The analysis of genetic variability at αs1-casein gene in 112 Indian dromedary (Camelus dromedari... more The analysis of genetic variability at αs1-casein gene in 112 Indian dromedary (Camelus dromedarius) was carried out. The 930 bp fragment of exon 5 of αs1 casein gene was successfully amplified by PCR. The nucleotide substitution at g.942G>T SNP (GenBank ID: JF429140) was detected by PCR-RFLP using SmlI restriction enzyme and verified by sequencing. The GT genotype was not observed in Bikaneri and Kachchhi camels but was observed in Jaisalmeri and Mewari camels at the frequency of 0.143 and 0.071, respectively. Though, the frequency of GT genotype was quite low but still it was enough to document the dynamic nature of the locus g.942G>T SNP in Indian dromedary. The present study comprised of 25 blood samples from domestic cats were cultured in duplicates f... more The present study comprised of 25 blood samples from domestic cats were cultured in duplicates for comparing efficacy of two mitogens i.e. Concanvalin A and Lectin. Concanvalin A was found to be significantly superior to Lectin. GTG- banding was used for preparation of domestic cat karyograms. Diploid number of chromosomes in domestic cats were found to 2n = 38 consisting of 18 pairs of autosomes and 1 pair of sex chromosomes. The 18 pairs of autosomes were classified into 5 groups as A,B,C,D,E, and F consisting of autosome pairs as Large submetacentric 3 pairs, Large subtelocentrics 4 pairs, Large metacentrics 2 pairs, Small submetacentric and subtelocentrics 4 pairs, Small metacentrics2 pairs and Telocentric or acrocentric 2 pairs, respectively. Whereas ‘X’ chromosome was found to be medium sized subtelomeric and ‘Y’ chromosome was small areocentric. No major significant abnormalities were reported in the present investigation.

The present study was undertaken to detect polymorphism at exon I of the prolactin locus using po... more The present study was undertaken to detect polymorphism at exon I of the prolactin locus using polymerase chain reaction -restriction fragment length polymorphism (PCR-RFLP) in the Pandharpuri buffalo breed. DNA from 50 Pandharpuri buffaloes was extracted by the phenol -chloroform method. A pair of bovine specific primers (forward 5'-ATTATCTCTCTCAT TTCCTTTTTACA-3' and reverse 5'-ACTCTGC TGTCACTGTCTGTATT-3') were used for amplification of PRL gene exon I. PCR was carried out in a final reaction volume of 25 µl and the reaction mixture was subjected to standard PCR protocol. The PCR product digested with Hae III (RE). Digestion with Hae III revealed an intact product of 857 bp. The results of PCR-RFLP using Hae III show a different band pattern from cattle and previously observed studies in Jaffarabadi buffaloes. The POU1F1 gene has been identified as one of the major gene for productivity, and its polymorphi... more The POU1F1 gene has been identified as one of the major gene for productivity, and its polymorphism is known to be associated with important production traits of various livestock. The present study was designed to investigate POU1F1 (exon-3) gene polymorphism and its association with body weight in Osmanabadi goat (n=217). The PCRSSCP and DNA sequencing revealed single ‘C’ > ‘T’ transition at nucleotide position 42 in the third exonic region (225 bp) POU1F1 gene of Osmanabadi goat. Genotyping revealed ‘TT’ and ‘CC’ genotypes with predominance of ‘C’ allele in the Osmanabadi goat population. Though no association between the said SNP and body weight was observed, these findings can be extended to find the genetic variability of POU1F1 gene associated with growth traits of various Indian goat breeds.

A1 and A2 are supposed to be related with human health. Hence, to know the status of breeding bulls used for AI services were screened for A1 and A2 variants. Total 47 blood samples of crossbred breeding bulls from Frozen Semen Laboratory, Pune and Aurangabad were used for present work. Methods: The extraction of genomic DNA by phenol-chloroform method were carried out from the blood samples of 47 breeding bulls. Further, DNA based PCR-RFLP method was used to identify the A1, A2 variants. Pure DNA fragments of b-casein (CSN 2) gene were amplified by PCR using suitable pairs of primer to amplify 121 bp and 244 bp products. The primer used to amplify 244 bp was allele specific. The 121 bp fragment then subjected to restriction enzyme digestion. The restriction enzyme digestions resulted into two types of band pattern. Identification of superior germplasm and incorporating them into breeding programme is the need of hour for augmenting productivity of Osmanabadi as well as other indigenous goat breeds. Hence, the present research was aimed to study Pituitary Specific Transcription Factor I (POU1F1) gene polymorphism and its associations with body weight of Osmanabadi goat. Methods: Genomic DNA was extracted from 217 blood samples of randomly selected Osmanabadi goats. The 450 bp fragment of POU1F1 gene encompassing exon-6 and its flanking region (3’UTR) was PCR amplified. The worldwide studies on various livestock species have identified various point mutations in Leptin gene, which are found to be influencing important economic traits including growth rate. The main objective of present study was to determine Leptin gene polymorphism and its association with body weights in Madgyal sheep using PCR-SSCP technique. PCR-SSCP is a reproducible, rapid and quite simple method for the detection of mutations through electrophoretic mobility differences in PCR amplified DNA. We have successfully amplified 180 bp and 275bp amplicons representing exon 1/2 and exon 3 of LEP gene, respectively. The PCR-SSCP analysis of these amplicons revealed a uniform band pattern in all 100 Madgyal sheep. The occurrence of monomorphic nature of Leptin gene in the present study indicates the lack of Leptin gene mutation in Madgyal sheep.
