RNaseA kinetic studies is performed with the drug metosartan using RNA as the substrate Metosartan, is a drug used as blocker in excretion and found to contain inhibitory property on RNaseA. Protein degradation and thiol titration assay has found to be that the drug has reducing property on RNaseA.

Aim: Assessment of genome integrity in testis and to explain the reasons for infertility with the... more Aim: Assessment of genome integrity in testis and to explain the reasons for infertility with the drug metosartan. Methodology: Comet assay with freshly isolated mitochondria, cryopreserved mitochondria and cryopreserved tissue mitochondria and cell proliferation assay by dye exclusion method and topoisomerase assay in fresh and deep freezed DNA. Results and Discussion: Comet assay of isolated mitochondria both from fresh and cryopreserved samples was examined by fluorescence microscope, which revealed that cryopreservation of testes mitochondria samples lead to injury to the tissue and DNA fragmentation. Topo isomerase assay has showed that increased incubation leads to DNA fragmentation and cell proliferation assay with the drug has showed the 0% viability of the cells by trypan blue. So, in vivo studies are necessary to confirm whether the drug is safe or not. Conclusion: Assessment of genome integrity lead to the finding that drug was not safe it resulted in so many difficulties in invitro condition.

Metosartan decreases testes viability and sperm count was found to be high in metosartan treated rats compared to Metosartan +RNase A group. RNase A induces Netosis in both invivo and invitro condition in both RNase treated group and as well as RNase A+ Metosartan. Deep cold storage of mitochondria induces expression of cyt P450gene in testes mitochondria but not in sperm.

From the previous studies it was clear that RNase A is present in pancreas and our new Finding in... more From the previous studies it was clear that RNase A is present in pancreas and our new Finding includes that RNase that is similar to RNase A was also present in testis along with other RNases in the tissue. In chromatofocussing the peak was observed in fraction 5 of PI 9.6 and in HPLC it was present in all fractions of the column. The aim of present work is to isolate and identify RNase A through HPLC and chromatofocussing. From the results we conclude that RNase A is present in testis in addition to its presence in epidydymis .Future perspectives include finding of other RNases present in the testis and also further evaluation by other methods.

So, we have chosen these two organisms because of their abundant availability, ease of plating and culturing on media. Proteases are most important in detergent industry and it is easy to isolate protease in large quantities from E. coli and Yeast strain S. cerevisiae and can recover the protease with low cost downstream processing methods. This article mainly focuses on the preliminary results about the secretion of protease in to the medium by fungi S. cerevisiae without requirement for autolysis and found to be cost effective. The protease can be recovered easily by simple pelleting of cells at 4000rpm for 15min.

This fruit is gaining increasing worldwide acceptance and has received extensive marketing and a wide distribution due to its relevant nutritional benefits for human health. This work presents relevant information on the production, composition and application of avocado, with an emphasis on its by-products, focusing on the proper use of waste and the possibility of monetizing waste for nutritional and environmental purposes. The entire avocado is rich in bioactive compounds (pulp, peel and seed) and presents several health benefits, such as antimicrobial, antioxidant and anticancer activities, as well as dermatological uses and others. Therefore, several food grade ingredients can be obtained from avocado wastes, particularly premium-grade fats or extracts with a high functional power. Studies should continue to identify the profiles and phytochemicals available to the business sector, which can also be implemented to valorize the nutritional and functional potential of avocado seeds and peels.

RNase is used as one of the model to study drug inhibition because less is known about metosartan... more RNase is used as one of the model to study drug inhibition because less is known about metosartan the drug of βreceptor blocker on heart that is used to treat cardiac arrhythmias. So it is important to do studies on this drug as previous reports of mine reported apoptosis in germinal tissues, and to find that whether it inhibits RNase A in vitro or not was one of the task that was to be determined by this study. As we have found out that as it inhibits RNase A, protein synthesis is also effected but not to that extent as the drug is active only for 2-3 hrs. It is important to study whether it arrest RNase A only in testis or globally. RNaseA an endonuclease that cuts the RNA from middle of it by cleaving after cytidine and also the uridine bases as fast as possible. Agarose gel electrophoresis is one of the technique used for the study to know if the drug inhibits RNase or not by simply monitoring through degradation of RNA.

Hormonal protection through suppression was proven excellent for the stem cell survival in testis, where as in other tissues it remains obscure. The hormonal suppression of male infertility was done mainly using testicular harmones like FSH and testosterone, Whereas GH is mainly involved in neuroprotective effects and protection against ischaemia compared to male fertility. Substances like resveratrol, also protect male fertility by improving the sperm motility. Stem cell survival through hormonal suppression and protective effects of other compounds like resveratrol against sperm motility was proven to be useful to the mankind with respect to male infertility.

Epigenetic modifications like gene regulation, DNA histone modification and methylation patterns are mostly affected during Hypertension condition and need to be carefully monitored. So, metosartan is the one of the antihypertensive drug used to study its effects on genome especially in invivo condition. Metosartan doesn’t affect the chromatin integrity in hypertensive rats but reduces the sperm count drastically and reaches up to zero at 1.25mg/ml concentration. Majorly the drug affects the sperm count but not the maturation of sperm. In hypertension condition the sperms are aggregated in to complex where as in metosartan treated group invitro lead to severe looping of sperms and resulted in formation of apoptotic bodies. I... more Metosartan induces so many deleterious effects in male Wistar rats treated with drug and RNase. It causes pre maturation of sperms and reduces the sperm count as well as reduces viable sperm count. It induces the xchromosome desynapsis leading to testicular carcinoma. The effects are so profound in case of drug than compared to the enzyme RNase. Metosartan induces apoptosis in testicular tissue mainly through intrinsic pathway which is mainly due to genotoxic agents. The drug mainly inhibits RNase A in rat testes and has positive effect on Mitosis.

RNase A is the most experimental protein in the 20th century. Disulphide bonds are necessary for enzymatic action of many proteins and it is also required for this protein. RNaseA kinetic studies is performed with the drug metosartan using RNA as the substrate Metosartan is an combination of metoprolol and telmisartan and found to contain inhibitory property on RNaseA. Protein degradation and thiol titration assay has found to be that the drug has reducing property on RNaseA. The study concludes that metosartin inhibits the activity of RNAse A allosterically by reducing the disulphide bonds present on it. The reduction of disulphides may dissociate the enzyme to its monomer which is not highly active hence reducing the catalytic activity of RNase A. HPLC studies confirm the RNaseA presence in testes and drug also induce toroid packing of DNA in immature sperms.

fibrosis is one of the well understood diseases that are discussed here. Unfolded proteins are guided by molecular chaperones folding them in
to its native state. Whereas some of the mutations lead to the generation of misfolded proteins whose study is difficult as aggregation properties
in the cell differ by type of fusion proteins used .So, continuous effort is being made in this area and the results are promisive but, as most of the
proteins contain un organised regions of 30 amino acids which is necessary for non specific interactions with proteins. So, as protein –proteininteractions play a major role, designing therapeutic drugs pertaining to protein –protein interactions is found to be useful.
