The Runx2 transcription factor is a key regulator of osteoblast differentiation. In response to 1... more The Runx2 transcription factor is a key regulator of osteoblast differentiation. In response to 1alpha,25 dihydroxy vitamin D3, Runx2 may interact with the 1alpha,25 dihydroxy vitamin D3 receptor (VDR) in the promoter of target genes, producing a synergic activation of their transcription. Previous studies have suggested that the motifs responsible for the VDR-Runx2 interaction are contained within the 230-361 domain of Runx2. In this work, we confirmed by GST-pull down that Runx2(I(209-361)) is sufficient to interact with the VDR. To obtain structural information, GST-Runx2(I(209-361)) protein was overexpressed in Escherichia coli, purified and crystallized using the hanging-drop vapor-diffusion method and polyethyleneglycol as a precipitant. The crystals were found to diffract to a maximum resolution of 2.7 A and a complete data set to a 3.3 A resolution was collected and analyzed. The crystals belong to the tetragonal system, with a space group P4 and unit-cell parameters of a = b = 90.8, and c = 57.2 A. The presence of a monomer of the recombinant GST-Runx2(I(209-361)) in the asymmetric unit gives a V(M) of 2.7 A(3) Da(-1) and a solvent content of 54.8%.

En este trabajo se diseñó una intervención para contribuir a la formación de científicos, a travé... more En este trabajo se diseñó una intervención para contribuir a la formación de científicos, a través de rediseñar la evaluación de una tarea de desempeño auténtica. Para ello, se creó y validó una rúbrica analítica para evaluar un informe de laboratorio en formato publicación científica en la asignatura bioquímica. Se obtuvo un consenso entre evaluadores de 70%. El análisis permitió mejorar el instrumento e identificar debilidades en el desempeño de los estudiantes. Se obtuvo la percepción de los estudiantes de la experiencia, quienes declararon utilizar la rúbrica, apreciándose acuerdo respecto a su calidad, objetividad y pertinencia. Sin embargo, no hubo acuerdo respecto de la utilidad para guiar y retroalimentar, ni hubo efecto en el rendimiento, evidenciando que se requiere robustecer el diseño. En consecuencia, se generó una propuesta que incorpora: involucrar a los estudiantes en la definición de estándares de calidad, guiar el análisis de datos, incluir coevaluación y retroalimentación.

The structure of phycobiliproteins and their spatial organization in the phycobilisome provide th... more The structure of phycobiliproteins and their spatial organization in the phycobilisome provide the environment for high efficiency in light harvesting and conduction towards photosystem II. This article focuses on the analysis of R-phycoerythrin, a light harvesting hexameric phycobiliprotein that is part of the phycobilisomes. The interaction surfaces and the environment of the chromophores of R-phycoerythrin were studied in order to explain its structural stability and spectroscopic sensitivity, properties revealed by perturbation experiments. Three interaction surfaces are described (alpha beta), (alpha beta)3 and (alpha beta)6. The analysis shows the importance of alpha subunits in the interaction between trimers, the homodimeric nature of the monomer (alpha beta) and also the presence of anchor points in every interaction surface studied: alpha18Phe and beta18Tyr for (alpha beta), beta76Asn for (alpha beta)3 and alpha25Asn for (alpha beta)6. Side chains of arginine, lysine or glutamine residues are located in the proximity of the chromophores providing the correct stabilization of their carboxylates. Aspartic acids residues are associated through H-bonds to the N atom of the two central rings of the tetrapyrrolic chromophores. Changes in the spectroscopic properties are observed in perturbation experiments, confirming the spatial requirement for an efficient resonance energy transfer among chromophores and through the phycobilisome.

Las instituciones de educación superior han transitado hacia un modelo de competen-cias, en el cu... more Las instituciones de educación superior han transitado hacia un modelo de competen-cias, en el cual las competencias genéricas han cobrado relevancia en la formación y el campo laboral. Esto necesariamente implica la incorporación explícita en la docencia, a través de actividades activas y auténticas. Los ambientes laborales se caracterizan por la multidisciplina, y requieren que el trabajo se desarrolle orientado hacia la colaboración. Ambas condiciones, si bien ampliamente reconocidas, se replican insuficientemente en el aula. De hecho, existen escasas experiencias de cómo incorporarlas en educación superior. El trabajo colaborativo en equipos multidisciplinarios se plantea como esen-cial para trascender hacia entornos creativos y resolución de problemas, de reflexión y crecimiento personal y grupal. En el ámbito de la enseñanza y aprendizaje sus ventajas han sido ampliamente reportadas. Este trabajo tuvo por objetivo diseñar una interven-ción para promover el trabajo colaborativo en entornos multidisciplinarios en un pro-grama de magíster, y evaluar la percepción de los estudiantes. El diseño del estudio es de carácter pre-experimental, cuantitativo, de alcance descriptivo. Se diseñó e imple-mentó una intervención didáctica y se obtuvo la percepción del estudiante en tres eta-pas de la intervención. Las dos cohortes reportaron no conocer el concepto de trabajo colaborativo, valorando y relacionándolo con la disciplina y aspectos afectivos, además de una percepción muy positiva de los beneficios de la retroalimentación. Ambas co-hortes percibieron alta utilidad de las actividades incorporadas en la asignatura, junto al logro equivalente de competencias disciplinares y genéricas, reportando aplicación de los conceptos en actividades subsecuentes de la asignatura y en el campo laboral. Higher education institutions have moved towards a competence-based model. In this context, generic competences are highlighted as important features in both education and professional practice. This necessarily involves planning and incorporating them explicitly into teaching, through active and authentic activities. Given the multidiscipli-nary nature of work environments, tasks must be based on collaboration. Unfortuna-tely, both conditions are insufficiently replicated in the classroom. In fact, there is little experience with incorporating them into higher education. In multidisciplinary teams is considered a key to interacting and contributing towards creative environments, pro-blem solving, reflection, and personal and group growth. Moreover, its advantages in the context of teaching-learning processes have been widely reported. This work aims to design an intervention to promote collaborative work in multidisciplinary settings in a master's program and assess students' perception. The study design is pre-experi-mental, descriptive, and quantitative. A didactic intervention was designed and imple-mented. The students’ perceptions were assessed at three stages of the intervention. The two cohorts reported not knowing the concept of collaborative work and prized the in-tervention activities. As a result, participants positively valued collaborative work and feedback, and were able to apply it in subsequent course activities and, also, at work.Keywords: Higher education; transversal competences; team work; multidisciplinary education; feedback

Problem-based learning using authentic material from the web was used to teach metabolism in a bi... more Problem-based learning using authentic material from the web was used to teach metabolism in a biochemistry course. In place of traditional lectures, students' analyzed health or nutrition articles from newspapers and magazines, which were debatable from a scientific point of view, following the principles of problem-based learning. A mixed method was used to assess the students' perception, use of sources of information and web services while performing the task, and changes in self-directed learning. Students' perception was particularly positive. The majority stated that the methodology helped them to apply knowledge to real life and that they learned about the topic analyzed by their group. The perception that problem-based learning promotes the ability to solve problems, critical thinking, and collaborative work is noteworthy. Tutors considered that teams identified the problem and concluded correctly, noticing students' enthusiasm and motivation. The methodology also promoted scientific reading. More importantly, a significant improvement in self-directed learning of the 2014 cohort was detected. This intervention suggests that this methodology is a valuable alternative to motive and promote self-learning; representing an opportunity to shift the focus of instruction from the teacher to the student. The design of the activity and materials are described in detail. Also, limitations and solutions are discussed.

However, it is one of the least intervened and researched areas in higher education institutions, all over the world. To compare the written tests applied to students of three health science undergraduate programs (Speech Therapy, Medical Technology and Nursing), with the written tests of three programs of other areas (Business and Administration, Psychology and Bioengineering). Comparisons were done using the Authentic Assessment Model's indicators. Also, the magnitude of the change in these variables was evaluated in these two groups of undergraduate programs, after the participation of the teachers in a training program based on this model. A quantitative and repeated measurements design was used. Crystal structures of the GTPase and PH domains from various species have been fitted to the stru... more Crystal structures of the GTPase and PH domains from various species have been fitted to the structure of the DPRD dynamin phospholipid tube in its constricted and non constricted states (Mears et al., 2007). The PRD interacts with the SH3 domains of several proteins involved in signaling pathways. We are using cryo-EM and a single particle approach to solve the structures of the full length proteinlipid tubes in the constricted and non-constricted states. In both states, we have observed a decrease in the number of subunits per turn compared to previous structures of DPRD dynamin tubes. This suggests that the presence of the PRD changes the arrangement of the dynamin domains around the phospholipid tube. Further evidence of a direct interaction between the GTPase domain and the PRD is provided by simultaneous immunogold labeling of the two terminal domains.

Background: The mineralized skeleton is a major evolutionary novelty that has contributed to the ... more Background: The mineralized skeleton is a major evolutionary novelty that has contributed to the impressive morphological diversifications of the vertebrates. Essential to bone biology is the solidified extracellular matrix secreted by highly specialized cells, the osteoblasts. We now have a rather complete view of the events underlying osteogenesis, from a cellular, molecular, genetic, and epigenetic perspective. Because this knowledge is still largely restricted to mammals, it is difficult, if not impossible, to deduce the evolutionary history of the regulatory network involved in osteoblasts specification and differentiation. In this study, we focused on the transcriptional regulators Runx2 and VDR (the Vitamin D Receptor) that, in mammals, directly interact together and stabilize complexes of co-activators and chromatin remodellers, thereby allowing the transcriptional activation of target genes involved in extracellular matrix mineralization. Using a combination of functional, biochemical, and histological approaches, we have asked if the interaction observed between Runx2 and VDR represents a recent mammalian innovation, or if it results from more ancient changes that have occurred deep in the vertebrate lineage.

To assess the effects of emergency remote teaching (ERT), we evaluated the expectations and, subsequently, the experiences of university students about online education. This study employed a simple prospective design as its method. We assessed the expectations of 1,904 students from different discipline areas (1,106 women and 798 men; age M = 21.56; SD = 3.07) during the beginning of the first semester, March 2020 (T1), and their experiences at the end of the same academic period, September 2020 (T2). We used convenience non-probability sampling. Participants responded to the questionnaire on Expectations toward virtual education in higher education for students and the questionnaire on virtual education experiences in higher education. Students' perception was particularly positive. The majority stated that the methodology helped them to apply knowledge to real life and that they learned about the topic analyzed by their group. The perception that problem-based learning promotes the ability to solve problems, critical thinking, and collaborative work is noteworthy. Tutors considered that teams identified the problem and concluded correctly, noticing students' enthusiasm and motivation. The methodology also promoted scientific reading. More importantly, a significant improvement in self-directed learning of the 2014 cohort was detected. This intervention suggests that this methodology is a valuable alternative to motive and promote self-learning; representing an opportunity to shift the focus of instruction from the teacher to the student. The design of the activity and materials are described in detail. Also, limitations and solutions are discussed.

To compare the written tests applied to students of three health science undergraduate programs (Speech Therapy, Medical Technology and Nursing), with the written tests of three programs of other areas (Business and Administration, Psychology and Bioengineering). Comparisons were done using the Authentic Assessment Model's indicators. Crystal structures of the GTPase and PH domains from various species have been fitted to the stru... more Crystal structures of the GTPase and PH domains from various species have been fitted to the structure of the DPRD dynamin phospholipid tube in its constricted and non constricted states (Mears et al., 2007). The PRD interacts with the SH3 domains of several proteins involved in signaling pathways. We are using cryo-EM and a single particle approach to solve the structures of the full length proteinlipid tubes in the constricted and non-constricted states. In both states, we have observed a decrease in the number of subunits per turn compared to previous structures of DPRD dynamin tubes. This suggests that the presence of the PRD changes the arrangement of the dynamin domains around the phospholipid tube. Further evidence of a direct interaction between the GTPase domain and the PRD is provided by simultaneous immunogold labeling of the two terminal domains.

Background: The mineralized skeleton is a major evolutionary novelty that has contributed to the ... more Background: The mineralized skeleton is a major evolutionary novelty that has contributed to the impressive morphological diversifications of the vertebrates. Essential to bone biology is the solidified extracellular matrix secreted by highly specialized cells, the osteoblasts. We now have a rather complete view of the events underlying osteogenesis, from a cellular, molecular, genetic, and epigenetic perspective. Because this knowledge is still largely restricted to mammals, it is difficult, if not impossible, to deduce the evolutionary history of the regulatory network involved in osteoblasts specification and differentiation. In this study, we focused on the transcriptional regulators Runx2 and VDR (the Vitamin D Receptor) that, in mammals, directly interact together and stabilize complexes of co-activators and chromatin remodellers, thereby allowing the transcriptional activation of target genes involved in extracellular matrix mineralization. Using a combination of functional, biochemical, and histological approaches, we have asked if the interaction observed between Runx2 and VDR represents a recent mammalian innovation, or if it results from more ancient changes that have occurred deep in the vertebrate lineage.

To assess the effects of emergency remote teaching (ERT), we evaluated the expectations and, subsequently, the experiences of university students about online education. This study employed a simple prospective design as its method. We assessed the expectations of 1,904 students from different discipline areas (1,106 women and 798 men; age M = 21.56; SD = 3.07) during the beginning of the first semester, March 2020 (T1), and their experiences at the end of the same academic period, September 2020 (T2).
