Research paper thumbnail of Neuromodulatory effects of hesperidin in mitigating oxidative stress in streptozotocin induced diabetes

BioMed research international, 2014

Recent studies have provided insights on antioxidants and their emergence as potential therapeutic and nutraceutical. The present study examined the hypothesis that hesperidin (HP) ameliorates oxidative stress and may be a limiting factor in the extent of CNS complication following diabetes. To test this hypothesis rats were divided into four groups: control, diabetic, diabetic-HP treated, and vehicle for HP treatment group. Diabetes mellitus was induced by a single injection of STZ (65 mg/kg body weight). Three days after STZ injection, HP was given (50 mg/kg b.wt. orally) once daily for four weeks. The results of the present investigation suggest that the significant elevated levels of oxidative stress markers were observed in STZ-treated animals, whereas significant depletion in the activity of nonenzymatic antioxidants and...

The current study investigated the effect of a vermiwash foliar spray on the response of bhut jolokia (Capsicum assamicum) exposed to two different arbuscular mycorrhizal fungi (AMF: Rhizophagus irregularis, RI and G. mosseae, GM) in acidic soil under naturally ventilated greenhouse conditions. The VW spray significantly influenced the growth of plants receiving the dual treatment of AMF+VW. Plant growth was more prominent in the GM+VW treatment group than that in the RI+VW treatment group. The plant-AMF interactions in relation to growth and nutrient requirements were also significantly influenced by the application of VW. Interestingly, the VW treatment appeared to contribute more N to plants when compared to that under the AMF treatment, which led to changes in the C:N:P stoichiometry in plant shoots. Furthermore, the increased potassium dependency, as observed i...

The purpose of this study was to determine the prevalence of the disease in the catchment area of the three teaching hospitals, to identify the systemic predisposing factors and local aetiological factors, and to assess the recommended role of aggressive surgical debridement as a part of treatment. This prospective study was conducted in the surgical departments of three tertiary care hospitals of the Khyber Medical University, Peshawar, Pakistan, from 1 January 2002 to 30 June 2007. Detailed history and examination of all patients were carried out to reach the diagnosis of Fournier's gangrene. After initial resuscitation, patients were treated aggressively, including surgical debridement. Sixty patients were studied in the study period. The male to female ratio was 5:1. The age range was 20-75 years with mean 47+17.4 (SD) years. The socioeconomic status of patients was poor (the average income was less than 50perweek)in36(6050 per week) in 36 (60%) and 24 (40%) were middle class (the average income being 50perweek)in36(6050-100 per week). Thirty-nine patients (65%) presented in the hot humid months of the year. Extent of the disease was scrotum in 18 patients (30%), perineum in 30 (50%) and abdominal wall in 12 (20%). Systemic predisposing factors identified in our study were diabetes mellitus in 20 (33.33%) patients, chronic alcohol abuse in two (3.33%) and long-standing steroid therapy in four (6.67%) while in 34 patients (56.67%) no cause was identified. The local aetiological origin of Fournier's gangrene was urogenital in 14 (23.33%) patients, anorectal in 10 (16.67%) and cutaneous in 8 (13.33%) and no local pathologies could be identified in 28 (46.67%) patients. The mean time interval between first symptom and initial treatment was 2.5 days with a range of 1-7 days. Number of debridement sessions per patient was 2-6 (mean, 3.15). Mean hospital stay was 31+7 (SD) with a range of 10-50 days. Morbidity was 80%. Four patients (6.67%) died. Three of these patients presented to hospital 5-6 days too late. Fournier's gangrene is not an uncommon disease in South Asia. Systemic predisposition such as diabetes mellitus, long-standing steroid therapy, chronic alcoholism and even the hot humid season can contribute to this dreadful disease. Local causes in the form of urogenital, anorectal and cutaneous disorders may trigger this disease in some patients. Early recognition of disease and aggressive surgical debridement are the main treatments.

The structures of the isolated compounds were established by means of 1D and 2D NMR spectroscopic ((1)H, (13)C, DEPT, COSY, HSQC and HMBC) and MS spectral analyses. All the fractions and isolated pure compounds were evaluated for antidiabetic activity by determining their inhibitory effects on PTP-1B enzyme as well as glucose uptake stimulation in C2Cl2 myoblasts cells. Compounds 4 and 7 showed significant in vitro PTP-1B inhibitory activity with 69.2 and 66.8% inhibition at 10 μg/ml concentrations respectively.

Their osteogenic effect was evaluated by using validated models including alkaline phosphatase (ALP) assay, mineralization assay and expression of osteogenic genes-bone morphogenetic protein-2 (BMP-2) and osteoblast transcription factor (RUNX2) - in primary calvarial cultures harvested from neonatal mice. Among them, compound 1 at a dose of 1.0 mg/kg body weight exhibited significant osteoprotective effects and did not show uterine estrogenicity at the same dose. Additionally, compound 1 treatment led to improved biomechanical properties as exhibited by increased power, energy and stiffness in femoral bones compared to untreated Ovx animals. Since osteoporotic compression fracture correlates with the mechanical characteristics of trabecular bone, so that it could effec...

Pure indium solder provides best combination of mechanical and thermo-mechanical properties for efficient thermal design. Two indium TIM assembly approaches were investigated: pre-attach approach and preform approach. Pre-attach refers to bonding indium to the lid cavity first, prior to microprocessor lid attach. Preform refers to direct bonding of indium to lid and silicon in a single reflow process. In the pre-attach approach, indium preform is attached to nickel-plated lid without gold metallization with aggressive flux. This type of flux must be cleaned thoroughly prior to microprocessor lid attach to avoid corrosion. After reflow, indium takes the shape of a dome in the lid cavity. This dome-shape indium can cause lid tilt reject during lid attach. Therefore, a follow-up coining step is required to flatten the indium dome prior to lid attach. At the early st...

Toxicology letters, 2009

Propylene oxide (PO) concentrations &... more Propylene oxide (PO) concentrations >or=300 ppm induced cell proliferation and tumors in rat nasal respiratory epithelium (NRE). Cell proliferation was suggested to result from depletion of glutathione (GSH) in NRE. In order to substantiate this hypothesis, cell proliferation - measured by bromodeoxyuridine incorporation into DNA of the epithelium lining middle septum, dorsal medial meatus, and medial and lateral surfaces of the nasoturbinate in transverse nasal sections taken immediately posterior to the upper incisor teeth - and water-soluble non-protein thiol (NPSH) in NRE were determined after exposing male Fischer 344 rats to 50 ppm, 100 ppm, 200 ppm, or 300 ppm PO (6 h/day, 3 days). Both parameters were also investigated after treating rats for 3 days with diethylmaleate (DEM; 2 x 250 mg/kg/day or 500 + 150 mg/kg/day) or buthionine sulfoximine (BSO; 500 mg/kg/day). Exposure to 50 ppm PO and treatment with 2 x2 50 mg/kg/day DEM resulted in NPSH levels approximating 50% and 80% of the level in untreated controls, respectively. Cell proliferation did not increase. After exposures to >or= 100 ppm PO or treatment with BSO or 500 + 150 mg/kg/day DEM, NPSH was depleted to <or=1/3 of the control level and cell proliferation increased 2.0-3.7-fold the control value. In conclusion, profound perturbation of the GSH status may represent a crucial step in PO induced rat nasal tumorigenicity.
