Background and Aims: In most of the studies, Toxoplasma gondii is maintained in laboratory mice o... more Background and Aims: In most of the studies, Toxoplasma gondii is maintained in laboratory mice or studied in vitro using non-lymphoid cell lines or primary mouse macrophages. The target of our research was to design a new axenic culture of Toxoplasma gondii tachyzoites to providing a sufficient quantity of them. Material and Methods: Theileria annulata-infected lymphoblastoids, which had been maintained up to 260 sub-cultures to attenuate the Theileria annulata, were evaluated for their suitability to the cultivation of Toxoplasma gondii tachyzoites. This cultivation process was carried out continuously for up to 10 passages, and after each 5 sub-culture, 0.1 ml of culture suspension (1×106 tachyzoites) was inoculated to each BALB/c mouse. Results: It was observed that the tachyzoites have attacked the lympho blastoids, multiplied inside them, and many fresh tachyzoites with typical shape and gliding movement were present in the culture suspension. A number of antigens have been identified as protective against leishmaniasis but a single antige... more A number of antigens have been identified as protective against leishmaniasis but a single antigen may not efficiently protect the human population with different immunological backgrounds. Thus, it is important to use several protective antigens. In the study, we fused three genes of L. major into pLEXSY- neo2 vector as a vaccine candidate. N-terminal part of LeIF gene was amplified by PCR from genomic DNA and was cut with Hind III and EcoRI enzymes then it was cloned into recombinant pEGFP-N1- LACK-TSA plasmid upstream of the GFP gene. The integrity of cloning was carried out by Colony PCR with forward LeIF and reverse TSA primers. Then, the LeIF-LACK-TSA (LLT) - EGFP Fusion was digested with BglII and NotI and subcloned into pLEXSY-neo2 vector at the same sites and the correct insert orientation was confirmed by enzymatic digestion. Respirable coal mine dust (RCMD) exposure is associated with black lung and silicosis diseases in... more Respirable coal mine dust (RCMD) exposure is associated with black lung and silicosis diseases in underground miners. Although only RCMD mass and silica concentrations are regulated, it is possible that particle size, surface area, and other chemical constituents also contribute to its adverse health effects. This review summarizes measurement technologies for RCMD mass concentrations, morphology, size distributions, and chemical compositions, with examples from published efforts where these methods have been applied. Some state-of-the-art technologies presented in this paper have not been certified as intrinsically safe, and caution should be exerted for their use in explosive environments. RCMD mass concentrations are most often obtained by filter sampling followed by gravimetric analysis, but recent requirements for real-time monitoring by continuous personal dust monitors (CPDM) enable quicker exposure risk assessments. The protozoan parasites of the genus Leishmania are the causative agents of different clinical di... more The protozoan parasites of the genus Leishmania are the causative agents of different clinical diseases. Fetal calf serum (FCS) is the main part and the most expensive ingredient of the Leishmania culture media. Here, the efficacies of different concentrations (1%, 2.5%, 5%, and 10%) of the filtered and autoclaved sheep and mouse urine were evaluated as a growth stimulator in Leishmania culture procedure. The results indicated that culture media enriched with the filtered sheep and mouse urine supported the growth of the parasites and can be used for cultivation of Leishmania parasites. In conclusion, this study has demonstrated an alternative low-cost medium that could be used in cultivation process of Leishmania major promastigotes.

Karaj is an area with large influx of refugee people in Iran. To increase knowledge about parasit... more Karaj is an area with large influx of refugee people in Iran. To increase knowledge about parasitic infections, we carried out this research during 2006-2008. We recorded the stool examination results and some of their personal characteristics. A total of 13,915 human stools were examined, and 649 (4.7%) were positive for intestinal parasites. Among them, 13 (0.09%) had worm and 636 (4.6%) had protozoan infections. Maximum infections belonged to Giardia intestinalis, and 534 (3.8%) samples had this infection. Other parasitic infections included Entamoeba coli (0.39%), Entamoeba histolytica (0.021%), Blastocystis hominis (0.08%), Trichomonas hominis (0.1%), Iodamoeba butschlii (0.06%), Chilomastix mesnili (0.007%), Endolimax nana (0.05%), Enterobius spp. eggs (0.028%), Taenia proglottids (0.028%), and Strongyloides stercoralis larvae (0.03%). The maximum numbers of referred people to laboratories were in July and the maximum percentage of infections was in August. There is a point that all 5 Strongyloides stercoralis infections were pertained to 2008. With attention to the rate of parasitic infections (4.7%), it seems that we should take additional educational information to wide spectrum of people living in this city.
Several methods of cultivation of Leishmania parasites are available, and among them, biphasic me... more Several methods of cultivation of Leishmania parasites are available, and among them, biphasic media like Nicolle-Novy-MacNeal medium are considered to be superior for the initial isolation of the parasites and their in vivo infectivity. In the present work, the efficacy of sheep blood-LB agar base medium (SLM) was evaluated for the cultivation of Leishmania major promastigotes. The SLM supports the proliferation of the parasites and can be used for routine isolation and cultivation of Leishmania parasites with acceptable in vivo infectivity for research purposes.
