Chemical and mineralogical analyses were performed on spent bed residues and cyclone ashes acquir... more Chemical and mineralogical analyses were performed on spent bed residues and cyclone ashes acquired from the TIDD pressurized fluidized bed combustion (PFBC) demonstration plant operated by American Electric Power in Brilliant, OH. The cyclone ashes were composed of fly ash, dolomite, anhydrite, periclase, and calcite in decreasing order of abundance. By comparison, bed residues contained less dolomite and fly ash but more anhydrite, calcite and periclase. All samples were highly alkaline with paste pH values ranging from 9.9 to 12.3. The major element chemistry of the by-products was dominated by Ca, Mg, S, Fe, Al and Si. All materials met the criteria for ceiling concentrations of Cd, Cr, Cu, Pb, Mo, Ni, Se and Zn as defined for land application of sewage sludges. Arsenic exceeded the ceiling level in one of six samples. An artificial weathering study was conducted to evaluate the impact of PFBC by-products on water quality in mined land reclamation. Fatty acid oxidation was studied in the presence of inhibitors of carnitine palmitoyltransferase ... more Fatty acid oxidation was studied in the presence of inhibitors of carnitine palmitoyltransferase I (CPT I), in normal and in peroxisome-proliferated rat hepatocytes. The oxidation decreased in mitochondria, as expected, but in peroxisomes it increased. These two effects were seen, in variable proportions, with (+)-decanoylcarnitine, 2-tetradecylglycidic acid (TDGA) and etomoxir. The decrease in mitochondrial oxidation (ketogenesis) affected saturated fatty acids with 12 or more carbon atoms, whereas the increase in peroxisomal oxidation (H2O2 production) affected saturated fatty acids with 8 or more carbon atoms. The peroxisomal increase was sensitive to chlorpromazine, a peroxisomal inhibitor. To study possible mechanisms, palmitoyl-, octanoyl- and acetyl-carnitine acyltransferase activities were measured, in homogenates and in subcellular fractions from control and TDGA-treated cells. Peroxisomal fatty acid oxidation in rat liver isolated hepatocytes occurs at a lower rate and wit... more Peroxisomal fatty acid oxidation in rat liver isolated hepatocytes occurs at a lower rate and with different substrate specificity, than in cell free in vitro systems. Acyl-CoA availability might explain the difference, but our finding that phenothiazines selectively inhibit peroxisomal fatty acid oxidation (Biochem. Biophys. Res. Commun. 120:505, 1984) raises the possibility of alternative explanations. Substrate access into the organelle might be a regulated step for which several mechanisms have been proposed (Ann. N.Y. Acad. Sci, vol. 386, 1982). Our recent description of channels or pores in the peroxisomal membrane (Labarca et al., this symposium) adds a novel structure relevant to transmembrane molecular exchanges.

To characterize the effects of inhibitors of Epstein‐Barr virus (EBV) reactivation, we establishe... more To characterize the effects of inhibitors of Epstein‐Barr virus (EBV) reactivation, we established Raji DR‐LUC cells as a new test system. These cells contain the firefly luciferase (LUC) gene under the control of an immediate‐early gene promoter (duplicated right region [DR]) of EBV on a self‐replicating episome. Luciferase induction thus serves as an intrinsic marker indicative for EBV reactivation from latency. The tumor promoter 12‐O‐tetradecanoylphorbol‐13‐acetate (TPA) induced the viral key activator BamH fragment Z left frame 1 (BZLF1) protein (“ZEBRA”) in this system, as demonstrated by induction of the BZLF1 protein‐responsive DR promoter upstream of the luciferase gene. Conversely, both BZLF1 protein and luciferase induction were inhibited effectively by the chemopreventive agent curcumin.
