The Ameliorative Role of a Marine Sponge Extract against Mixture of Persistent Organic Pollutants... more The Ameliorative Role of a Marine Sponge Extract against Mixture of Persistent Organic Pollutants induced Changes in Hematological Parameters in Mice Persistent organic pollutants (POPs), especially polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) are highly toxic to the marine environment. Since little is known about the effect of natural life mixture of POPs on the hematological parameters in Mice, the present study investigated the effect of POPs compounds to induce toxicity in mice using hematological parameter as biomarkers .The protective role of Hyrtios aff. Erectus sponge extract isolated from red sea was also examined. A mixture of PCB 28, PCB 52, , PCB 101, PCB 118, PCB 153, PCB 138 and PCB 180,alpha-Hexachlorocyclohexane, beta-Hexachlorocyclohexane, gamma-hexachlorocyclohexane, Aldrin , O,P’-DDE, Dieldrin, P,p DDE, O,P DDD, Endrin, P,p DDD and P,pDDT were extracted from sediments collected from Lake Mariout. Mice groups were assign into four groups and hematological parameters were compared between these different groups. Total white blood cell (WBC) count, red blood cells (RBC), hemoglobin, and hematocrit were decreased significantly in the induced group. No significant increase were observed for MCV, HCH, MCHC for induction group; while a significant increase in WBC, RBC, and PLT was observed in the pretreated group. On the other hand, a little or almost no change was observed in the pre-treated group by Hyrtios aff. Erectus extract. These findings were confirmed with immunohistochemistry results which showed marked eosinophilia of hepatocyte cytoplasm. At the same time the centrilobular showed hypertrophy and midzonal cytoplasmic vacuolization of hepatocytes in contrast to the pretreated group and the extract group. The present study concluded the existence of significant associations between POPs induce toxicity and hematological different parameters, i.e. WBC, RBC, HCT and PLT. Also the present results confirmed the protective role of Hyrtios aff. Erectus extract which harbor a great diversity of bioactive compounds and minerals.

Twenty-eight male Sprague-Dawley rats were divided into four groups (n=7). Group I served as −ve control, group II served as the induced group receiving subcutaneously for seven days 0.25 mg heavy metal mixtures, group III received (i.p.) TS extract of dose 40 mg for seven days, and group IV served as the protected group pretreated with TS extract for seven days as a protection dose, and then treated with the heavy metal-mixture. The main pathological changes within the liver after heavy-metal mixtures administrations marked hepatic damage evidenced by foci of lobular necrosis with neutrophilic infiltration, adjacent to dysplastic hepatocytes. Inhibition of acetylcholinesterase (AChE.3.1.1.7), the key enzyme in the breakdown of acetylcholi... more Inhibition of acetylcholinesterase (AChE.3.1.1.7), the key enzyme in the breakdown of acetylcholine, is considered as a promising strategy for the treatment of neurological disorders such as Alzheimer's disease (AD). The brain AChE from female Egyptian Mediterranean buffalo (Bas Buballus) was purified by ammonium sulphate precipitation, Sephadex G-25, Sephadex G-100 and DEAE-cellulose. Finally, Polyacrylamide gel electrophoresis was carried out to clarify the enzyme purity. The effect of the natural extracts of Ginkgo biloba and berberry and a synthetic derivative of genistein; ipriflavone on the activity of pure AChE were carried out in an in vitro study. Ginkgo biloba ands berberry extracts inhibited AChE. The increase in the K m-values with no differences in the V max-values pointed toward competitive type of inhibition. On the other hand, ipriflavone could be accounted as non-competitive inhibitor to AChE, where it caused three fold decrease in V max – value and did not alter the K m-value. Moreover, AChE is inhibited in a mixed type of inhibition by Aricept ®. Interestingly, the inhibitory per cent of ipriflavone nearly equal to that of Aricept ® (70%). It is evident from the present study that ipriflavone is the strongest inhibitor.

Marine derived fungi are considered as a promising source of novel drugs due to their biodiversit... more Marine derived fungi are considered as a promising source of novel drugs due to their biodiversity and consequent chemo-diversity. Although marine microorganisms especially fungi are not well defined taxonomically, making this a promising frontier for the discovery of new medicines. This study focused on marine derived fungi as a model for bio-active exploration for new entities with anti-inflammatory and antioxidant capacity. Three in-vitro assays were used to investigate the bioactive antioxidant potentiality of fungal extracts. Thiobarbituric acid (TBARS), α,α-Diphenyl-ß-picrylhydrazyl (DPPH) and NO assay are based on their total phenolic and flavonoid content of each extract group. Ch. globosum recorded the highest antioxidant activity (92.82%) in TBARS assay, while G. dankaliensis came first by recording 59.28% in DPPH assay in comparison with ascorbic acid (61.83%). In NO inhibition assay, N. oryzae showed 49.3% comparing with ascorbic acid (73.12%). From the preliminary result of our extracts, we can consider the marine derived fungi extracts as promising antioxidant and anti-inflammatory drug candidate.
