The second-largest cause of losing quality in vegetables and fruits (VFs) is enzymatic browning (... more The second-largest cause of losing quality in vegetables and fruits (VFs) is enzymatic browning (EB). As a result of polyphenol oxidases (PPOs), EB leads to the color performance of vegetable and fruit yields. To inhibit the activity of PPOs, chemical and physical methods have been developed and several synthetic chemical compounds are widely used as Anti-Browning Agents (ABA) in VFs products. In recent times, emphasis was placed on customer-oriented food manufacturing innovations. Customers have a tendency to encourage the usage of PPO inhibitors that are natural and environmentally friendly. The aim of present review is to illustrate the underlying mechanisms of PPO chemical inhibitor anti browning action and current trends in some of these inhibitor studies. Studies developed over the last decade have been reported and discussed, such as Natural, chemical, physical, controlled atmosphere and coating techniques to avoid EB. The purpose of this review article will be to assemble and reveal an up-to-date demonstration of browning inhibiting natural and synthetic compounds. The details available in this review could also take to facilitate the ultimate objective of developing new inhibitors of enzymatic browning (plant extracts, natural and synthetic compounds) that are acceptable, healthy and beneficial for the food industry.

Lipoxygenase LOX is a lipolysis enzyme that oxidises unsaturated fatty acids like arachidonic and... more Lipoxygenase LOX is a lipolysis enzyme that oxidises unsaturated fatty acids like arachidonic and linoleic acids to form unhealthy chemicals such as dienes, leukotrienes and malondialdehyde in advanced oxidation, which is harmful to cells. In this study, the polyphenols and saponins compounds were extracted from Castanea sativa using Soxhlet for 3 days. Colourimetric and HPLC techniques were used to identify polyphenols and saponins in the extracts respectively. The effects of these extracts were evaluated on LOX activity, which was purified from the liver of Iraqi Cows (AL-Sharabi Cows) as well as on E. coli and Pseudomonas bacteria resistance. According to the LOX purification procedures, the specific activity increased from 0.001 to 0.03 U/mg with a purification fold of 30 times and a yield percentage of 174. Gallic acid 7.72 mg, Rutin 29.25 mg, Quercetin 27.6 mg, kaempferol 34.42 mg, Apigenin 5.25 mg and Catechin 25.8 mg/100 gm of Castanea fruit was obtained. The inhibitory effe...
Male factor infertility contributes partially and solely to the problem of childlessness in aroun... more Male factor infertility contributes partially and solely to the problem of childlessness in around 50% of the cases. Unfortunately, 30 -50% of the etiologies of male infertility are unknown and therefore, no specific therapy can be instituted. Sperm count and sperm motility are prime parameters that determine the functional ability of spermatozoa.This study included determination of lactate dehydrogenase-C(LDH) activity in male infertility (n=23)and its relationship with the different parameters of the seminogram. Also deals with partial purification lactate dehydrogenasefrom semen plasma of male infertility patients by dialysis, and ion exchange DEAEcellulose techniques. The research includes the determination of Glutathione peroxidase activity and its relationship w... more The research includes the determination of Glutathione peroxidase activity and its relationship with some biochemical parameters in serum of patients of diabetes type II. The activity Glutathione peroxidase, Glutathiones-transferase and the level of Glutathione are significantly decreased (p<0.05). whereas the activity of Superoxide dismutase, Polyamine oxidase and the levels of glucose, Glycocylated hemoglobin, Malondialdehyde, insulin hormone and insulin hormone resistance are increased significantly in patients compared to that of healthy subjects. A positive correlation is found between Glutathione peroxidase with Glutathione-s-transferase (r=0.417), Superoxide dismutase (r=0.644) and Glutathione (r=0.521), whereas a significant negative correlation is found between Glutathione peroxidase with Malondialdehyde (r= -0.452). This research was included partially purification of lipoxygenase (LOX) from serum of patients wi... more This research was included partially purification of lipoxygenase (LOX) from serum of patients with epilepsy using ammonium sulfate precipitation, dialysis and ion-exchange chromatography with specific activity (0.751 U / mg), (0.935 U/mg) and (2.60 U/mg) respectively. Optimum conditions for enzyme activity were determined. The best enzyme activity was showed at 7 minutes of incubation time, 3 minutes of reaction time, pH = 7, temperature 40Cº and finally substrate concentration (linoleic acid) was (1.2 mmol/L). By applying Michaelis-Menten and Lineweaver-Burk Equations, Michaelis-Menten constant (km) and Vmax values were found 0.3 (mM) and (0.9 U/ml) respectively. Also this study included the effect of some antiepileptic drugs such as valproic acid, carbamazepine and acetozolamide. Acetazolamide showed the highest inhibition of lipooxygenase activity (96.2)٪ , valproic acid(95.6) and Carbamazapine (95.1)٪. Inhibition type was studied and the result showed noncompetitive inhibition by using Lineweaver-Burke plot for all above drugs.

The mechanism of SDH inhibition by using both drugs were noncompetitive. There is no change in Km value 100 mM. Vmax value without inhibitor was 22.175 but 13.634 and 14.615 unit/ml with Glibenclamide and Rosiglitazone respectively. Accordingly, the inhibition constant Ki was calculated and appeared 7mM for both inhibitors above. Finally, we propose that action of these drugs may contribute to decrease patients with type I diabetes complications through inhibition SDH which plays an important role in polyol pathway.
