It has been used around the world as a spice, flavoring agent, and ingredient in traditional medicine. Ginger essential oils (GEOs) are derivatives of ginger that can be found in various products used in daily life, such as food, pharmaceutical, and cosmetics. The present study analyzed the chemical compositions, antioxidant, and antibacterial activities of three commercially available GEOs. The compositions of GEOs were identified using the gas chromatography method. The antioxidant activity was evaluated using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and 2,2'-azinobis-(3ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay methods. The antibacterial activity was determined using a disc diffusion assay based on the diameter of the inhibition zone (DIZ). The main compounds identified from the samples were zingiberene, α-curcumene, β-sesquiphellandrene, camphene, α-farnesene, β-bisabolene, α-pinene, and 3-carene. The IC values were found to be 5.3023 and 1.4504 mg mL for GEO1; 0.9249 and 0.5276 mg mL 50-1-1 for GEO2; and 10.4463 and 3.3535 mg mL for GEO3 when evaluated using DPPH and ABTS assay methods,-1 respectively. All samples showed antibacterial activity against ATCC 13420 and Staphylococcus aureus Bacillus subtilis (the collection of National Research and Innovation Agency), while only GEO2 and 3 displayed inhibitory effect against ATCC 9637. Antioxidant and anticancer activities of sand sea cucumber Holothuria scabra (dried and fresh) ex... more Antioxidant and anticancer activities of sand sea cucumber Holothuria scabra (dried and fresh) extracts were studied. The highest extraction yield of sea cucumber H. scabra (3.9%) was obtained using dried H. scabra at 60 o C. The highest antioxidant activity was found in fresh H. scabra extract at 60°C with an IC50 value of 629.89 ± 0.15 µg/mL using the ABTS method, and the highest antioxidant activity by DPPH method was found in dried H. scabra extract at 70°C with an IC50 value of 32017.18 ± 0.82 µg/mL. The best antioxidant activity based on FRAP and TBARS methods was found in fresh H. scabra extracts at 80°C, respectively. The highest total phenol and flavonoid contained in dried H. scabra extract were 317.54 ± 8.91 mg GAE/100 g sample and 247.56 ± 11.70 mg QE/100 g sample. H. scabra extracts inhibited more than 50% of the growth of the MDA-MB-231 cell line at concentrations of 25 and 50 μg/mL except for dried H. scabra extracts at 80°C. Similarly, the extracts showed the highest cytotoxic effect up to 100% at the highest concentration (100 μg/mL) except for dried H. scabra extracts at 70°C and 80°C.

Endophytic fungi are widely underexplored in the discovery of novel bioactive compounds with prom... more Endophytic fungi are widely underexplored in the discovery of novel bioactive compounds with promising pharmaceutical uses. Aglaia foveolata, which has important medicinal value, was investigated for its endophytic fungi to produce bioactive compounds by determining antioxidant, antibacterial, and antibiofilm activities. In this study, two endophytic fungi were collected from A. foveolata leaves and identified phenotypically along with genotypically by Internal Transcribed Spacer (ITS) gene sequence and known as Penicillium sp. SAF6-EGY strain AFL.2 and Arthrinium sp. R22-1 strain AFL.3. These fungal cultures were further grown on Yeast Malt (YM) medium and the culture was extracted by ethyl acetate. The ethyl acetate extract of AFL.2 and AFL.3 showed relatively similar in Total Phenolic Content (TPC) and Total Flavonoid Content (TFC) ranging from 114.71 to 119.42 mg GAE/g extract and 74.09 until 76.31 mg QE/g extract for TPC and TFC, respectively. Further, AFL.3 showed the highest antioxidant activity in 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2azino-bis (3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) scavenging at IC 50 value of 123.41 and 256.95 µg/mL, respectively. As for antibacterial activity, AFL.2 exhibited the strongest activity against Eschericia coli (acrB JW0451-2) and Staphylococcus aureus (Newman) with the same MIC value of 250 µg/mL. Meanwhile, AFL.3 showed the best antibacterial activity against Bacillus subtilis (DSM10) and S. aureus with the same MIC value of 500 µg/mL. In addition, AFL.2 showed the best antibiofilm activity against E. coli, while AFL.3 against B. subtilis biofilm with the inhibition value of 42 and 35%, respectively. Liquid Chromatography Quadrupole-Mass Spectrometry Analysis (LC-MS/MS) revealed that both AFL.2 and AFL.3 extract contained several putative antibacterial compounds which are correspond to its promising activity.

In this work, essential oil was extracted from nettle leaves and encapsulated in chitosan nanopar... more In this work, essential oil was extracted from nettle leaves and encapsulated in chitosan nanoparticles by a two-stage of emulsion-ionic gelation method. An acceptable retention rate (59.5 to 68.2%) of nettle essential oil (NEO) loaded in chitosan nanoparticles was achieved. Also, Fourier transform infrared (FTIR) spectroscopy and X-ray diffraction (XRD) also confırmed successful introducing and loading NEO in chitosan. The nanoparticles displayed an average size of 208.3-369.4 nm. NEO-loaded chitosan nanoparticles had greater antioxidant activity than the free form of NEO. Furthermore, the antibacterial activity of NEO-loaded chitosan nanoparticles was high against S. aureus and E. coli (inhibition zone diameter of 4.11-3.95 cm). Based on these results, the encapsulation of EOs in chitosan nanoparticles are promising candidate to be used in food and pharmaceutical products for novel applications.

Watercress contains protein, calcium, phosphorus, iron, vitamin A, vitamin E, vitamin C, flavonoids and phenols. Antioxidants are components that can delay or prevent the oxidation of lipids, nucleic acids, or other molecules by inhibiting the initiation or propagation of oxidation chain reactions. The function of antioxidants is to neutralize free radicals, so that the body is protected from various degenerative diseases and helps suppress the aging process. The purpose of this study was to isolate and identify antioxidant compounds in watercress. The studies included maceration and reflux extraction with 96% ethanol, partitioning (with n-hexane and ethyl acetate), antioxidant activity determination, and compound determination. Antioxidant activity test by 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenger method showed that the watercress ethyl acetate extract had the highest antioxidant activity with IC 50 of 67.77 ppm. Fractionation by first column chromatography [SiO 2 ; n-hexane: ethyl acetate (10:1-1:1), CHCl 3 : MeOH (15:1-1:1)] obtained 5 combined fractions (F1-F5) where the results of the antioxidant activity test of fraction 5 (F5) had a great inhibition value compared to other fractions of 61.04%. Furthermore, F5 fractionated second column chromatography [SiO 2 ; n-hexane: ethyl acetate (1:1), CHCl 3 : MeOH (5:1)] obtained 2 combined fractions (F5.1 and F5.2). After two purifications using column chromatography and testing for antioxidant activity, fraction 5.2 had the highest activity with an IC 50 value of 75.65 ppm. Fraction 5.2 contained phenols and/or flavonoids according to phytochemical screening, while UV-Vis, Fourier transform infrared (FTIR) spectrophotometry and gas chromatography-mass spectrometry (GC-MS) spectroscopic results indicated that, the active ingredient in fraction 5.2 is expected to be 2-methoxy-4-vinylphenol.

Scientifically, celery extract has the ability as an antioxidant, antibacterial, anti-breast cancer, and inhibiting ACE enzyme activity as antihypertensive. The use of maceration and ultrasound-assisted extraction (UAE) methods will give different results on the amount and biological activity of the extract. This study aims to determine the biological activity of celery extract using maceration and ultrasound-assisted extraction method. TEAC, FRAP, NO, and DPPH scavenging assays were used to determine in vitro antioxidant activities. Folin-Ciocalteu reagent and aluminium chloride colorimetric assay were used to determine total phenolic and flavonoid contents, respectively. The sulforhodamine B and angiotensin-converting enzyme (ACE) inhibition assays were used to determine anti-breast cancer and antihypertensive activities, respectively. The celery extract from both extraction methods had antioxidants activity, inhibited the growth of MDA-MB-231 breast cancer cell line, and showed as antihypertensive by ACE inhibition. The maceration gave better activities than UAE with significant results in antioxidants, especially in a water solvent. The use of solvents had more effect on antioxidant activity, especially the FRAP and DPPH free radical scavenging method, as well as the TPC and TFC values of the extract compared to the extraction technique. It was found that ethanol solvent was better than water. The maceration provided better activities than ultrasound-assisted extraction in antibreast cancer. However, there was no significant difference in antihypertensives. The UAE extraction method produced higher yields but had lower antioxidant and anticancer activity than a maceration.

Scientifically, garlic extract has the ability as an antioxidant, antibacterial, anticancer, and antihypertensive. The use of different extraction techniques and solvents will give different results on the amount and biological activity of the extract. This study aims to determine the biological activity of garlic extract using two extraction techniques and solvent types. The TEAC, FRAP, NO and DPPH scavenging assays were used to determine in vitro antioxidant activity. Follin-Ciocalteu reagent and aluminium chloride colorimetric assay were used to determine total phenolic (TPC) and flavonoid (TFC) content. In vitro disc diffusion, sulforhodamine B (SRB), and angiotensin converting enzyme (ACE) inhibition assays were used to determine the antibacterial, anticancer, and antihypertensive activities, respectively. The 70% ethanol solvent and maceration method had better antioxidant activity than the aqueous and ultrasonic-assisted extraction (UAE). TPC values were not significantly different between extraction methods and solvents in all treatments, meanwhile aqueous solvent gave better TFC values than 70% ethanol. Garlic extract only had inhibition against B. subtilis bacteria. The anticancer against MDA-MB231 breast cancer cell line and ACE inhibitory activity of 70% ethanol solvent and UAE method was higher than that of aqueous and maceration. It can be concluded that the different solvent and extraction method were affecting on the biological activities of the garlic extract. However, the UAE technique provides a more effective extraction alternative that requires less solvent and a shorter time.

Antioxidants such as phenol and flavonoid compounds are known to counteract these free radicals. Long term use of synthetic antioxidants is known to cause bad effects on the body. Therefore, the necessity to search for natural antioxidants from plants and its endophytic microbes continues with the hope to obtain potential natural antioxidants with minimum side effects. The purpose of this study was to determine the antioxidant activity, total phenolic, and flavonoid contents of the extract of endophytic fungi which were derived from the Bogor-originated turmeric leaves via in vitro isolation. DPPH free radical scavenging method was used to determine in vitro antioxidant activity. The total phenolic and flavonoid contents assays were based on the Follin-Ciocalteu and aluminium chloride reagents, respectively. The extract of endophytic fungi of Bo.Ci.Cl.D1 and Bo.Ci.Cl.D2 showed antioxidant activity with IC50 value at 24.04 and 96.08 mg/L, respectively. Total phenolic content of Bo.Ci.Cl.D1 and Bo.Ci.Cl.D2 extracts were 113.47 and 81.83 mg gallic acid equivalent/g extract respectively. Total flavonoid content of Bo.Cl.D1 and Bo.Ci.Cl.D2 extracts were 41.79 and 38.50 mg quercetin equivalent/g extract, respectively. Based on these assays, it could be concluded that the extract of Bo.Ci.Cl.D1 has better antioxidant activities than Bo.Ci.Cl.D2.

In this study, two endophytic fungi were collected from A. foveolata leaves and identified phenotypically along with genotypically by Internal Transcribed Spacer (ITS) gene sequence and known as Penicillium sp. SAF6-EGY strain AFL.2 and Arthrinium sp. R22-1 strain AFL.3. These fungal cultures were further grown on Yeast Malt (YM) medium and the culture was extracted by ethyl acetate. The ethyl acetate extract of AFL.2 and AFL.3 showed relatively similar in Total Phenolic Content (TPC) and Total Flavonoid Content (TFC) ranging from 114.71 to 119.42 mg GAE/g extract and 74.09 until 76.31 mg QE/g extract for TPC and TFC, respectively. Further, AFL.3 showed the highest antioxidant activity in 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2azino-bis (3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) scavenging at IC 50 value of 123.41 and 256.95 µg/mL, respectively. As for antibacterial activity, AFL.2 exhibited the strongest activity against Eschericia coli (acrB JW0451-2) and Staphylococcus aureus (Newman) with the same MIC value of 250 µg/mL. Meanwhile, AFL.3 showed the best antibacterial activity against Bacillus subtilis (DSM10) and S. aureus with the same MIC value of 500 µg/mL. In addition, AFL.2 showed the best antibiofilm activity against E. coli, while AFL.3 against B. subtilis biofilm with the inhibition value of 42 and 35%, respectively. Liquid Chromatography Quadrupole-Mass Spectrometry Analysis (LC-MS/MS) revealed that both AFL.2 and AFL.3 extract contained several putative antibacterial compounds which are correspond to its promising activity.

An acceptable retention rate (59.5 to 68.2%) of nettle essential oil (NEO) loaded in chitosan nanoparticles was achieved. Also, Fourier transform infrared (FTIR) spectroscopy and X-ray diffraction (XRD) also confırmed successful introducing and loading NEO in chitosan. The nanoparticles displayed an average size of 208.3-369.4 nm. NEO-loaded chitosan nanoparticles had greater antioxidant activity than the free form of NEO. Furthermore, the antibacterial activity of NEO-loaded chitosan nanoparticles was high against S. aureus and E. coli (inhibition zone diameter of 4.11-3.95 cm). Based on these results, the encapsulation of EOs in chitosan nanoparticles are promising candidate to be used in food and pharmaceutical products for novel applications.

Watercress contains protein, calcium, phosphorus, iron, vitamin A, vitamin E, vitamin C, flavonoids and phenols. Antioxidants are components that can delay or prevent the oxidation of lipids, nucleic acids, or other molecules by inhibiting the initiation or propagation of oxidation chain reactions. The function of antioxidants is to neutralize free radicals, so that the body is protected from various degenerative diseases and helps suppress the aging process. The purpose of this study was to isolate and identify antioxidant compounds in watercress. The studies included maceration and reflux extraction with 96% ethanol, partitioning (with n-hexane and ethyl acetate), antioxidant activity determination, and compound determination. Antioxidant activity test by 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenger method showed that the watercress ethyl acetate extract had the highest antioxidant activity with IC 50 of 67.77 ppm. Fractionation by first column chromatography [SiO 2 ; n-hexane: ethyl acetate (10:1-1:1), CHCl 3 : MeOH (15:1-1:1)] obtained 5 combined fractions (F1-F5) where the results of the antioxidant activity test of fraction 5 (F5) had a great inhibition value compared to other fractions of 61.04%. Furthermore, F5 fractionated second column chromatography [SiO 2 ; n-hexane: ethyl acetate (1:1), CHCl 3 : MeOH (5:1)] obtained 2 combined fractions (F5.1 and F5.2). After two purifications using column chromatography and testing for antioxidant activity, fraction 5.2 had the highest activity with an IC 50 value of 75.65 ppm. Fraction 5.2 contained phenols and/or flavonoids according to phytochemical screening, while UV-Vis, Fourier transform infrared (FTIR) spectrophotometry and gas chromatography-mass spectrometry (GC-MS) spectroscopic results indicated that, the active ingredient in fraction 5.2 is expected to be 2-methoxy-4-vinylphenol.

Scientifically, celery extract has the ability as an antioxidant, antibacterial, anti-breast cancer, and inhibiting ACE enzyme activity as antihypertensive. The use of maceration and ultrasound-assisted extraction (UAE) methods will give different results on the amount and biological activity of the extract. This study aims to determine the biological activity of celery extract using maceration and ultrasound-assisted extraction method. TEAC, FRAP, NO, and DPPH scavenging assays were used to determine in vitro antioxidant activities. Folin-Ciocalteu reagent and aluminium chloride colorimetric assay were used to determine total phenolic and flavonoid contents, respectively. The sulforhodamine B and angiotensin-converting enzyme (ACE) inhibition assays were used to determine anti-breast cancer and antihypertensive activities, respectively. The celery extract from both extraction methods had antioxidants activity, inhibited the growth of MDA-MB-231 breast cancer cell line, and showed as antihypertensive by ACE inhibition. The maceration gave better activities than UAE with significant results in antioxidants, especially in a water solvent. The use of solvents had more effect on antioxidant activity, especially the FRAP and DPPH free radical scavenging method, as well as the TPC and TFC values of the extract compared to the extraction technique. It was found that ethanol solvent was better than water. The maceration provided better activities than ultrasound-assisted extraction in antibreast cancer. However, there was no significant difference in antihypertensives. The UAE extraction method produced higher yields but had lower antioxidant and anticancer activity than a maceration.

The highest extraction yield of sea cucumber H. scabra (3.9%) was obtained using dried H. scabra at 60oC. The highest antioxidant activity was found in fresh H. scabra extract at 60°C with an IC50 value of 629.89 ± 0.15 µg/mL using the ABTS method, and the highest antioxidant activity by DPPH method was found in dried H. scabra extract at 70°C with an IC50 value of 32017.18 ± 0.82 µg/mL. The best antioxidant activity based on FRAP and TBARS methods was found in fresh H. scabra extracts at 80°C, respectively. The use of different extraction techniques and solvents will give different results on the amount and biological activity of the extract. This study aims to determine the biological activity of garlic extract using two extraction techniques and solvent types. The TEAC, FRAP, NO and DPPH scavenging assays were used to determine in vitro antioxidant activity. Follin-Ciocalteu reagent and aluminium chloride colorimetric assay were used to determine total phenolic (TPC) and flavonoid (TFC) content. In vitro disc diffusion, sulforhodamine B (SRB), and angiotensin converting enzyme (ACE) inhibition assays were used to determine the antibacterial, anticancer, and antihypertensive activities, respectively. The 70% ethanol solvent and maceration method had better antioxidant activity than the aqueous and ultrasonic-assisted extraction (UAE). TPC values were not significantly different between extraction methods and solvents in all treatments, meanwhile aqueous solvent gave better TFC values than 70% ethanol. Garlic extract only had inhibition against B. subtilis bacteria. The anticancer against MDA-MB231 breast cancer cell line and ACE inhibitory activity of 70% ethanol solvent and UAE method was higher than that of aqueous and maceration. It can be concluded that the different solvent and extraction method were affecting on the biological activities of the garlic extract. However, the UAE technique provides a more effective extraction alternative that requires less solvent and a shorter time.

Antioxidants such as phenol and flavonoid compounds are known to counteract these free radicals. Long term use of synthetic antioxidants is known to cause bad effects on the body. Therefore, the necessity to search for natural antioxidants from plants and its endophytic microbes continues with the hope to obtain potential natural antioxidants with minimum side effects. The purpose of this study was to determine the antioxidant activity, total phenolic, and flavonoid contents of the extract of endophytic fungi which were derived from the Bogor-originated turmeric leaves via in vitro isolation. DPPH free radical scavenging method was used to determine in vitro antioxidant activity. The total phenolic and flavonoid contents assays were based on the Follin-Ciocalteu and aluminium chloride reagents, respectively. The extract of endophytic fungi of Bo.Ci.Cl.D1 and Bo.Ci.Cl.D2 showed antioxidant activity with IC50 value at 24.04 and 96.08 mg/L, respectively. Total phenolic content of Bo.Ci.Cl.D1 and Bo.Ci.Cl.D2 extracts were 113.47 and 81.83 mg gallic acid equivalent/g extract respectively. Total flavonoid content of Bo.Cl.D1 and Bo.Ci.Cl.D2 extracts were 41.79 and 38.50 mg quercetin equivalent/g extract, respectively. Based on these assays, it could be concluded that the extract of Bo.Ci.Cl.D1 has better antioxidant activities than Bo.Ci.Cl.D2.
