Until recently grape cultivars were identified based entirely on ampelographic criteria. Currentl... more Until recently grape cultivars were identified based entirely on ampelographic criteria. Currently a number of molecular techniques based on PCR can be applied for exhaustive genetic analysis on plants. These methods proceed through the direct analysis of DNA, in this way avoiding the interference due to environmental interactions. In this paper the application of RAPD and AFLP for the analysis of genetic diversity in a group of more than 50 grape cultivars is presented. Using 18 RAPD primers, 103 information bands were identified, corresponding to 29.9% of polymorphism. In comparison, using only four AFLP primers produced 86 information bands, with a very similar percentage of polymorphism (29.6%). Both methodologies exhibited an adequate degree of reproducibility, evaluated on a collection of 48 Cabernet Sauvignon clones. The dendrograms based on these data sets graphically depicted the ability of both methods to differentiate all the cultivars studied. In the case of Cabernet Sauvignon clones, AFLP permitted the identification of a number of differences that could be related to morphological differences detected among these clones, such as cluster length. The comparison of both methods, principally related to their ability to differentiate cultivars and clones, suggests that AFLP is the more adequate method for both purposes. Nevertheless, both methods could be implemented, given that under controlled conditions appropriate levels of polymorphism and reproducibility are displayed.

Thirty-one apricot cultivars were evaluated, including one Japanese plum and one apricot x plum hybrid as outgroup. Eleven primer pairs developed in peach were assayed. Seven primer pairs (64% of the assayed markers) were amplified successfully and resulted in 44 polymorphic bands, ranging from 2 (UDP96008) to 8 (UDP98406 and UDP98411) alleles. The observed heterozygosity ranged from 0.09 (UDP96008) to 0.88 (UDP96003). There is a high degree of homology for the SSR loci between peach and apricot and transportability of these markers among Prunus. According to genetic distances obtained within the apricot group, four clusters were identified: Western European;, North American; one cluster including 'Bebeco', 'Tirynthos' and 'Ninfa' and finally another cluster including other North American cultivars. (Lamiaceae) are used for centuries in traditional medicine of Balcanic people in the healing of the lymphatic nodule and respiratory system inflammation. In this paper the amount of total phenols and flavonoids (analyzed by UV/Vis spectrophotometry), phenolic compounds profile (analyzed by HPLC), antimicrobial and antioxidant activities were studied in samples collected in seven per species populations of S. montana and S. subspicata in Croatia. Eight phenolic compounds (rutin, quercetin, caffeic, p-coumaric, ellagic, protocatehuic, rosmarinic, and syringic acid) were identified and quantified using HPLC in methanolic and ethanolic extracts. Results showed that both species contained polyphenolics and other antioxidant compounds with chelating and radical-scavenging properties. The extracts prepared from both species showed broad spectrum of antimicrobial activity on in vitro tested microbial species (Staphylococcus aureus, Escherichia coli, Candida albicans, C. dubliniensis, C. krusei, C. glabrata, C. parapsilosis, and Microsporum gypseum).

Currently a number of molecular techniques based on PCR can be applied for exhaustive genetic analysis on plants. These methods proceed through the direct analysis of DNA, in this way avoiding the interference due to environmental interactions. In this paper the application of RAPD and AFLP for the analysis of genetic diversity in a group of more than 50 grape cultivars is presented.
