Materials and Methods In a case-control study, semen samples were collected from infertile and healthy control men. MTHFR promoter region was amplified by methylation-specific polymerase chain reaction (PCR). Finally, the promoter region of MTHFR was analyzed by bioinformatics software. Results Our data revealed significant associations of CpG island promoter methylation with oligozoospermia in a case-control study. In silico analysis showed that promoter contains a strong nucleosome exclusion region, a bonafide CGIs, six PROSITE motifs without a defined TATA box and 14 transcription factor (TF) binding sites, which are directly involved in spermatogenesis Conclusion Based on our findings, methylation of the MTHFR gene promoter region may be a risk factor for oligozoospermia. Post-acrosomal sheath WW domain binding protein (PAWP) is one of the proteins that is expressed d... more Post-acrosomal sheath WW domain binding protein (PAWP) is one of the proteins that is expressed during spermatogenesis process and has several roles in sperm differentiation, fertilization, and early embryonic development. The aim of this study was assessment of the relationship between PAWP expression with fertilization rate in infertile men candidate for intracytoplasmic sperm injection (ICSI). In addition, sperm DNA fragmentation was also assessed in these infertile men. Materials and method: In this descriptive study, semen samples were collected from 54 infertile men, of aged 32.5±4.44 candidate for ICSI referring to Isfahan Fertility and Infertility Center. Sperm parameters, DNA fragmentation, expression of PAWP at RNA, and protein level were assessed by Computer Assisted Semen Analysis, TUNEL assay, real time PCR, and western blot, respectively. Data was analyzed by SPSS (version 16) using Independent t-test and Pearson Correlation. P-values <0.05 were considered statistically significant. Resultst: Significant correlations were observed between expression of PAWP at RNA and protein level with fertilization rate. When infertile men were categorized according to the percentage of fertilization lower and higher than 50%, the expression of PAWP was significantly lower and percentage of DNA fragmentation was significantly higher In the first group,in infertile men with fertilization rate lower than 50% compared to with the other group. (p<0.05). Conclusion: Normal expression of sperm PAWP at gene and protein levels, and also low percentage of sperm DNA fragmentation may be considered as normal spermatogenesis processes in men.

AimTo investigate if there is an association between M235T polymorphism of angiotensinogen gene a... more AimTo investigate if there is an association between M235T polymorphism of angiotensinogen gene and myocardial infarction (MI) risk and perform a meta-analysis and an in silico approach.MethodsThis case-control study included 340 participants (155 MI patients and 185 controls) examined at Kashan University of Medical Sciences (Kashan, Iran) between 2013 and 2015. Meta-analysis included 25 studies with 6334 MI patients and 6711 controls. Bioinformatics tools were applied to evaluate the impact of M235T polymorphism on angiotensinogen function and structure.ResultsGenetic association study revealed a significant association between TT genotype (odds ratio [OR] 2.08, 95% confidence interval [CI] 1.08-4.00, P = 0.029) and T allele (OR 1.45, 95% CI 1.06-1.99, P = 0.021) and MI risk. Meta-analysis also revealed a significant association between M235T polymorphism and MI risk in allelic (OR 1.55, 95% CI 1.10-2.18, P = 0.012) and recessive (OR 1.69, 95% CI 1.13-2.53, P = 0.010) models within Asian population. In silico-analysis revealed that M235T fundamentally changed the function of angiotensinogen (score 32; expected accuracy 66%).ConclusionsOur study suggests that M235T polymorphism might be a helpful biomarker for screening of susceptible individuals for MI in Asian population.
