnamic derivative.

Both stresses originate from an accumulation of all-trans-retinal (atRAL) and are involved in bisretinoid formation by condensation of atRAL with phosphatidylethanolamine (carbonyl stress) in the photoreceptor and its transformation into lipofuscin bisretinoids (oxidative stress) in the retinal pigment epithelium (RPE). As atRAL and bisretinoid accumulation contribute to RPE and photoreceptor cell death, our goal is to select powerful chemical inhibitors of COS. A series of arylhydrazones derived from various 6,8-diacetoxy-or 6,8-dihydroxy-9-oxo-9H-xanthene ... more A series of arylhydrazones derived from various 6,8-diacetoxy-or 6,8-dihydroxy-9-oxo-9H-xanthene carboxaldehydes were synthesized and evaluated for their in vitro antifungal properties against two human pathogenic yeasts (Candida albicans and C. krusei ) according to a diffusion method. The activity was strongly dependent from the position of the (1-arylhydrazinyl-2-ylidene)methyl chain in the xanthone molecular skeleton. Compounds having the nitrogen side chain in the 4-position, with a further halogen substitution on the terminal phenyl ring showed fungistatic effects. Within this series, the 4-fluorophenylhydrazinyl derivative 13g exhibited the highest activity, particularly against C. krusei, with a greater efficacy than that of econazole, used as reference.

T... more Lipooligosaccharides (LOSs) have recently been proposed as markers of mycobacterial avirulence. They have been characterized in Mycobacterium kansasii cell wall and were investigated in Mycobacterium gastri since the distinction between the two mycobacterial species remains in some question. A set of unknown LOSs was isolated from M. gastri W471. The highly antigenic lipooligosaccharide, LOS-III, was purified and appeared in all the M. gastri strains investigated regardless of their morphology. Moreover, by enzyme-linked immunosorbent assay and chromatographic approaches, it was found that LOS-III unambiguously distinguished M. gastri from the opportunistic pathogen M. kansasii. The LOS-III structure was established from its native form using NMR spectroscopy. This strategy revealed the presence of a supplementary monosaccharide (X) which was not characterized by routine carbohydrate analysis. The contribution of phenols, absorbing above 300 nm, to the photodegradation of lignin was approa... more The contribution of phenols, absorbing above 300 nm, to the photodegradation of lignin was approached by studying a phenolic phenylcoumarone lignin model molecule PCO. The compound was irradiated in solution and adsorbed on filter paper. The isolation and analysis of the photoproducts formed during the irradiation indicate the presente of a catechol structure (involving demethylation of the starting material) and two dimeric stilbene compounds: a ketone anda conjugated quinone methide giving high colouration to the irradiated solution. Those compounds have been detected as well in the solid state. The structure elucidation of the photoproducts was mainly based on mass spectrometry and 2D NMR experiments at 500 MHz. The major role played by the phenol group was supported by studying the O-methylated analogue PCOMe.

Within this series, the 4-fluorophenylhydrazinyl derivative 13g exhibited the highest activity, particularly against C. krusei, with a greater efficacy than that of econazole, used as reference.

T... more Lipooligosaccharides (LOSs) have recently been proposed as markers of mycobacterial avirulence. They have been characterized in Mycobacterium kansasii cell wall and were investigated in Mycobacterium gastri since the distinction between the two mycobacterial species remains in some question. A set of unknown LOSs was isolated from M. gastri W471. The highly antigenic lipooligosaccharide, LOS-III, was purified and appeared in all the M. gastri strains investigated regardless of their morphology. Moreover, by enzyme-linked immunosorbent assay and chromatographic approaches, it was found that LOS-III unambiguously distinguished M. gastri from the opportunistic pathogen M. kansasii. The LOS-III structure was established from its native form using NMR spectroscopy. Leur découverte et la compréhension des mécanismes qui les sous-tendent sont des enjeux considérables pour la santé de demain. En effet, que ce soit pour améliorer la qualité de vie des bien-portants ou pour restaurer celle des malades, le médecin généraliste, le cancérologue, le cardiologue, le pharmacien ou le nutritionniste ont à mener un même combat, le plus efficacement possible, celui qui les oppose aux dégâts causés par des entités dévastatrices que sont les radicaux libres et autres espèces oxygénées. Par leur extrême réactivité, les radicaux libres sont responsables des processus inéluctables du vieillissement mais aussi, de ceux observés dans un grand nombre de pathologies, qui ont toutes comme corollaire une augmentation sensible des désordres du métabolisme oxydatif.

The contribution of phenols, absorbing above 300 nm, to the photodegradation of lignin was approa... more The contribution of phenols, absorbing above 300 nm, to the photodegradation of lignin was approached by studying a phenolic phenylcoumarone lignin model molecule PCO. The compound was irradiated in solution and adsorbed on filter paper. The isolation and analysis of the photoproducts formed during the irradiation indicate the presente of a catechol structure (involving demethylation of the starting material) and two dimeric stilbene compounds: a ketone anda conjugated quinone methide giving high colouration to the irradiated solution. Those compounds have been detected as well in the solid state. The structure elucidation of the photoproducts was mainly based on mass spectrometry and 2D NMR experiments at 500 MHz. The major role played by the phenol group was supported by studying the O-methylated analogue PCOMe.
